Thioridazine induces phospholipid accumulation and necroptosis in parental and tamoxifen-resistant breast cancer cells.

The development of acquired resistance to tamoxifen poses a significant clinical challenge in breast cancer treatment. Tumour heterogeneity has emerged as a primary reason for the clinical implications of resistance, yet we still lack actionable targets to address this issue. Repurposing existing drugs has become an emerging trend to tackle demanding medical indications. Therefore, we aim to study the efficacy of the antipsychotic drug Thioridazine against both parental and tamoxifen-resistant breast cancer cells. In this study, we have demonstrated that Thioridazine induces phospholipid accum

Laser-Induced Dimeric Photoproducts of Chlorpromazine: LC-MS Identification and Molecular Docking Evidence of Enhanced Anticancer Potential.

Breast cancer treatments, such as chemotherapy, radiation, and surgery, often face significant limitations, highlighting the need for more effective and targeted therapies. Here, we investigate the potential of 266 nm laser irradiation of chlorpromazine as a novel approach to develop new antitumoral compounds. We identify six chlorpromazine photocompounds with masses in the range of 178-334 u, along with several dimeric compounds with masses between 566 and 600 u, using an HPLC-MS. In silico approaches assess their pharmacokinetic and pharmacodynamic properties while comparing their toxicity w

Membrane Partition and Structural Reorganization Induced by Antipsychotics with Distinct Clinical Profiles.

Antipsychotics (APs) are used in the treatment of severe mental disorders. Their mechanism of action involves interaction with multiple brain targets, notably the dopamine D2 receptor (D2R), where they compete with dopamine. Due to their lipophilic nature, APs also partition and accumulate in lipid membranes, particularly around the D2R and in synaptic vesicles. When intercalated into brain membranes, APs slowly accumulate and act as reservoirs, allowing their rapid release on demand to modulate neurotransmitter signaling. They also modify the physicochemical and mechanical properties of the l

Revealing the different effects induced by antipsychotic drugs using an electrochemical microsensor to measure exocytosis in living cells.

Real-time monitoring of neurotransmitter release in living cells is crucial for understanding neural functions and the efficacy of drug actions. Here, we developed an electrochemical microsensor chip using a multidimensional nanosensitive material (MNG-1) that detects dopamine (DA) with high sensitivity, enabling real-time analysis of exocytosis in living cells. Our sensor-based technique does not require advanced equipment and can detect single exocytotic events using a standard electrochemical workstation and a small Faraday cage, allowing for rapid and statistically significant data collect

Lysosomal TPC2 channel as a new target of chlorpromazine and clomipramine to induce protective autophagy in L-BMAA-induced neurodegeneration.

Several neurodegenerative diseases including amyotrophic lateral sclerosis (ALS) are characterized by toxic aggregates accumulation due to autophagy blockade, prompting researchers to identify new autophagy-activating drugs. Here we tested, in an in vitro ALS/PDC model, the neuroprotective effects of the antipsychotic Chlorpromazine (CPZ) and the antidepressant Clomipramine (CMI), chosen by drug repurposing approach for their ability to stimulate TPC2 lysosomal channel. Patch-clamp electrophysiology on enlarged lysosomes in NSC-34 motor neurons showed that CPZ and CMI induced large inwardly-re

Study of the Lipophilicity of Tetracyclic Anticancer Azaphenothiazines.

Although chlorpromazine is primarily used in psychiatry, it has been shown since its introduction to influence the course of neoplastic diseases. According to the strategy of drug repurposing, chlorpromazine has been successfully tested for its potential antitumor effects on multiple cancer cell lines. This effect is consistent with the overlap of molecular pathways observed for years between schizophrenia and cancer. The main objective of this work was to evaluate the lipophilicity of 17 previously synthesized tetracyclic chlorpromazine analogues exhibiting diverse anticancer and antimicrobia

Effects of Sedation on Rectal Temperature During Equine Dental Procedures Performed in Cold to Warm Ambient Temperatures.

Sedation is important for safe equine dental procedures, but it is unknown if there is a higher risk of sedation causing hypothermia in procedures performed during cold months. The authors hypothesize that sedation for a dental procedure will significantly decrease rectal body temperature in cold as compared to warm ambient temperatures. Adult equids (N = 246), from a private equine dentistry practice, that underwent dental equilibration, were selected for the study. Each horse's weight was estimated and sedation was administered intravenously (0.01 mg/kg detomidine and 0.01 mg/kg butorphanol)

The antipsychotic chlorpromazine induces oxidative stress in endothelial cells: an in vitro assessment employing Fourier-transform infrared spectroscopy.

Chlorpromazine (CPZ) is a first-generation antipsychotic that has been widely used to treat an array of neurological conditions, including schizophrenia, bipolar disorder, and anxiety. Treatment of these chronic conditions with CPZ has been linked to elevated levels of reactive oxygen species (ROS), and accumulating evidence supports a link between ROS and chronic and degenerative pathologies, including cardiovascular diseases. Therefore, the aim of this study was to observe the presence of oxidative stress in porcine aortic endothelial cells (PAE) exposed to different concentrations of CPZ in

Phenothiazine Derivatives and Their Impact on the Apoptosis Processes: A Review.

Phenothiazine derivatives have been used for decades as antipsychotic drugs in multiple mental health and physical conditions treatment (schizophrenia, mania in bipolar disorder, and psychosis). Epidemiological studies have shown that people with schizophrenia are less likely to suffer from cancer, which indicates the ability of antipsychotics to inhibit the development of cancer cells. It is our third review about the impact of phenothiazine derivatives on cell death. Here, we focused on apoptosis. We selected 92 papers that investigated the effect of phenothiazine derivatives on the apoptosi
