SARS-CoV-2 seeds IAPP aggregation and β-cell failure.

The global diabetes incidence has surged post-COVID-19, yet how SARS-CoV-2 drives long-term beta cell (β-cell) failure remains incompletely elucidated. We identified a mechanism whereby SARS-CoV-2 structural proteins directly hijack islet amyloid polypeptide (IAPP) to induce pathogenic amyloid deposition, disrupting insulin granule maturation, trafficking, and secretion in an age-susceptible islet microenvironment. In macaque models, aged subjects showed extensive islet IAPP aggregation with increased β-cell necroptosis, whereas adult infected animals exhibited minimal IAPP pathology. Multiple

Non-Newtonian blood rheology significantly alters hemodynamic predictions during cardiac looping: a computational study.

INTRODUCTION: Hemodynamic forces play a key role in early cardiac morphogenesis, yet many computational studies assume Newtonian blood behavior. Here, we evaluate the impact of non-Newtonian shear-thinning rheology on flow patterns, pressure distributions, and wall shear stress (WSS) during cardiac looping using idealized three-dimensional models of the embryonic heart tube. METHODS: Five geometries representing progressive looping stages, from a linear tube to an S-shaped configuration with ventricular ballooning, were analyzed under pulsatile flow using both Newtonian and power-law viscosit

Distant Homology BLASTP Searches.

Many neuropeptides have been widely conserved across species during evolution. Valuable insights may be gained by studying their biological activity in different species and identifying common functions. Standard basic local alignment search tool for proteins (BLASTP) searches often fail to detect homologous counterparts in distantly related species such as Drosophila and Caenorhabditis elegans, despite evidence for functional equivalents. This can be remedied by employing distant homology BLASTP with less stringent search parameters. Here, I describe the use of BLASTP searches with relaxed se

Structure of pancreatic hIAPP fibrils derived from patients with type 2 diabetes.

Type 2 diabetes (T2D) impacts the quality of life and lifespan of nearly 10% of the global population. Human islet amyloid polypeptide (hIAPP) constitutes a major component of islet amyloid deposition in patients with T2D, with hIAPP fibrils believed to play a key role in the pathogenesis of T2D. In this study, we determined the cryo-electron microscopy (cryo-EM) structure of hIAPP fibrils extracted from surgically resected pancreases of three donors with T2D. These fibrils exhibit a uniform morphology, comprising two symmetrical protofilaments encompassing residues 2-37 of hIAPP and adopting

Probing the Effect of α-Helical Stapling Strategies on the Inhibition of Peptide Aggregation and Amyloid Cytotoxicity.

Side chain-to-side chain peptide macrocyclization or stapling is a chemical modification that is frequently used to increase the metabolic stability, the cell permeability, and/or the binding affinity of peptide drugs. Interestingly, it was recently reported that α-helical stapling can also protect the amyloidogenic peptide hormone islet amyloid polypeptide (IAPP) from aggregation and amyloid-associated toxicity. IAPP is the major component of insoluble amyloid deposits found in diabetic patients, and its derivatives constitute potential therapeutic candidates to treat metabolic disorders. Her

The long-term effectiveness of the anti-obesity medication phentermine (LEAP) trial: Rationale, design, and baseline characteristics.

BACKGROUND: Current clinical practice guidelines support the long-term use of pharmacotherapy for obesity treatment. Historically, phentermine has been one of the most prescribed obesity medications (OMs), however, its long-term efficacy and safety have never been evaluated in a randomized trial as its market approval predates such requirements. Here we describe the design, rationale, and baseline characteristics for a 24-month, double-blind, randomized controlled trial evaluating the efficacy, cardiovascular risk, and safety of phentermine in adults with overweight or obesity. METHODS: This

Synergistic Effect of Mutations in the Amyloidogenic Core of Human Amylin (hAM) Affords a New Potent Inhibitor of hAM Fibrillation: Computational Design and Experimental Validation.

The human amylin, hAM, also known as the human islet amyloid polypeptide (hIAPP), fibrils are identified as a substantial risk element in type 2 diabetes (T2D). Thus, targeting hAM fibrillation could significantly influence the prevention and treatment of T2D. In this work, a library of computationally designed peptides based on the hAM amyloidogenic sequence NFGAILSS, an effective inhibitor of hAM fibrillation, has been generated, which is further evaluated for its ability to effectively attenuate hAM fibrillation. Notably, MM-PBSA analysis identified two new peptides, QYGPILSS (ΔGbinding = -

Islet Amyloid Polypeptide Modelled to Simulate Diabetes Co-Oligomerized with β-Amyloid 1-42 Reproducing the Pathological Cascade of Alzheimer's Disease in Human Cerebral Organoids.

Previous animal models of sporadic Alzheimer's disease (sAD), based on the β-amyloid (Aβ) cascade hypothesis and induced by Aβ1-42 oligomers (AβO), only recapitulated early AD pathological features. sAD cerebral organoids (COs) model employed Aβ coculture approach and found no typical features of AD pathology. Type 2 diabetes (T2DM) is one of the most important modifiable AD risk factors, so we hypothesize that T2DM could substantially exacerbate Aβ neurotoxicity and reproduce typical AD pathology. Human islet amyloid polypeptide (hIAPP) was used to mimic T2DM and was co-oligomerized with Aβ1-

Mammals that can develop type 2 diabetes have a similarly structured β-sheet amyloid oligomer.

