Brain mechanisms supporting movement-evoked chronic low back pain: an observational brain imaging study.

BACKGROUND: Chronic low back pain (cLBP) is the most prevalent chronic pain condition and the leading cause of disability worldwide. Acute movement-evoked pain strongly predicts cLBP exacerbation, reflecting maladaptive amplification of nociceptive input, yet the neural mechanisms supporting movement-evoked cLBP flares remain poorly characterised. METHODS: A total of 114 participants with cLBP performed the cLBP-evoking leg raise test during perfusion-based arterial spin labelling functional MRI. Leg raise test-based pain intensity and unpleasantness ratings were assessed with an 11-point vis

Closing the Surveillance-to-Stewardship Gap in antimicrobial resistance: a narrative review of global lessons from WHO GLASS, UK ESPAUR, and low- and middle-income countries.

BACKGROUND: Antimicrobial resistance (AMR) surveillance has expanded faster than any other component of the global response, yet resistance continues to rise. In a growing number of settings, the principal constraint is the failure to translate available data into action, a disconnect we describe as the Surveillance-to-Stewardship (S2S) Gap. OBJECTIVES: To define the S2S Gap and its component gaps, examine where it binds across health systems of different maturity, analyse why it persists, and propose an operationalisable, testable framework for closing it. SOURCES: A cross-sectional analysi

Metabolomics in Microbial Genetic Resource Conservation: From Strain Identity to Functional Preservation.

The conservation of microbial genetic resources (MGR) is increasingly recognized as a strategic pillar for food security, biotechnology, ecosystem resilience, and climate change adaptation. Traditionally, ex situ conservation efforts have prioritized strain identity, viability, and genetic stability, relying mainly on cryopreservation and lyophilization as long-term storage strategies. However, growing evidence indicates that cellular survival and genomic integrity do not necessarily guarantee the preservation of microbial functionality, as preservation procedures may induce subtle yet biologi

In Silico Pipeline for the Development and Primer Design of Microsatellite Markers in Mangifera similis Blume.

Mangifera similis Blume, a wild relative of the mango, is an important genetic resource for improving mango varieties. This chapter outlines an in silico pipeline for the development and primer design of microsatellite (SSR) markers for the chloroplast and the whole genome of M. similis, using bioinformatics and next-generation sequencing. The method enables efficient microsatellite identification, saving laboratory time and costs, and is particularly useful for tree species with limited genomic data. The workflow comprises steps such as DNA extraction, sequencing, library construction, in sil

Optimized DNA Extraction from Coffee Roots Colonized by Arbuscular Mycorrhizal Fungi Using a Commercial Kit.

The molecular identification of arbuscular mycorrhizal fungi (AMF) from root tissues can often be limited by the co-extraction of polysaccharides, phenolic compounds, and other PCR inhibitors, particularly in woody or highly lignified roots, where cell lysis is also often difficult. Although commercial extraction kits offer standardized workflows and reduce the handling of harmful reagents, the manufacturer's protocols are insufficient for root samples with high levels of secondary metabolites. This protocol describes the optimization of DNA extraction from coffee (Coffea arabica L.) roots col

Purification of RNA G-Quadruplex Binding Proteins.

G-quadruplex (G4) nucleic acids have been shown to regulate a wide range of cellular processes, including replication, telomere maintenance, epigenetic landscape, and various aspects of transcription and translation. In this method, we describe an experimental purification procedure for identifying RNA G-quadruplex-binding proteins from nuclear extracts. Using biotinylated oligonucleotides allows immobilization to streptavidin-coated magnetic beads and convenient washing during the pulldown. Importantly, the experimental protocol presented here can distinguish protein interactions with structu

In Vitro RNA Pulldown and Validation of lncRNA-Protein Interactions.

Long noncoding RNAs (lncRNAs) play important roles in gene regulation and cancer progression through interactions with proteins, DNA, and other RNA molecules. Identification of lncRNA-protein interactions is therefore essential for understanding the molecular mechanisms by which lncRNAs regulate cellular processes. Among the available approaches, in vitro RNA pulldown coupled with mass spectrometry has become a widely used strategy for identifying proteins that associate with specific lncRNAs. In this chapter, we describe a detailed protocol for in vitro RNA pulldown using biotin-labeled RNA t

Capture and Purification of Endogenous Long Noncoding Ribonucleic Protein Complexes for Protein Identification Using Grad-MS.

