Triazinyl-benzenesulfonamide derivatives as hCA IX inhibitors: Design, synthesis, and activity determination using an optimized stop-flow methodology.

This study presents the design, synthesis, and biological evaluation of a new series of 1,3,5-triazinyl benzenesulfonamide derivatives incorporating substituted piperazines, aminobenzenes, or adamantane moieties. The compounds were tested for inhibitory activity against human carbonic anhydrase isoenzymes II and IX, aiming for selectivity towards the latter, cancer associated isoenzyme IX, on the basis of an initial molecular docking screening. Several compounds showed inhibitory activity and selectivity exceeding that of the clinical benchmark acetazolamide. Among the most effective compounds

Click Chemistry-Enabled Parallel Synthesis of N-Acyl Sulfonamides and Their Evaluation as Carbonic Anhydrase Inhibitors.

A synthetically accessible library of N-acyl sulfonamides was constructed using a combination of copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) and N-acylation of primary sulfonamides. The proposed two-step reaction sequence had a high experimentally confirmed synthetic success rate (up to 85%) and gave reasonable product yields (up to 61%). As a result of the validation process, a 262-member compound library (out of >70K accessible combinations) was prepared. Biological profiling of the synthesized library by differential scanning fluorimetry and enzymatic assays identified several lo

Design and Synthesis of Bendamustine-Carbonic Anhydrase Inhibitors with Antiproliferative Effects in Clear Cell Renal Cell Carcinoma.

Human carbonic anhydrase IX (hCA IX) is markedly overexpressed in clear cell renal cell carcinoma and plays a key role in establishing an acidic tumor microenvironment associated with intrinsic chemoresistance. Extracellular acidification limits the uptake and efficacy of several cytotoxic agents, including bendamustine, a bifunctional alkylating agent whose activity depends on intracellular accumulation and DNA cross-link formation. Herein, we report the design and synthesis of novel bendamustine-carbonic anhydrase inhibitor (CAI) hybrids aimed at combining CA IX targeting with DNA-damaging a

Carbonic Anhydrase I and II as Biomarkers and Therapeutic Targets in Human Disease: From Physiology to Clinical Translation.

Carbonic anhydrases (CAs) are zinc-containing metalloenzymes that catalyze the reversible conversion of carbon dioxide and water into bicarbonate and protons, contributing to acid-base balance, pH regulation, and ion transport. Among human cytosolic isoforms, carbonic anhydrase I (CA I) and carbonic anhydrase II (CA II) are abundant and clinically relevant, yet their distinct roles are often obscured within broader discussions of the CA family. This narrative review evaluates CA I and CA II as biomarkers and therapeutic targets in glaucoma, atherosclerosis and vascular calcification, anemia, e

Water-soluble phthalocyanines with non-peripheral naphthoxy-sulfonate substituents: computational docking, biological and sono-photochemical investigations.

This study reports the synthesis of a novel ligand, sodium 6-(2,3-dicyanophenoxy)naphthalene-2-sulfonate (1), and the corresponding non-peripherally substituted metallophthalocyanines (MPcs) [M = Zn(II) (2), Ga(III) (3); X = Cl, In(III) (4); X = Cl]. These compounds were functionalized with 6-naphthoxy-2-sulfonic acid sodium salt groups. Given the limitations of conventional photodynamic therapy (PDT), we investigated the potential of sonophotodynamic therapy (SPDT), a dual-modality approach combining light and ultrasound, to enhance singlet oxygen (¹O2) production. Synthesis of novel bromophenolic sulfonamides with their acetylcholinesterase and carbonic anhydrase inhibitory actions.

Because of the important biological properties of halogenated benzene sulfonamides, here we report the first synthesis of bromophenolic sulfonamides and their human carbonic anhydrase I (hCA I), human carbonic anhydrase II (hCA II) and acetylcholinesterase (AChE) inhibition. For this purpose, 4-bromo-1,2-dimethoxybenzene was reacted with chlorosulfonic acid fallowed by amination with ammonia, N-alkyl amines and N,N-dialkyl amines to give a series of novel sulfonamides. Subsequent O-demethylation of these sulfonamides using BBr3 afforded the target bromophenolic benzene sulfonamides. The synthe

Acetazolamide inhibition of carbonic anhydrase 4 reverses opioid-induced synaptic rearrangements in nucleus accumbens and reduces drug-seeking behavior.

Persistent vulnerability to drug-seeking is driven by enduring synaptic adaptations, yet current μ-opioid receptor-targeting pharmacotherapies provide limited efficacy against these neuroadaptations. Thus, there is a critical need for mechanistically distinct, non-opioid interventions. We recently found that carbonic anhydrase 4 (CA4) disruption reduces cocaine-induced synaptic adaptations and drug-seeking. Building on this foundation, we sought to determine whether deleting CA4 or pharmacological inhibition with acetazolamide (AZD), a clinically employed carbonic anhydrase inhibitor-could mit

CA IX Inhibition by a Sulfonamide Compound: A Therapeutic Approach Against Breast Cancer.