Some mammals develop amyloid plaques and type 2 diabetes, much like humans, depending on the sequence of their islet amyloid polypeptide (IAPP). In humans, IAPP forms a toxic oligomer with a parallel β-sheet across residues 23FGAIL28S. Using two-dimensional infrared spectroscopy, we monitor the structure of IAPP from seven different mammals, five of which are from species that can develop type 2 diabetes (ferret, raccoon, cat, baboon, and human) and three from those that do not (hamster, rat, and pig). G24 is isotope labeled to monitor for the presence of the oligomeric β-sheet previously foun

A structural model of toxic amyloid oligomers involved in type 2 diabetes.

Amyloid oligomers of the human islet amyloid polypeptide (hIAPP) are a likely cytotoxic species driving β-cell death in type 2 diabetes, but their transient nature has precluded atomic-level structural characterization. We obtained a high-resolution structure of a physiologically relevant hIAPP oligomer. Using 2D IR spectroscopy, we identified three substitutions that slowed aggregation sufficiently for comprehensive 2D/3D NMR analysis while retaining the key wild-type structural features and cytotoxicity. The structural model reveals a dimeric assembly with N-terminal helices and a kink that

Trifunctional flavoenzyme-catalyzed asymmetric 4-alkyl-butenolide assembly in avenolide biosynthesis.

Butenolides are important features of many bioactive compounds. Current petrochemical-based methods for asymmetric butenolide synthesis are unsustainable. Biocatalysis could enable cheaper, greener, and more sustainable alternatives. However, known butenolide biosynthetic pathways are complex and challenging to harness. Here we show that avenolide, a 4-alkylbutenolide that regulates avermectin production in Streptomyces avermitilis, is assembled from a fatty acyl thioester by a multifunctional flavoenzyme and an iterative cytochrome P450. The flavoenzyme catalyzes homochiral 4-alkylbutenolide

APOE isotype-dependent regulation of IgG and IgA levels against different IAPP epitopes.

The efficient clearance of IAPP oligomers (IAPPo) by autoantibodies is crucial as increased plasma levels of IAPPo can induce microvascular alterations and Alzheimer's disease (AD)-characteristic amyloid-β deposition in the brain. We have recently demonstrated that plasma immunoglobulin (Ig) A levels against IAPPo, but not IgG, are reduced in an Apolipoprotein E (APOE) ε4 allele dose-dependent manner. In this study, we aimed to investigate if this APOE genotype-dependent impact can be explained by differences in IAPP epitope recognition by IgA and IgG. We found that the specificity for IAPP ep

In vitro metabolic profiling of weight-loss-inducing amylin receptor agonists in the context of preventive doping research.

Peptide hormone-based weight-loss therapeutics have gained increasing attention, driven amongst other reasons, by the clinical and commercial success of semaglutide. Their increasing accessibility raises concerns about their potential misuse in sports, especially in disciplines where weight management is decisive for athletic performance. Semaglutide has been included in the World Anti-Doping Agency's monitoring program since 2024. Given that amylin signalling is a key therapeutic target for next-generation weight-loss drugs, amylin receptor agonists such as pramlintide, cagrilintide and KBP-0

The story of amylin: from physiology to therapy.

Amylin is a glucoregulatory peptide hormone discovered in 1986. Almost 20 years later, pramlintide, a human amylin analogue, emerged as the first amylin-based drug, approved as an adjunct treatment to insulin for type 1 diabetes (T1D) and type 2 diabetes (T2D). Despite its effects on multiple organ systems, the therapeutic potential of amylin has remained relatively underexplored until recently, when growing interest in amylin has prompted advancement of several amylin-based therapies towards clinical use. This Review contextualizes the evolving therapeutic potential of amylin, focusing on rec

Modulating human IAPP aggregation in type 2 diabetes: inhibitors, mechanisms, and translational challenges.

Islet amyloid polypeptide (IAPP) aggregation is a characteristic pathological feature of type 2 diabetes (T2D), driving pancreatic β-cell dysfunction and loss through the formation of toxic oligomeric and fibrillar species. In recent years, significant advances in structural biology and chemical biology have deepened our understanding of the molecular basis of IAPP misfolding and enabled the development of various molecular strategies to modulate its aggregation pathway. This review summarizes recent advances in IAPP aggregation inhibitors, encompassing natural products, small synthetic molecu

Long-acting amylin-related peptides as therapies for obesity and type 2 diabetes.

The first therapeutically useful amylin receptor (AMYR) agonist, pramlintide was based upon the structure of non-aggregating rat amylin and found limited application as an adjunct to insulin therapy. It acts centrally to induce satiety, promote weight loss, suppress prandial glucagon release and reduce prandial hyperglycaemia. Recent understanding of the heterodimeric structure of amylin-calcitonin receptor complexes and of amylin structure-function properties has assisted the design of a second generation of non-aggregating, long-acting amylin analogues. These are now advancing in clinical de

Structural and morphological dynamics of "on-path" and "off-path" oligomers of human islet amyloid polypeptide.

The deposition of cytotoxic human islet amyloid polypeptide (IAPP) aggregates is a hallmark feature of Type 2 Diabetes. However, the structural evolution and cytotoxicity of IAPP aggregate species remain poorly understood. This study combines kinetics, biophysical and cell assays to resolve the morphological dynamics of IAPP aggregation. Using atomic force microscopy (AFM) and atomic force microscopy Infrared (AFM-IR) spectroscopy, we observed two distinctly different types of oligomers, donut-like (DO) and round oligomers (RO), formed at the early stages of protein aggregation. DO were domina
