BACKGROUND AND AIM: Waterfowl, particularly Anas platyrhynchos and Cairina moschata, are important reservoirs and amplifiers of transboundary viral diseases, including Duck viral hepatitis (DVH), Avian influenza (AI), and Newcastle disease (ND). Despite their epidemiological significance, data from the Indonesia-Malaysia border region of West Kalimantan remain scarce. This study aimed to determine the seroprevalence and epidemiological patterns of DVH, AI, and ND in waterfowl populations in this high-risk transboundary interface.
MATERIALS AND METHODS: A cross-sectional study was conducted in

Despite the availability of vaccines and treatments for COVID-19, the emergence of new SARS-CoV-2 variants continues to challenge vaccine-induced immunity, emphasizing the need to develop antiviral therapies to combat these variants and other viruses. Umifenovir (UMF), marketed as Arbidol, is a broad-spectrum antiviral drug approved in Russia and China for influenza viruses A and B. Recent studies have suggested its potential against SARS-CoV-2, demonstrating its ability to inhibit viral replication and obstruct viral entry by targeting the spike protein, despite having low oral bioavailabilit

Seasonal influenza causes considerable morbidity and mortality, with influenza A and B viruses driving most influenza-associated hospitalizations and deaths. Vaccination remains a key influenza prevention strategy; however, current seasonal influenza vaccines based on traditional platforms provide inconsistent protection. Messenger RNA - based vaccines may offer several key advantages over other vaccines, including the flexibility to optimize antigen expression to enhance immunogenicity without the need for adjuvants. mRNA-1010 is an investigational seasonal influenza vaccine candidate that en

Swine influenza viruses (SIVs), particularly the H1N1 and H3N2 subtypes, pose ongoing threats to the swine industry and global public health. Current commercial vaccines generally provide limited cross-protection against heterologous viruses due to the frequent antigenic drift and shift in SIVs, highlighting the need for broad-spectrum vaccines. Self-assembling nanoparticles (NPs) can elicit strong and broad immune responses, representing a promising platform for the development of broad-spectrum nanovaccines. In this study, two newly-screened conserved epitopes (H1-3 and H1-5) and (H3-1 and H

Although vaccination is recognised as the optimal method to reduce the health burden associated with influenza virus infections, current vaccines regularly demonstrate low efficacy despite annual reformulation. To address the critical need for a more effective vaccine, our group has developed a gamma-irradiated influenza A virus (IAV) vaccine (γ-flu) and illustrated broad-spectrum immunity against drifted and heterosubtypic IAV infections following intranasal vaccination. For clinical development, highly purified vaccine preparations based on clinically relevant IAV isolates are required. Here

Squalene-based adjuvants like MF59 and AddaVax induce transient muscle injury, but their downstream mechanisms remain unclear. We show that intramuscular injection of a quadrivalent inactivated influenza vaccine (QIV) with AddaVax triggers a muscle regenerative-like immune response, increasing CX3CR1⁺Ly6C⁺ macrophages in muscle and draining lymph nodes within four days. This promotes a type II-skewed immune response, with elevated IgG1 titers and Th2 cytokines in the lungs after influenza challenge. In aged mice, the macrophage response and type II skewing are diminished. Adoptive transfer of

Current licensed influenza vaccines primarily protect by eliciting antibodies against the viral hemagglutinin (HA) glycoprotein, thereby blocking viral attachment and fusion with host cells. Unlike most vaccines, influenza vaccines must be administered annually because circulating viruses undergo continuous antigenic drift and population antibody titers wane over time. Despite yearly reformulation, influenza vaccine effectiveness remains highly variable, often below 45%, largely due to antigenic mismatches. These mismatches arise from ongoing HA evolution following strain selection and from eg

Recombinant measles virus (rMeV) vectors are promising platforms for vaccine development against emerging infectious diseases due to their safety, stability, and potent immunogenicity. However, conventional rMeV rescue systems frequently exhibit low efficiency, thereby constraining their scalability and throughput. In this study, we developed a modular, helper-virus-free and high-efficiency rescue platform based on an orthogonal transcription system utilizing orthogonal promoters and engineered RNA polymerases fused to an mRNA capping enzyme. This innovative system facilitated robust cytoplasm

Influenza D virus (IDV) is an emerging bovine respiratory pathogen associated with bovine respiratory disease complex (BRDC), highlighting the need for a safe and effective live attenuated vaccine. In this study, we developed a replication-restricted IDV vaccine candidate by introducing a point mutation (S57Q) into the esterase active site of the hemagglutinin-esterase-fusion (HEF) protein, rendering the virus incapable of replication in conventional host cells. To enable efficient virus production, we established a swine testis (ST) cell line expressing wild-type HEF under a doxycycline-induc

