The use of the white biotechnology toolkit to edit natural purines for studying their anticancer activity via mTOR pathway.

Purine alkaloids were proven to have significant cytotoxic activity against different cancer cell lines via modulating several cellular pathways, leading to inhibition of cell proliferation and increasing cell death. The search for new potential cytotoxic compounds produced by natural eco-friendly means is of great importance. The microbial transformation of natural purine alkaloids, caffeine (Cf), theophylline (Tp), theobromine (Tb), and theacrine (Tc) via filamentous fungi was explored using Aspergillus versicolor (AUMC 4807), Aspergillus niger (NRRL 328), Cunninghamella echinulata (ATCC 138

The Impact of Controlled Fermentation Temperature on Chemical Composition and Sensory Properties of Cacao.

The content of phenolic compounds can affect the quality of cacao beans (Theobroma cacao L). The variation in the concentration of these compounds is influenced by factors such as cacao variety, fermentation conditions, and temperature, which play a crucial role in the method of bean drying. In this study, the analytical method of ultra-performance liquid chromatography (UPLC-DAD-RI) was developed to identify, quantify, and examine variations in the concentrations of catechins (catechin, epicatechin, and epigallocatechin) and methylxanthines (theobromine and caffeine) by subjecting the beans t

Principal Components Analysis of Spot Urine Caffeine and Caffeine Metabolites Identifies Promising Exposure Biomarkers of Caffeine and Theobromine Intake in a Cross-Sectional Survey of the United States Population (National Health and Nutrition Examination Survey 2009-2014).

BACKGROUND: Spot urine concentrations of caffeine and certain caffeine metabolites correlate well with 24-h caffeine intake and have potential use as caffeine exposure biomarkers. However, a high degree of intercorrelation exists among these urine compounds, and their correlations with intake are very similar, making the selection of a single intake biomarker challenging based on bivariate analyses alone. OBJECTIVES: To use principal components (PCs) analysis (PCA) as a dimensionality reduction tool to identify underlying correlation structures in the relationship of caffeine intake with spot

[Omics Study of Ovarian Malignancies: From Urine Metabolomic Profile to Minimally Invasive MicroRNA Markers].

A search for efficient biomarkers of ovarian cancer is one of the current trends in gynecologic oncology. Metabolic profiling by ultra high-performance liquid chromatography and mass spectrometry (UHPLC-MS) yields information about the total set of low-molecular-weight metabolites of a patient's biological fluid sample. The metabolites may provide potential disease markers, and their combination with microRNA level data significantly increases the diagnostic value. To identify the potential noninvasive diagnostic markers of serous ovarian adenocarcinoma, the metabolomic profile and microRNA tr

Exploration of Biomarkers of Food Intake in a Caribbean Hispanic Population.

Valid measures of dietary intake are essential for health and nutrition research, but typical forms- or interview-based measurements are susceptible to random and systematic errors. Although many biomarkers of food intake (BFIs) have been validated, we aimed to explore how food-BFI relationships are affected by genetic and lifestyle factors among Caribbean Hispanic adults. Dietary, clinical, anthropometric, blood metabolomics, and genotype data from 782 Puerto Rican adults were available. Thirty-one BFI-food intake relationships were assessed using linear regression, including covariates based
