Background Alzheimer's disease (AD) is a progressive neurodegenerative disorder mainly characterized by progressive cognitive decline and memory loss. Identifying candidate biomarkers before the clinical onset of AD is crucial for enabling earlier diagnosis and timely therapeutic intervention. Among different molecular targets, N-acylsphingosine amidohydrolase 2 (ASAH2), a key enzyme in ceramide metabolism, has been linked to many neurodegenerative diseases, including AD. This study investigates ASAH2 expression in human serum and AD mouse models to explore its potential as an early biomarker

PURPOSE: Hepatoblastoma (HB) is the most common and lethal malignant solid tumor of liver in children. Targeting the progression of tumor glucose metabolism represents a promising strategy to attenuate the tumor promoting microenvironment. The Warburg effect, a hallmark metabolic feature of tumor cells, plays an important role in mediating tumorigenesis and progression. In this study, we elucidated the critical role of SKP2 in the development of hepatoblastoma. Materials and Methods SKP2 was identified by GEO database to play an important role in hepatoblastoma. The high expression of SKP2 in

BACKGROUND: The 2018 Farm Bill states that cultivars of Cannabis sativa L. (industrial hemp) are legal for industrial use if total tetrahydrocannabinol (THC) concentrations are less than 0.30%. Due to this legislation, hemp cultivars with low total THC have found a wide range of uses, from animal feed to paper production. Although cannabinoids are the most widely studied compounds in hemp, hemp produces numerous other compound classes as well, and these phytochemicals may have uses in the functional food and pharmaceutical industry.
METHODS: Initial liquid chromatography profiling of hemp roo

Hepatoblastoma (HB), the most common childhood liver cancer and the simplest of human neoplasms molecularly, commonly deregulates Wnt/β-catenin (B), Hippo/YAP (Y) and/or NFE2/NRF2 (N) signaling. Murine HBs that closely resemble human tumors can be generated by over-expressing pairwise or triple combination of mutant forms of B,Y and N. However, research is hampered by a paucity of established cell lines from either species. We show here that immortalized cell lines can be routinely established from murine HBs that over-express oncogenic B + Y and B + Y + N if the Cdkn2a tumor suppressor locus

Patients with familial adenomatous polyposis (FAP) have increased risk of hepatoblastoma (HB). We report monozygotic twins with HB in a FAP family. To explore genetic alterations in the HBs of the twins, we carried out whole exome sequencing (WES), RNA-seq, and immunohistochemical analyses of the tumors. Additional multiregional digital PCR was performed to profile clonality of each tumor. To determine a pathogenic germline variant in APC, Sanger sequencing was applied for the twins, the father, and the siblings of the father. A pathogenic variant of the APC gene was identified in the father a

BACKGROUND: Hepatoblastoma (HB) is the most common primary pediatric hepatocellular carcinoma, and the five-year survival rate for HB is the lowest among childhood cancers, as 20% of cases are chemotherapy resistant or unresectable. Understanding the global burden of HB and future trends in HB burden is urgently needed for gastroenterology and hepatology specialists and pediatricians. This study aims to provide a comprehensive assessment of the global burden of hepatoblastoma, predict future trends over the next decade, and offer insights into its management.
METHODS: Using data from the Glob

The enhancement of cell motility by bioactive molecules such as growth factors, hormones, and tissue factors is pivotal in cancer invasion and metastasis. However, the molecular mechanisms underlying this enhancement remain incompletely understood. In this study, we demonstrate that hepatoblastoma HepG2 cell motility is significantly increased following hepatocyte growth factor (HGF) treatment, as assessed by phagokinetic track assays, Transwell assays, and scratch assays. This enhancement is mediated by reactive oxygen species (ROS), which activate the PKCζ/Rho GTPase signaling pathway. Notab

BACKGROUND: The causal relationship between interleukin-7 levels and neuroblastoma risk remains unclear, and optimal radiation therapy timing lacks definitive evidence. This study investigated causal associations using Mendelian randomization while examining radiation therapy timing effects.
METHODS: We conducted two-sample Mendelian randomization analysis using GWAS summary statistics for interleukin-7 levels and neuroblastoma with three SNPs as instrumental variables. Multiple MRmethods included inverse variance weighted (IVW), MR-Egger, weighted median, and mode approaches. Additionally, 1

