Аннотация

BACKGROUND: The widespread use of pesticides like acetamiprid has raised serious concerns about food safety and environmental pollution. Conventional techniques such as chromatography are accurate but require sophisticated instrumentation and complex sample preparation. Although electrochemical sensors offer advantages in portability and cost-effectiveness, most nucleic acid-based amplification approaches still rely on multi-step solution-phase reactions, limiting their practical application. Herein, a novel surface-confined sensing platform was designed that seamlessly integrates target recognition with signal amplification in a one-step procedure, significantly simplifying the operation process while maintaining high sensitivity.

RESULTS: A simple yet efficient electrochemical sensor was developed for pesticide detection. Using acetamiprid as a model target, introducing aptamer as target recognition element, designing primer exchange reaction (PER) cascade reaction as signal amplification strategy, target recognition and signal amplification were confined on the electrode surface and completed in one step. Target binding aptamer triggered PER, generating long DNA strands enriched with multiple guanine-rich sequences, forming numerous G-quadruplex-hemin complexes in situ, producing strong electrochemical signal that enabled sensitive acetamiprid detection with a detection limit of 2.73 pM (S/N = 3). The entire detection process from target addition to signal output only requires two simple steps, greatly simplifying the operation process and improving detection efficiency. Meaningfully, the sensor has demonstrated reliable performance in detecting acetamiprid in various vegetable, fruit, and soil samples, showing great potential for monitoring pesticide pollution in agricultural and environmental applications.

SIGNIFICANCE: This work establishes a new paradigm for pesticide detection by combining surface-confined reactions with nucleic acid amplification, offering superior simplicity and sensitivity. The demonstrated performance in real samples highlights its potential for food safety monitoring and environmental protection. The developed platform provides a versatile foundation for detecting various contaminants through adaptable nucleic acid designs.
