Poloxamer-Based Th1-Biased NKT Cell Nano-Agonist for Tumor Immunotherapy.

BACKGROUND: Natural killer T (NKT) cells bridge innate and adaptive immunity and are promising targets for tumor immunotherapy. However, its agonist α-GalCer induces NKT cells to secrete both Th1-type and Th2-type cytokines. Coupled with the challenges of in vivo delivery, these drawbacks collectively limit its clinical application. METHODS: In this study, we constructed Th1-biased NKT cell nano-agonists (LGCP Ms) by using Th1-preferred glycolipids (Lys-α-GalCer) and Poloxamer through thin-film hydration combined with probe sonication to enhance NKT cell-mediated anti-tumor immunity. RESULTS

Emerging Lipid Nanoparticle Targeting Strategies for Extrahepatic Delivery of RNA Therapeutics.

Lipid nanoparticles (LNPs) have transformed RNA therapeutics through their high delivery efficiency, scalable manufacturing, and favorable safety profiles, as demonstrated by COVID-19 mRNA vaccines and clinically approved small interfering RNA (siRNA) therapeutics. However, extending RNA delivery beyond the liver to extrahepatic tissues, including the spleen, lungs, lymph nodes, brain, and tumors, remains challenging because of the intrinsic hepatotropism of conventional LNPs and their limited cell-type specificity. Recent advances in lipid chemistry, particle engineering, surface functionaliz

Palmitoyltransferase DHHC13 controls lysosomal cholesterol egress through palmitoylating Niemann-Pick C1.

Protein S-palmitoylation provides a key regulatory mechanism for diverse cellular activities. Although palmitoylated proteins are frequently observed in cholesterol-enriched membrane microdomains, how protein palmitoylation reciprocally regulates cholesterol metabolism remains elusive. Here, we identify DHHC13 as a critical regulator linking protein palmitoylation to lysosomal cholesterol egress. Through an shRNA screen, we found that lack of DHHC13 led to pronounced lysosomal cholesterol accumulation. Mechanistically, DHHC13 directly palmitoylated Niemann-Pick C1 (NPC1). Loss of palmitoylatio

Third lumbar transverse process syndrome with acute abdominal pain as the main clinical manifestation: A case report.

RATIONALE: Third lumbar transverse process syndrome is an uncommon musculoskeletal cause of referred abdominal pain. Because acute abdominal pain is appropriately evaluated first for visceral, vascular, urinary, and surgical emergencies, a spinal or paraspinal pain generator may be overlooked. PATIENT CONCERNS: A 66-year-old man presented with persistent left lower-quadrant abdominal pain radiating to the left lower back. Baseline pain intensity was 7/10 on a VAS, and the pain was aggravated by postural change, standing, and walking. DIAGNOSES: Serial laboratory tests, endoscopy, ultrasonogr

Polymeric Nanoparticle Gene Delivery to Reprogram the Tumor Immune Microenvironment in Ovarian Cancer for an Anti-Tumor Response.

The tumor immune microenvironment (TIME) in ovarian cancer is highly immunosuppressive, and existing immunotherapies are not effective in 85-90% of patients. To address this, we fabricated nanoparticles (NPs) composed of biodegradable poly(β-amino ester) (PBAE) complexed with plasmids encoding 4-1BB ligand and interleukin-12, for delivery into ovarian cancer cells. Collectively with the intrinsic major histocompatibility complex (MHC):antigen signals in tumor cells, these NPs generate tumor-associated antigen-presenting cells (tAPCs), leading to a T-cell-mediated anti-tumor response in an anti

Decoding extracellular vesicle-biomolecule interactions: How the biomolecular corona shapes extracellular vesicle biodistribution and clearance in vivo.

Extracellular vesicles (EVs) are promising endogenous nanocarriers, yet how biomolecules adsorption shapes their in vivo fate remains unclear. Here, we examine how biomolecular corona formation regulates the biodistribution, cellular recognition, and clearance of extracellular vesicles. Using EVs derived from macrophages and breast cancer cells, together with artificially prepared cell membrane-derived vesicles (CVs) generated from the corresponding parental cells as controls, we observed markedly different circulation and clearance behaviors despite their comparable physicochemical properties

The advances and challenges of mRNA-LNP application in fields of hematology and oncology therapeutics.

The rapidly global deployment of COVID-19 mRNA-LNP vaccines established the clinical feasibility of combining RNA medicine with LNP delivery systems, transforming a long-standing experimental concept into a scalable therapeutic modality, previously considered impossible. Currently, the value of mRNA-LNPs has been expanding from infectious disease vaccines to cancer immunotherapy, personalized neoantigen vaccination, in vivo immune-cell engineering, protein replacement therapy, mRNA-encoded biologics and genome editing, with particular relevance to hematology and oncology. However, in hematolog

Clinically Significant Portal Hypertension in Hepatocellular Carcinoma: Non-Invasive Assessment Across the Continuum.

PURPOSE OF REVIEW: Clinically significant portal hypertension (CSPH) is a key determinant of surgical candidacy, locoregional therapy safety, and systemic treatment outcomes in hepatocellular carcinoma (HCC), yet its non-invasive assessment remains challenging because the Baveno criteria were derived in HCC-free cirrhosis. This review synthesises current evidence on non-invasive tests (NITs) for CSPH across the HCC care continuum, organised around three clinical scenarios: pre-operative assessment, on-treatment monitoring, and pre-immunotherapy evaluation. RECENT FINDINGS: Liver stiffness-bas

TNFα promotes brain-predominant EAE independent of IFNγ and IL-17A in HLA-DR3 transgenic mice.