Paediatric liver transplantation (PLT) is a life-saving procedure for children with advanced liver disease or hepatoblastoma. The number of available grafts is limited in relation to the number of children on PLT waiting list. This graft shortage has led transplant societies and healthcare organizations to explore ways to investigate possible options and expand the donor pool. The safe use of grafts from obese donors has always been a subject of debate among PLT specialists. Donors' obesity is strongly associated with hepatic steatosis which can affect graft function by impairing microcirculat

Origanum vulgare L. is an important aromatic plant traditionally used in folk medicine since ancient times. Its growing interest for the scientific community is mainly attributed to its distinctive chemical profile, which includes bioactive compounds, such as polyphenols (phenolic acids and flavonoids) and volatile compounds (essential oil). These components collectively contribute to oregano's wide spectrum of biological activities. In this study, the volatile components of the essential oil (WEO_OR) and the polyphenolic fraction of the methanolic extract (ME_OR) obtained from leaves and infl

The incorporation of a palliative evaluation and care plan development for patients with serious illness is becoming a standard of care in modern medicine. For patients with complex challenges, palliative care physicians are well trained to offer specialized treatment plans and interventions to mitigate symptoms. This case presents the challenges of a mixed hepatocellular carcinoma and hepatoblastoma initially diagnosed in a 17-year-old male. Despite attempts at aggressive cancer-directed treatment, disease progression prompted referral for a palliative evaluation at age 20. Challenges unique

Congenital hepatic haemangioma is a rare benign vascular tumour in neonates. This case describes a neonate with an antenatally detected abdominal mass, suspected to be a hepatic neoplasm. Postnatal imaging revealed a large hepatic lesion with diagnostic uncertainty between hepatic haemangioma, hepatoblastoma and mesenchymal hamartoma. The infant developed progressive anaemia requiring transfusions, and he underwent surgical excision of the mass. Histopathological examination confirmed hepatic haemangioma, showing thin-walled vascular channels with areas of haemorrhage and fibrosis. This case b

BACKGROUND: Liver transplantation (LT) has been recommended for hepatoblastoma (HB) involving all four liver sectors at diagnosis (PRETEXT IV). However, recent studies question the universal indication for primary LT in PRETEXT IV HB.
METHODS: We reviewed 32 PRETEXT IV HB patients treated at our center from February 2007 to July 2024. A multidisciplinary team reassessed surgical resectability to evaluate treatment strategies for PRETEXT IV HB.
RESULTS: Of 32 patients, 28 (87.5%) underwent LT, while 4 (12.5%) underwent liver resection (LR). Median follow-up was 4.6 years (IQR 2.3-8.5). The tu

INTRODUCTION: Children with hepatoblastoma (HB) and other solid tumors frequently undergo intraoperative blood transfusion (IBT) with unknown impact on oncologic outcomes and scant data to guide transfusion in this population. This study tested the hypothesis that IBT is associated with poorer survival in children with HB.
METHODS: A multicenter retrospective observational study of patients aged <18 years with HB who underwent primary tumor resection, including liver transplantation, from 2010 to 2019 was performed at 19 institutions. The association of IBT with risk of recurrence and death w

BACKGROUND: Most patients with trisomy 18 (18T) die of cardiac or other severe anomalies within the first few years. Although surgical intervention for cardiac lesions or other diseases has improved these patients' prognoses in recent years, some survivors face the risk of developing neoplasms, such as hepatoblastoma. However, the treatment strategy remains controversial because of the poor prognosis. We evaluated the merits of aggressive therapeutic interventions for malignant tumors in 18T patients.
METHODS: We encountered five patients who underwent hepatoblastoma treatment within the last

BACKGROUND AND AIMS: Hepatoblastoma (HB) is a rare form of pediatric liver cancer currently treated with chemotherapy, which has major side effects, and surgery. We aimed to characterize the immune landscape of HB to improve our understanding of the immunologic contribution to this disease and explore immunotherapeutic options.
METHODS: To investigate the tissue landscape, a high-plex imaging mass cytometry panel was developed and applied to tissue of treatment-naïve HB and chemotherapy-treated HB, with paired adjacent normal liver tissue. Moreover, immunofluorescence was used to stain HB and