Carbonic anhydrase IX (CA IX) is overexpressed in many solid tumors, contributing to cancer cell proliferation, survival, invasion, and metastasis. Sulfonamide-based compounds have emerged as potential anticancer agents by inhibiting this enzyme. In this study, we investigated the anticancer potential of a previously synthesized sulfonamide derivative, MMH-I, focusing on its CA IX-targeted activity and therapeutic efficacy in both in vitro and in vivo models of breast cancer. Cytotoxicity was assessed using MTT assays in 4T1 breast cancer cells, while apoptosis was evaluated by acridine orange

X-ray analysis of complexes of carbonic anhydrase II with 1,3-oxazole-containing sulfonamide derivatives elucidates the structural basis for their exceptionally high inhibitory potency.

Carbonic anhydrase II (CA2) is a zinc metalloenzyme that catalyzes the reversible hydration of CO2 and is widely used as a model for structure-guided inhibitor development. Previously reported crystal structures of human CA II (α-hCA2) in complex with the picomolar sulfonamide inhibitors 8V5 and 8V8 (PDB entries 5nee and 5nea) did not allow the complete ligand geometry of 8V5 to be resolved. To clarify the structural basis of binding, we determined high-resolution (1.5-1.7 Å) crystal structures of bovine CA II (α-bCA2) in complex with both inhibitors. α-bCA2 was selected as a crystallographica

p-Aminobenzene-Sulfonamide Derivatives of Substituted Pyrimidines as Human Carbonic Anhydrase Inhibitors.

The essential reaction of CO2 hydration, fundamental to all living organisms, is facilitated by the enzyme carbonic anhydrase (CA, EC 4.2.1.1). This enzyme plays a crucial role in regulating various physiological and pathological processes. A series of heterocyclic benzenesulfonamide derivatives (19 compounds) were evaluated as possible inhibitors of human CAs. Their inhibitory properties were tested against several isoforms such as the cytosolic hCA I and hCA II, as well as the transmembrane isoforms hCA IV, hCA IX and hCA XII. The tested molecules demonstrated notable inhibitory potential, p

Multi-target pyrazolopyrimidine-coumarin derivatives as potent CA IX/XII and tubulin polymerization inhibitors: Design, synthesis, and biological evaluation.

In the current medical era, developing multi-target anticancer agents that simultaneously engage complementary cellular vulnerabilities represents a promising strategy to overcome the inherent limitations of single-target therapeutics. Building on insights from our previous investigation, we synthesized a novel series of coumarin-pyrazolo [1,5-a]pyrimidine hybrids (13a-n) designed to target tumor-associated carbonic anhydrases (CA IX/XII) and tubulin polymerization through rational molecular hybridization. Structure-activity relationship analysis revealed that a zinc-binding sulfonamide group

Alternate day vs daily topical brinzolamide for the treatment of cystic maculopathy in inherited rod cone retinal degenerations.

Introduction: Cystic maculopathy (CM) occurs in 10-50% of inherited rod-cone retinal degenerations (RCD), with carbonic anhydrase inhibitors reported to show improvement, but rebound may occur. Alternate day treatment with topical Brinzolamide may alleviate this effect. We assessed treatment response in alternate day vs continuous day dosing. Methods: A retrospective chart review of patients with RCD undertaken, identifying those with CM, recording treatment regimen, vision, and OCT parameters. Outcomes were visual acuity (VA) improvement ≥3 letters significant, OCT reduction of mean cyst size

Molecular docking, MM/GBSA, FEP/MD, and DFT/MM MD studies on predicting binding affinity of carbonic anhydrase II inhibitors.

Human carbonic anhydrase II (hCAII) is one of the zinc-containing metalloenzymes that catalyzes various hydration reactions. We report an investigation of whether modern computational tools can predict inhibitory potency of a set of sulfonamides against hCAII. The methods used are molecular docking, molecular dynamics simulations coupled with the free energy perturbation theory (FEP/MD), molecular mechanics combined with the generalized Born and surface area continuum solvation (MM/GBSA), and quantum mechanics/molecular mechanics (QM/MM) metadynamics simulations. A comparison is presented betw

Trends in drug discovery for carbonic anhydrases: FDA approvals, clinical trials, molecular functions, and therapeutic potential.

Human carbonic anhydrases (CAs) catalyse the rapid conversion of CO₂ and bicarbonate, supporting pH balance, ion transport and many metabolic processes. All 15 human CA family proteins share a conserved structure, but they differ in activity, localisation and tissue distribution. Several CA isozymes are directly linked to distinct clinical conditions. Although CA-inhibiting drugs have long been clinically used, research across individual isozymes, chemical compound classes, and therapeutic uses has progressed unevenly. This review brings together current structural, biochemical and pharmacolog

Glycosyl Coumarins as Selective Inhibitors of Tumor-Associated Carbonic Anhydrase IX and XII: Synthesis, Structure-Activity Relationships, and Molecular Modeling.

Coumarins represent a distinctive class of non-classical carbonic anhydrase inhibitors that interact with the entrance region of the catalytic pocket rather than directly coordinating the catalytic Zn2+ ion. In this study, a series of glycosylated coumarins was synthesized through a copper-catalyzed multicomponent reaction involving propargyl glycosides, salicylaldehyde, and tosyl azide, providing efficient access to iminocoumarin-based glycosides derived from natural carbohydrates. The inhibitory activity of the synthesized compounds was evaluated against human carbonic anhydrase isoforms I,