Peptides alone often exhibit limited immunogenicity, necessitating the development of effective antigen delivery systems to facilitate recognition and presentation, ultimately eliciting robust immune responses and activating T and B lymphocytes. Filamentous bacteriophages, such as M13, are recognized as efficient platforms for peptide expression and presentation via their capsid surfaces. The conserved amino acid sequence HA2 1-9 (GLFGAIAGF) from the haemagglutinin (HA) transmembrane protein of Influenza A virus (IAV) has been identified as a promising immunogen for eliciting broad-spectrum im

BACKGROUND: Hemagglutinin (HA), one of the main surface glycoproteins of the influenza virus, is essential for viral entry into host cells. Antibodies targeting HA can neutralize the virus, offering protection against infection. HA-specific antibody titers are therefore key markers of immune protection and vaccine efficacy. However, data on humoral immune responses to influenza A(H3N2) and influenza A(H1N1)pdm09 and viruses in India remain limited.
METHODOLOGY: Laboratory-confirmed influenza specimens from 2016 to 2017 were analyzed in this retrospective longitudinal study. A systematic rando

Influenza A viruses (IAVs) are significant respiratory pathogens characterized by high mutation rates and frequent genetic reassortments, underscoring the need for vaccines that can induce robust and broadly protective mucosal immunity. While replication-competent vesicular stomatitis virus (VSV) vectors have the potential to elicit mucosal immunity, their neurovirulence raises significant safety concerns. Herein, we report that a semi-replicating VSV (srVSV) vector, composed of one VSV with the glycoprotein (G) gene deleted (rVSVΔG) and another with the L gene deleted (rVSVΔL), has improved s

Influenza viruses continue to pose a significant threat to human and animal health. However, the limited number of licensed antivirals is increasingly compromised by drug resistance driven by high mutation rates. This highlights the urgent need for broad-spectrum therapeutics with novel mechanisms of action. Nanobodies, a new generation of antibody drugs, have great potential in the treatment of influenza virus infections. Neuraminidase (NA) mediates the sialic acid cleavage required for viral release, and its genetic drift is generally slower than that of hemagglutinin (HA) in influenza virus

BACKGROUND: Influenza caused by the influenza virus poses a significant global health threat, highlighting the urgent need for effective antiviral drugs. Ganhuangcao, a traditional Chinese medicine, is known for its heat-clearing, detoxifying, diuretic, and jaundice-reducing effects, but its anti-influenza properties and mechanisms have been scarcely studied.
OBJECTIVE: This study aims to elucidate the effects and molecular mechanisms of Ganhuangcao extracts, particularly its main component, gallic acid (GA), against influenza virus (H1N1) infection.
METHODS: Ganhuangcao extracts were prepar

BACKGROUND: Recent cases of H5N1 avian influenza spilling over into human populations have raised concerns once again about the unpredictable emergence of zoonotic viral diseases. This phenomenon highlights the persistent threat posed by viruses that jump from animals to humans, underscoring the need for preparedness for antigens that humans have not encountered. Nasal vaccines, which can effectively block transmission at the site of viral entry, represent a promising strategy and should be prioritised for stockpiling to halt zoonotic spillover.
METHODS: Here, we immunised a preclinical anima

Swine influenza virus (SIV) continues to evolve and possesses notable zoonotic potential, making it an important respiratory pathogen of concern for both the global swine industry and public health. Owing to antigenic drift, genetic reassortment, and regional lineage diversity, vaccine efficacy against SIV shows marked variability across different epidemiological contexts. Therefore, establishing appropriate animal models to dissect its pathogenic mechanisms, transmission characteristics, and immune response patterns is of critical importance. This review systematically summarises the animal m

Vaccination is the best way to combat annual influenza epidemics, yet the breadth of vaccine-induced humoral immunity toward decades of future differentially evolving influenza A (IAV) and influenza B (IBV) viruses is unclear. Using historic 1994 influenza vaccination cohorts of young and older adults, we defined antibody responses elicited by 1994 vaccination against future influenza strains spanning three decades of differentially evolving IAV and IBV strains. Quality of antibody responses and vaccine-induced and cross-reactive B cell memory responses were investigated. Vaccination increased

BACKGROUND: There is an urgent need to understand the immune correlates of protection against avian influenza viruses (AIV), where pre-existing immunity may be limited.
METHODS: Here, we characterized the antibody response in 12 severely ill A(H7N9) patients and examined its association with early-life imprinting and clinical outcome.
RESULTS: We find that A(H7N9) patients imprinted with A(H2N2) during early life show minimal H7-IgM and a rapid IgG response across diverse hemagglutinin subtypes. They also have more high avidity H7-antibodies compared to older or younger patients. Early antib

Clade 2.3.4.4b highly pathogenic avian influenza A(H5N1) viruses continue to expand geographically and across mammalian hosts, raising concern about pandemic potential. The degree and specificity of pre-existing immunity in humans are key determinants of this risk. We analyzed hemagglutinin (HA)- and neuraminidase (NA)-specific antibody responses in 300 sera collected from adults in New York City. While HA directed binding antibodies to clade 2.3.4.4b H5 were low and hemagglutination-inhibiting antibodies were absent, we detected widespread binding and functional NA antibodies against N1 neura
