Chios Mastic in Well-Controlled Hypertension: Findings From the HYPER-MASTIC Randomized Controlled Trial.

Residual cardiovascular risk persists in patients with adequately controlled hypertension, driven partly by inflammation, oxidative stress and endothelial dysfunction. Chios mastic (Pistacia lentiscus var. Chia) has shown antihypertensive, antioxidant and vascular effects in experimental and clinical settings. This trial evaluated whether Chios mastic supplementation confers additional cardiovascular benefits in well-controlled hypertensive individuals. In this double-blind, randomized, placebo-controlled trial, 106 adults with stable, well-controlled hypertension and variable cardiovascular r

Monoclonal Antibodies Targeting Bacillus Calmette-Guérin (BCG) Outer Membrane Proteins Confer Protection Against Tuberculosis.

Tuberculosis (TB), caused by the Mycobacterium tuberculosis complex (MTBC), is a persistent global health crisis, remaining the world's leading infectious killer. There were abundant outer membrane proteins (OMPs) distributed in the bacterial cell wall, while fewer proteins were identified and characterized as antigens that could induce protective antibodies against TB. The OMPs of BCG were extracted with the detergent N-octyl-β-D-glucopyranoside, and the extracted proteins were used to immunize mice to generate an antibody library. Subsequently, the library underwent HAT/HT screening, and fiv

Radial Tiny Golden Angle UTE (tyGA-UTE) for Cardiac MRI in the Small Animal Model.

Robust cardiorespiratory motion correction is critical for high-quality ultra-high-field (UHF) preclinical cardiac magnetic resonance imaging (MRI). Whereas radial tiny golden angle (tyGA) sampling, data-driven self-gating (sg), compressed sensing (CS), and ultrashort echo time (UTE) are individually well-established techniques, their integration into a unified acquisition and reconstruction pipeline, together with systematic validation at 11.7 T remains insufficiently explored. This study presents and validates a UHF-optimized tyGA-based UTE workflow that enables high-temporal-resolution slid

The First Case of Digenic MODY Diabetes Involving Compound Heterozygosity of Three Variants in the PDX1 and HNF1B Genes.

OBJECTIVES: We report a rare case of digenic MODY. In 2016, at age 15, the patient was diagnosed with type 1 diabetes and began insulin therapy. After discrepancies in her glycaemic control were observed, an extensive differential diagnosis was conducted. In 2024, genetic testing confirmed monogenic diabetes, allowing for tailored treatment of the patient. METHODS AND RESULTS: The initial diagnosis was type 1 diabetes, later revised to MODY after Sanger screening. Genetic testing detected c.293C>T, p.Ala98Val in the HNF1A gene, which was initially misinterpreted as a variant causing Maturity

Discovery of Avian TCRVβ2+ T Cells Recognizing the Endogenous Ganglioside via a chCD1.1-Dependent Pathway.

This study presents the discovery and first functional characterization of chicken lipid-reactive Natural Killer T (NKT)-like cells, resolving a long-standing knowledge gap in avian immunology. Using purified TCRVβ2+ lymphocytes, we demonstrate through rigorous experiments combining shRNA-mediated gene silencing, pharmacological pathway blockade, primary MACS-purified co-cultures, and intracellular cytokine flow cytometry that these cells are activated by CpG-ODN-stimulated antigen-presenting cells (APCs). Activation selectively depends on APC-driven glycosphingolipid (GSL) biosynthesis, with

NFS1 as a Metabolic Vulnerability in Acute Myeloid Leukemia Through Iron Dysregulation and STAT1/IFIT2-Associated Signaling.

Acute myeloid leukemia (AML) is a metabolically heterogeneous malignancy with poor outcomes and limited durable therapeutic options. We investigated NFS1, a key enzyme involved in iron-sulfur cluster biogenesis, as a potential metabolic vulnerability in AML. NFS1 was overexpressed in AML samples, and higher expression was associated with inferior overall survival in the TCGA-LAML cohort. Genetic knockdown of NFS1 markedly suppressed AML cell proliferation in vitro and reduced tumor growth in subcutaneous xenograft models. NFS1 knockdown induced disturbances in iron homeostasis accompanied by i

Cytokine-induced killer cells combined with temozolomide suppress glioblastoma progression in an orthotopic model and demonstrate clinical feasibility in recurrent glioblastoma.

INTRODUCTION: Glioblastoma (GBM) remains an aggressive primary brain tumor with limited therapeutic options after recurrence. Cytokine-induced killer (CIK) cells are ex vivo-expanded cytotoxic lymphocytes with T-cell and natural killer-like properties. We evaluated CIK-cell-based therapy using an orthotopic LN229-luciferase GBM model and a retrospective cohort of patients with recurrent GBM. METHODS: Twelve independently manufactured patient-derived CIK-cell products were phenotypically characterized. In the orthotopic model, control, temozolomide (TMZ), and TMZ+CIK groups (n = 4 per group) w

Checkpoint-Directed Glycoengineering Reprograms the Vascular Lumen for Siglec-Mediated Immune Regulation.

