Precursor-Encoded Metal-Free Carbon Dots Reshape the Bone-Regenerative Microenvironment: Materials Chemistry, Multiscale Mechanisms, and a Translational Roadmap.

Bone-defect healing is a spatiotemporally coordinated continuum involving inflammatory initiation and resolution, redox re-equilibration, immune-cell state transitions, angiogenesis, osteogenic commitment of mesenchymal stromal/progenitor cells, and osteoblast-osteoclast coupling. Although metal-containing nanomaterials can enhance regeneration through ion release, catalysis, or photothermal effects, their performance is often constrained by local dose, oxidation state, release kinetics, and long-term accumulation. Metal-free carbon dots (CDs) combine nanoscale interfacial activity, programmab

Going viral - Virus-targeting nanobodies for research, diagnostics and therapy.

The increasing prevalence of viral diseases represents a major threat to global human health, highlighting the urgent need for advanced tools to study viral infections, improve diagnostics, and develop effective antiviral therapies. Antibodies are central to the immune response against viruses and are widely used in research and diagnostics; however, their structural complexity and manufacturing process limit their areas of applications. Single-domain antibodies-better known as nanobodies-derived from heavy chain-only antibodies found in camelids have emerged as a promising alternative. Their

IL-2 and CD40L can establish and sustain a human B cell-mediated immune regulatory network.

UNLABELLED: B cells can control inflammation, but the factors governing B cell-mediated immune regulatory network remain unclear. We report that IL-2/CD40L enable class-switched memory (CSM) B cells to acquire an immunosuppressive phenotype characterized by the expression of surface TIGIT as well as multiple other inhibitory receptors (PD-1, LAG-3, GITR, CD39, and CTLA-4) and soluble molecules (IL-10, granzyme B, and IL-35). TIGIT + CSM B cells expressing functional CCR9 exhibit significant defects in total immunoglobulin secretion, while preserving IgA2 secretion. Furthermore, IL-2/CD40L-indu

Corneal Confocal Microscopy (CCM) in Patients With Central Neurodegenerative Disease.

Corneal confocal microscopy (CCM) is a rapid, non-invasive ophthalmic imaging technique that enables detailed visualization and quantification of small corneal nerve fibres. Although initially developed for the assessment of diabetic neuropathy, its application has broadened substantially, with increasing evidence supporting its value in a range of central neurodegenerative diseases. This narrative review synthesizes current findings on the use of CCM in dementia, multiple sclerosis, stroke, burning mouth syndrome, amyotrophic lateral sclerosis, trigeminal neuralgia, migraine and Parkinson's d

A novel CAF-related gene signature for prognostic and immunotherapy response prediction in PDAC based on single-cell RNA-seq and bulk RNA-seq data.

Increasing evidence shows that cancer-associated fibroblasts (CAFs) contribute to tumor progression, create an immunosuppressive microenvironment, and promote resistance to immunotherapy. However, the CAF-related gene signature in pancreatic ductal adenocarcinoma (PDAC) remains largely unexplored. In our study, we constructed a nine-gene CAF-related signature and found that it strongly correlated with the prognosis of PDAC patients in different cohorts. Subsequently, we found that the CAF-related gene signature was associated with the genomic alteration landscape, immune infiltration landscape

Mutations in Chromatin Regulator Genes Associate With TMB and Immunotherapy Response in Melanoma.

Melanoma is a high-tumor-mutational-burden (TMB) cancer, largely driven by ultraviolet-induced DNA damage. Although TMB predicts response to immune checkpoint inhibitors (ICIs), the genomic factors underlying its variability remain unclear. In addition to canonical DNA-repair defects such as mismatch repair (MMR) and POLE/POLD1 mutations, alterations in chromatin regulators (CRs) may influence genome stability and mutational load. We analyzed melanoma cohorts from the MSK-ICI, TCGA, MSK-Met, DFCI, and UCLA datasets. TMB was compared between CR-mutant and CR-wild-type tumors and overall cohorts

Neutrophil-to-lymphocyte ratio as a predictor of severity and adverse outcomes in complicated acute diverticulitis.

Complicated acute diverticulitis (CAD) requires accurate risk stratification to guide emergency management and surgical decision-making. Hemogram-derived inflammatory biomarkers, including the neutrophil-to-lymphocyte ratio (NLR) and platelet-to-lymphocyte ratio (PLR), may provide rapid and accessible prognostic information. This study evaluated the diagnostic and prognostic utility of these biomarkers compared with the APACHE II score in patients with CAD. This retrospective study included 204 adult patients with CT-confirmed CAD treated at a tertiary referral center in Mexico between March 2

Automated triage of non-informative frames in minimally invasive surgical videos: a comparative study of domain-specific vision transformers versus general-purpose multimodal models.

