BACKGROUND: the association of Non-polio Enteroviruses (NPEVs) with acute flaccid paralysis (AFP) is growing anxiety worldwide, particularly, In Iraq.

OBJECTIVES: This study aimed to highlight the importance of the molecular techniques in the detection of the NPEVs and to understand if there are any effects of the capsid genes polymorphisms on the virus' behavior in the tissue culture.

STUDY DESIGN: During 2014-2016, from 799 stool samples of AFP' patients, 90 NPEVs cell culture positive and negative isolates have been collected on the basis on their cytopathic effect levels in the cell culture for detection by the reverse transcriptase polymerase chain reaction (RT-PCR). For further molecular study, the Viral Protien-4 capsid gene (VP4) of 17 NPEVs RT-PCR positive isolates and the evolutionary relationship of these isolates were analyzed by the nucleotide sequence analysis technique and the phylogenetic tree, respectively.

RESULTS: the prevalence of the NPEVs positive results have been increased by the RT- PCR and the nucleotide sequence analyzed isolates were characterized as members of Enterovirus B with a predominance to the echoviruses, including echovirus (E)-4, E-7, E-9, E-14 and E-20. As well as, Coxsackie B virus, Enterovirus- 74, Enterovirus B-106 and untyped Enterovirus B strain. Noticeably, the E-4 and EV- 74 strains have been belonged for the recorded two death cases in this study. While an obvious coordination have been demonstrated between the evolutionary relationships of these isolates and their behavior in the tissue culture, the death case associated E-4 strain showed a different grouping pattern.

CONCLUSION: the VP4 gene sequencing, well typed the local strains into Enteroviruses B group. For best results, it's recommended to use other viral protein- capsids in the characterization of the NPEVs isolates.
