This work introduces the design and preparation of a new type of crosslinked polymers template based on chitosan-pectin polysaccharides (CHT-PEC) to encapsulate silver nanoparticles as a novel biological agent by using Anethum graveolens leaf extract as a natural reducing reagent. CHT-PEC hydrogel was formed by hydrogen bonds and crosslinked by glutaraldehyde to have a natural reducing/capping and stabilizer template for immobilizing nanoparticles (CHT-PCT/Ag NPs). The prepared CHT-PCT/Ag nanocomposite was confirmed by various advanced techniques like FT-IR, UV-vis, FE-SEM, TEM, elemental mapping, EDX, and XRD techniques. TEM images showed the particles shape was spherical and monodispersed with a size of 10-15 nm. The CHT-PCT/Ag NPs pure crystalline nature was recorded by the related XRD pattern. In the in vivo study, gestational diabetes mellitus was induced in rats via intraperitoneal administration of streptozotocin. The subjects were subsequently divided into five distinct groups: the CHT-PCT/Ag nanocomposite-100 μg/kg group, the CHT-PCT/Ag nanocomposite-400 μg/kg group, the glibenclamide group, the untreated rats, and the normal pregnancy rats (n = 10). The CHT-PCT/Ag nanocomposite was administered intragastrically over 25 days. On the concluding day, the concentrations of ALT, AST, ALP, and blood glucose were evaluated. After processing the tissues, the volumes of the liver and its sub-compartments were estimated. The CHT-PCT/Ag nanocomposite demonstrates the capability to lower the increased levels of AST and ALP enzymes. In the untreated group, the introduction of CHT-PCT/Ag nanocomposite led to a reduction in blood glucose levels. The streptozotocin administration caused a significant increase in the volume of hepatocytes and sinusoids. Nevertheless, following treatment with CHT-PCT/Ag nanocomposite-400, a marked decrease in their volume was observed. Conversely, the volumes of the portal vein and bile ducts remained consistent across the experimental groups. The presence of CHT-PCT/Ag nanocomposite resulted in alterations in the volume of the hepatic arteries and central vein. This study highlights the antidiabetic and hepatoprotective properties of CHT-PCT/Ag nanocomposite, suggesting its potential as a supplementary measure to protect liver while simultaneously prevent the gestational diabetes.
