Аннотация
BACKGROUND: While proton pump inhibitors (PPIs) control gastric acid symptoms in reflux disease, they cannot fully reverse the core pathology of NF-kB-driven inflammatory damage, creating an urgent need for therapies that directly target this mechanism. The aim of this study was to determine whether quercetin combined with curcumin modulates the nuclear factor-kappa B (NF-kB) inflammatory signalling pathway in a murine model of oesophageal erosive reflux disease.
MATERIALS AND METHODS: A reflux oesophagitis (RE) model was established in mice surgically. The mice were treated with quercetin (dietary), curcumin (topical), or their combination for 6 weeks. We analysed the expression of NF-kB and inflammatory factors, such as interleukin (IL)-1b, IL-8 and tumour necrosis factor (TNF)-a, using real-time PCR. We performed enzyme-linked immunosorbent assays to validate the real-time PCR findings, and western blots to determine the effects on NF-kB p-p65, TNF-a, IL-1b and IL-8.
RESULTS: The combination of quercetin and curcumin significantly suppressed the NF-kB inflammatory pathway by reducing the expression of various pro-inflammatory cytokines in our murine model of oesophageal erosive reflux disease. The combination was more effective than either compound alone.
CONCLUSIONS: Our findings suggest that administering quercetin in combination with curcumin is a promising approach for mitigating inflammation via modulation of the NF-kB pathway in oesophageal erosive reflux disease.
