ETHNOPHARMACOLOGICAL RELEVANCE: Mesosphaerum pectinatum (L.) Kuntze is known in the Northeast of Brazil as "canudinho" or "sambacaitá", where it is mainly used as tea or infusion to treat gastrointestinal disorders.

OBJECTIVE: To evaluate the gastroprotective activity of the aqueous extract of the aerial parts of M. pectinatum (AEMP).

MATERIAL AND METHODS: AEMP was prepared and characterized by HPLC. Male Swiss mice were subjected to acidified ethanol (AE)-induced ulcers to evaluate, macroscopically and histologically, the gastroprotective effect of AEMP (25, 50, and 100 mg/kg, p.o.), and to measure myeloperoxidase (MPO) activity and IL-6 and sulfhydryl group levels in gastric tissue. Additionally, the involvement of non-protein sulfhydryl compounds (NP-SH), nitric oxide (NO), prostaglandins, ATP-sensitive K+ channels (KATP), and α2-adrenoceptors was investigated. The pylorus ligation model was used to assess mucus content.

RESULTS: HPLC analysis detected rosmarinic acid in AEMP. Pretreatment with AEMP (at all doses) significantly reduced the area of AE-induced gastric lesions and MPO activity. It also decreased IL-6 levels, increased sulfhydryl groups in gastric tissue, and attenuated the histological alterations induced by AE. Pretreatment with the NP-SH blocker or indomethacin reversed the gastroprotective response of AEMP. However, this reversion did not occur after pretreating mice with a KATP blocker, an inhibitor of NO synthases, or an α2-adrenoceptors antagonist. Additionally, treatment with AEMP increased mucus production in the pylorus-ligation model.

CONCLUSIONS: These results provide evidence that AEMP exerts gastroprotective action in mice by reducing the inflammatory response and involving NP-SH and possibly prostaglandins.