The composition of long noncoding ribonucleic protein (lncRNPs) complexes has a major impact on lncRNA function, revealing a multitude of modalities for RNA-protein interactions. However, long noncoding RNAs (lncRNAs) also interact with an array of proteins during their processing that are not related to their function. For example, different proteins involved in splicing, maturation, and nuclear export are likely to interact with a newly transcribed lncRNA. Thus, a major limitation is the purification of fully complexed lncRNPs after the transient, intermediate RNA-protein interactions. Here

Development and Application of Expressed Sequence Tags (ESTs) for the Improvement of Cotton Crops.

Expressed sequence tags (ESTs), are short cDNA sequences obtained from expressed genes in any plant tissue of Gossypium spp. and represent a suitable approach for studying the transcriptome in the absence of a reference genome. In this study, protocols for EST development were established to explore expressed genes in cotton under diverse biotic and abiotic stress conditions. ESTs were generated by using two approaches: suppression subtractive hybridization (SSH) and next-generation sequencing (NGS) techniques. The process begins with RNA isolation, followed by mRNA purification, cDNA synthesi

Technical Guidelines for Cryopreservation and Lyophilization of Pathogenic Microorganism Strains.

Pathogenic microorganisms, defined as microbial entities capable of invading the host and causing infection or communicable disease, encompass a taxonomically diverse assemblage that includes viruses, bacteria, fungi, and rickettsiae.Cryopreservation is a technique that stores microbial cultures in a low-temperature environment, reducing their metabolism to the minimum or even to a complete standstill so that the organisms enter a semi-dormant or fully dormant state. Lyophilization (freeze-drying) is a preservation technique in which microorganisms are first frozen in a cryoprotective medium.

Isolation of Rhizospheric Bacteroidota Using Oligotrophic Supplemented Culture Media.

Isolation of Bacteroidota is crucial in microbial ecology laboratories because these organisms play important roles in the environment, including relevant interactions with plants and animals. Therefore, effective methods are needed for their successful isolation. Here, we describe modified Soil Extract (SE), Tryptic Soy (TS), and Reasoner's 2A (R2A) culture media that facilitate the isolation of Bacteroidota, primarily by addressing their auxotrophies and specific growth requirements. This approach enables both morphological and molecular identification of the isolates.

Conclusions and Perspectives on the Conservation of Microbial Genetic Resources.

Microbial genetic resources (MGRs) play a significant role in global ecological stability and in the development of modern biotechnology. In this context, in situ conservation strategies are essential to preserve the ecological interactions of the vast majority of non-culturable microorganisms, while ex situ conservation strategies ensure their availability in biobanks under controlled standards. This book compiles and systematizes the main strategies, methodological criteria, and experimental protocols aimed at the conservation of MGRs. In particular, effective methodologies for the conservat

Protocol for the Manipulation of Fossil Amber to Reduce Allergenic Effects.

Amber has a significant role in science, traditional medicine, art, education, and communication. Fossil amber inclusions are relevant for the study of the history and evolution of life on Earth. Amber has also been associated with allergic reactions in some individuals, and thus protocols should be implemented to reduce these risks when working with amber. In this chapter, we describe the experimental approaches to process fossil amber and measures to reduce the risks associated with allergic reactions to this fossilized tree resin. These methods allow the processing of amber pieces for the s

RAPD Marker Optimization and Genetic Diversity Assessment in Averrhoa Genus.

The genus Averrhoa includes economically and nutritionally important fruit crops like Averrhoa carambola (star fruit) and Averrhoa bilimbi (bilimbi). However, it has not been studied enough genetically, even though it is important for horticulture. For germplasm conservation, cultivar identification, and crop improvement programs, it is important to be able to accurately measure genetic diversity. This chapter elucidates the optimization and utilization of random amplified polymorphic DNA (RAPD) markers for the analysis of genetic diversity in Averrhoa species. We used a modified CTAB method t

Application of Single Nucleotide Polymorphisms (SNPs) in Sesame (Sesamum indicum L.) for Genetic Diversity, Mapping, and Marker-Assisted Selection.

Sesamum indicum L. is one of the oldest oilseed, cultivated oleaginous species, which is highly valued due to the excellent quality of the oils and owing to its antioxidative effects. Even though it is a major economic crop, there is a historical gap in breeding of sesame compared to the major staple crops, mainly because of a lack of genomic resources. With the emergence of next-generation sequencing methods, single-nucleotide polymorphisms (SNPs) with their noted benefits, compared to older marker systems, such as dense marker coverage, whole-genome coverage, and high-throughput automation c