BACKGROUND: Minimally invasive surgery (MIS) videos support education and quality review, but raw recordings contain non-informative frames that impede review and increase storage. METHODS: We retrospectively analyzed 300,832 frames from 120 MIS videos performed by 30 surgeons. An EndoViT-initialized Vision Transformer (ViT) was evaluated using leave-six-surgeons-out cross-validation and compared with conventional vision models. GPT-4o-mini, LLaVA, and Doubao were separately evaluated under zero- and few-shot prompting on a balanced 400-frame subset that was not sample-size matched to the ViT

MRI evaluation of hypermobile lateral meniscus beyond distance measurements: the role of integrated popliteomeniscal fascicle assessment.

BACKGROUND: Hypermobile lateral meniscus (HLM) causes lateral knee pain and mechanical symptoms, but preoperative identification remains challenging because HLM is a dynamic abnormality that is not always captured on static magnetic resonance imaging (MRI). Previous MRI-based studies have mainly focused on linear measurements or qualitative popliteomeniscal fascicle (PMF) abnormalities. This study aimed to evaluate whether spatial MRI assessment beyond conventional linear distance measurements could help characterize arthroscopically confirmed HLM. We also aimed to determine whether MRI-derive

Optimal Resources for Children's Surgery 2: Updates on Training and Education Recommendations.

OBJECTIVE: The practice of pediatric surgery, and surgery in general, in low- and middle-income countries (LMICs) presents multiple challenges. Beyond limitations in physical and human resources, there is a great disparity in the quality and accessibility of training programs in LMICs. In response to input from high-income countries (HICs) and a desire to standardize training, regional associations have formed with the aim of facilitating knowledge sharing and enhancing surgical education around the world. Despite this, no clear guidelines exist regarding the types of initial training programs

Periprocedural body temperature and clinical outcomes after successful reperfusion in large vessel occlusion stroke.

BACKGROUND: Functional recovery remains heterogeneous among patients with large-vessel occlusion stroke, even after successful reperfusion. Body temperature may constitute a potentially modifiable physiological factor associated with functional outcome. In particular, whether clinical factors, including baseline stroke severity, modify this temperature-outcome association remains uncertain. METHODS: We retrospectively analyzed 219 patients with large-vessel occlusion acute ischemic stroke (LVO-AIS) who underwent EVT and achieved successful reperfusion, defined as a modified Thrombolysis in Ce

Modified frailty index versus risk analysis index for 30-day mortality after noncardiac surgery: a systematic review and meta-analysis.

BACKGROUND: mFI and RAI are widely used in noncardiac surgical risk stratification, but they differ in content and data requirements. Because construct validity and outcome discrimination are distinct, we evaluated their prognostic discrimination for 30-day all-cause mortality without treating either instrument as a criterion-standard multidimensional frailty measure. METHODS: This PRISMA-aligned, prospectively registered systematic review used two complementary evidence levels: tool-specific random-effects meta-analyses across independent cohorts and direct within-study comparison. Oligopeptide-binding protein A is a potential target for combating Salmonella infection.

INTRODUCTION: Salmonella infection remains a pervasive diarrheal illness, particularly affecting populations in developing countries. The lack of an effective vaccine against non-typhoidal Salmonella is attributable to antigenic diversity among serotypes, incomplete understanding of protective immune mechanisms, and the difficulty in identifying conserved antigens capable of eliciting broad cross-protective immunity. METHODS: To identify conserved Salmonella antigens, we established a Salmonella-specific monoclonal antibody library using heat-killed bacteria. Candidate antigens recognized by

TSC1-mTORC1 signaling in Alzheimer's disease: an integrated bioinformatic analysis linking gene expression to neuropathology.

BACKGROUND AND OBJECTIVE: Alzheimer's disease (AD) is characterized by the accumulation of amyloid-β (Aβ) plaques and tau pathology, with mTORC1 hyperactivation implicated in disease pathogenesis via suppression of autophagy. However, systematic evidence regarding its key upstream repressor, TSC1, in human AD brains remains limited. METHODS: Using an integrated, multi-level bioinformatic approach combining RNA-seq, neuropathological, and clinical data from the ROSMAP cohort, we comprehensively investigated the TSC1-mTORC1 axis in AD. Gene co-expression network analysis and gene set enrichment