Dysregulated immune activation at the vascular surface drives microvascular inflammation and tissue injury in many diseases, yet current therapies do not restore immune regulation at the blood-vessel interface. Terminal sialic acids engage inhibitory Siglec receptors on circulating immune cells and function as endogenous glyco-immune checkpoint ligands. Loss of sialylation on vascular endothelial cells during injury disrupts this regulatory axis and promotes vascular damage. Here, we introduce a checkpoint-directed glycoengineering strategy that installs multivalent Siglec-engaging sialic acid

FAM3A Inhibits Plasma Triglyceride Levels via Microvasculature-Involved Enhancement of VLDL-Triglyceride Hydrolysis and Lipid Uptake.

FAM3 metabolism regulating signaling molecule A (FAM3A) is ubiquitously expressed. However, its biological role remains largely unknown. This study investigated the regulatory roles of FAM3A in plasma triglyceride (TG) levels. Proteomics revealed upregulated lipid metabolism-related proteins, including CD36 and TCF15, in skeletal muscle of FAM3A-transgenic mice. Clinically, plasma FAM3A was reduced in patients with metabolic disorders. FAM3A-transgenic mice and recombinant FAM3A (rcFAM3A) injection in mice significantly reduced plasma TG levels, upregulated tissue lipid uptake and lipid transp

Optimizing Concentrated Growth Factor Therapy: Sodium Citrate Reduces Pain in Hair Loss Treatment.

BACKGROUND: Androgenetic alopecia (AGA) is a prevalent condition causing significant psychosocial distress. While concentrated growth factor (CGF) therapy has emerged as a promising regenerative treatment, the choice of anticoagulant remains a critical but underexplored variable that may affect the growth factor release, and patient experience. OBJECTIVE: To compare the efficacy, procedural pain, and patient satisfaction of CGF prepared with sodium citrate, heparin sodium, or no anticoagulant in patients with AGA. METHODS: This phase 2, multicenter, single-blind, randomized controlled trial

G6PD suppresses thrombopoiesis via interfering with TMOD3-mediated actin dynamics.

Glucose-6-phosphate dehydrogenase (G6PD), the committed enzyme of the pentose phosphate pathway, plays a pivotal role in glucose metabolism, which is integral to platelet biology. Although G6PD deficiency is a prevalent enzyme defect worldwide, the specific role of G6PD itself in platelets remains unclear. Herein, we discover a previously unrecognized phenotype in both human patients and murine models, demonstrating that G6PD deficiency leads to elevated platelet counts through enhanced proplatelet formation (PPF). Mechanistically, we reveal a non-canonical function of G6PD in regulating actin

Phytochemicals for Ameliorating Aluminum Toxicity: Dissecting the Mechanisms for Therapeutic Potential.

Although aluminum is the third common metal of the earth, it is physiologically nonessential and its higher levels on regular exposure can be harmful to the human body, affecting several vital organ systems. Environmental aluminum contamination and toxicity are therefore regarded as a "silent killer" as it is associated with the precipitation of several serious health complications chiefly involving the nervous, blood, and musculoskeletal systems. Various natural products obtained from medicinal plants; that is, phytochemicals have been reported to possess aluminum toxicity ameliorative potent

A new prognostic perspective on preoperative coagulation parameters for lung adenocarcinoma patients: development and validation of the DFP score.

BACKGROUND: Emerging evidence suggests a pivotal role of coagulation-related factors in tumor progression and survival outcomes. However, their integration into risk stratification models remains limited. This study aimed to develop and validate a novel prognostic scoring system based on preoperative coagulation parameters to improve survival prediction in surgically treated lung adenocarcinoma (LUAD) patients. METHODS: A total of 1081 patients with pathologically confirmed LUAD who underwent pulmonary resection at a single center were retrospectively enrolled and divided into training (n = 7

Mitochondrial Protection and Neural Repair via ROS-Responsive Biomimetic Nanoparticles in Intracerebral Hemorrhage.

Intracerebral hemorrhage (ICH)-induced secondary brain injury is driven in part by oxidative stress, blood-brain barrier disruption, mitochondrial dysfunction, and neuronal loss. Here, we developed a brain endothelial cell membrane-coated ROS-responsive biomimetic nanoplatform (NP@D+M) for co-delivery of the VDAC1 inhibitor DIDS and the mitochondrial fission inhibitor Mdivi-1. NP@D+M showed uniform size, preserved endothelial membrane proteins, and accelerated drug release under elevated H2O2. In ICH-like neuronal injury models, NP@D+M enhanced uptake, protected neurites, reduced LDH release a

Mass Spectrometry Imaging Reveals Biodistribution and Biotransformation of Black Phosphorus.

Black phosphorus (BP) holds significant potential in biomedicine, biosensing, and phototherapy; however, its instability and the poor understanding of its in vivo fate have limited its applications. Here, we developed an integrated label-free mass spectrometry imaging (MSI) platform to investigate BP's in vivo distribution, transformation, clearance, and metabolism in tumor-bearing mice. We detected BPs and related nitrogen/oxygen-containing phosphorus (P) clusters in six organs and tumor tissues, revealing distinctive tissue distributions. BP and its degradation products primarily accumulated
