Physics-constrained and geometry-aware deep reconstruction for limited-angle CBCT.

Limited-angle cone-beam computed tomography (CBCT) is attractive for imageguided radiotherapy and imaging-guided procedure when full-arc acquisition is impractical, but the incomplete angular sampling leads to severe streaking, shading, and structural distortion. This study aimed to develop a physics-constrained and geometry-aware deep reconstruction framework for recovering anatomically faithful volumetric CBCT from limited-angle projections.
Approach: We propose PCGA-Net, a dual-domain reconstruction framework that combines projection completion, analytic geometric reconstruction, and im

Iodine-131 S-values of post-thyroidectomy adult Filipino mesh-type computational phantoms.

PURPOSE: Adult Filipinos are highly susceptible to differentiated thyroid carcinoma (DTC), which is managed with thyroidectomy followed by radioactive iodine (RAI) therapy. Currently, there is a lack of iodine-131 S-values for adult Filipinos undergoing RAI therapy post-thyroidectomy, posing internal dose assessment challenges. This study established iodine-131 S-values of post-thyroidectomy adult Filipino mesh-type computational phantoms, enabling organ-level internal dose assessment. METHODS: The phantoms were developed by scaling the adult mesh-type reference computational phantoms (MRCPs)

Targeting androgen receptor as a novel radiosensitizing therapy to improve long-term survival and anti-tumor immunity in glioblastoma via TGF-β/Smad3 Axis reprogramming.

Glioblastoma (GBM) remains the most aggressive primary adult brain cancer with limited therapeutic options. Our prior research demonstrated that androgen receptor antagonists (ARAs) enhance survival in GBM mouse models by preferentially suppressing glioma stem cells. This study investigates the potential of ARAs as radiosensitizers in combination with radiotherapy (RT). Combined effects of ARAs and RT were evaluated using an orthotopic GBM mouse model. RNA-Seq and TCGA analyses delineated AR-associated regulatory networks, while integrated cellular and molecular approaches utilizing human and

Preclinical Evaluation of [225Ac]Ac-NM600 Therapeutic Potential for Localized and Metastatic Triple-Negative Breast Cancer.

The absence of estrogen receptor, progesterone receptor, and HER2 expression limits the utility of targeted therapies in triple-negative breast cancer (TNBC). Chemotherapy is the standard of care for advanced TNBC but offers limited responses and an overall poor prognosis. We have developed several tumor-selective alkylphosphocholine analogs (NM600) for radiopharmaceutical therapy (RPT) of TNBC. Herein, we investigate the preclinical [225Ac]Ac-NM600 therapeutic efficacy and safety in two murine models of TNBC. NM600 was radiolabeled with 225Ac following previous methods. In vitro cytotoxicity

Peroxiredoxin 6 Silencing Remodels Lipid Metabolism and Redox Pathways to Promote Ferroptosis-Mediated Radiosensitization in Nonsmall Cell Lung Cancer.

Ionizing radiation (IR) induces oxidative stress and lipid peroxidation with ferroptosis contributing to radiation-induced cancer cell death. Peroxiredoxin 6 (PRDX6), a multifunctional enzyme involved in phospholipid repair and redox regulation, has been implicated in ferroptosis regulation, but its role in the radiosensitivity of cancer cells remains unclear. Here, we show that PRDX6 depletion enhances radiosensitivity in A549 lung cancer cells. PRDX6-silenced A549 cells exhibited elevated lipid peroxidation and labile iron levels, indicating a ferroptosis-prone state that was amplified by ra