Multiple sclerosis (MS) is thought to be mediated by human leukocyte antigen (HLA) class II-restricted autoreactive CD4 T cells producing interleukin (IL)-17A and/or interferon γ (IFNγ), but the necessity of these cytokines for disease pathogenesis is unclear. Utilizing HLA-DR3 (DRβ1*0301) transgenic mice lacking IFNγ or IL-17A in experimental autoimmune encephalomyelitis (EAE), a model of MS, we found that neither cytokine is essential for disease induction, although both have distinct roles. The absence of IFNγ caused atypical, brain-predominant EAE characterized by high levels of IL-17A, wh

Stereotyped CLL-like immunoglobulin rearrangements in healthy repertoires: implications for leukemogenesis.

Chronic lymphocytic leukemia (CLL) is characterized by a highly biased B-cell receptor (BcR) immunoglobulin repertoire and by recurrent stereotyped receptors defined as quasi-identical BcR recurring in unrelated patients. High-throughput sequencing studies have shown that immunoglobulin rearrangements resembling these CLL stereotypes can also be detected in healthy individuals. Beyond their relevance to leukemogenesis, these findings provide a distinctive opportunity to investigate how recurrent BcR configurations are generated, selected, and maintained during normal B-cell development. In thi

Casein Hydrolysate Peptides Attenuate Immunosenescence and Inflammaging in a D-Galactose-Induced Mouse Model of Aging.

Aging populations worldwide face increasing burdens from physiological deterioration. Bioactive peptides derived from milk proteins have attracted interest for their biological activities, yet systematic evidence against aging remains limited. Here, we investigated casein hydrolysates in a D-galactose-induced murine aging model. Following eight weeks of oral administration at graded doses, we quantified systemic oxidative stress markers, inflammatory cytokine profiles, hepatic and cerebral histopathology, and multiple immune competence parameters. Our data demonstrate that casein peptides dose

Effects of pyridaben on the spleen and T lymphocytes and intervention strategy in grass carp (Ctenopharyngodon idella).

The widespread use of pyridaben (PDB) poses serious threat to aquatic organism health. 6-Gingerol, a bioactive component of ginger, has been demonstrated to possess both antioxidant and immunomodulatory properties. However, the effects of spleen injury of PDB in aquatic species and the potential mitigating effects of 6-Gingerol remain insufficiently characterized. The present study aimed to characterize the toxic effects of PDB on the spleen of grass carp and to evaluate whether 6-Gingerol could effectively counteract PDB-induced toxicity. The results showed that PDB exposure could induce sple

Canonical and Non-Canonical Regulators of Lipid Metabolism in the Liver and the "Liver-Other Organs" Axes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common chronic liver disease and is closely associated with obesity, insulin resistance, and the recently conceptualized cardiovascular-kidney-metabolic syndrome. For decades, the pathogenesis of steatosis has been viewed primarily through the lens of insulin and the canonical transcriptional programs it regulates-sterol regulatory element-binding protein 1c, carbohydrate response element-binding protein, peroxisome proliferator-activated receptors, and AMP-activated protein kinase. At the same time, recent data point

Brain-Specific Quantification of Somatostatin Receptor Expression: SUVmax and SUVRsss as Quantitative Correlates of the Krenning Score in Meningioma.

Somatostatin receptor type 2 is highly expressed in meningiomas and can be quantified using [68Ga]Ga-DOTATATE PET. The Krenning score, originally developed for [111In]In-octreotide scintigraphy, remains the standard visual grading system for somatostatin receptor expression and is widely used to guide eligibility for peptide receptor radionuclide therapy. However, visual grading may not optimally translate to intracranial disease. This study evaluated the relationship between brain-specific quantitative PET parameters-SUVmax and SUV ratio normalized to the superior sagittal sinus (SUVRsss)-and

Splenic Volume Changes After Left Nephrectomy: A Retrospective Computed Tomography-Based Morphometric Study.

Background/Objective: Spleen volume can vary depending on physiological conditions, systemic diseases, and various clinical factors. However, the relationship between the anatomical changes resulting from the surgical removal of an organ adjacent to the spleen and spleen volume has not been sufficiently studied. The aim of this study was to evaluate changes in spleen volume following left nephrectomy and to investigate the associations of sex and the time interval between preoperative and postoperative computed tomography (CT) examinations with these changes. Methods: In this retrospective stu

MRI Anatomy of Pseudopus apodus: A Descriptive Study with High-Field and Low-Field MRI.

Pseudopus apodus is an anguimorph lizard belonging to the order Squamata. Anatomical information on this species remains limited, particularly regarding soft tissues, and has primarily focused on the axial skeleton of both living and fossil specimens. Recently, the anatomy of its coelomic cavity has been described using dissection and multi-detector computed tomography (CT), and its brain has been characterized neuroanatomically by combining ultra-high-resolution magnetic resonance imaging (MRI) with histological analysis using immunofluorescence. In the present study, four specimens (one fema

GoAstV Induced Mitophagy and Ferroptosis in the Spleen of Goslings.

A novel goose astrovirus (GoAstV) has emerged in various provinces of China, leading to a mortality rate of up to 50% among GoAstV goslings. The spleen is an important immune organ in geese. Studies have shown that goslings infected with GoAstV exhibit higher viral loads in their spleens, and the spleens sustain damage. However, the mechanism underlying its induction of spleen injury remains to be elucidated. Therefore, a gosling infection model with GoAstV was established in this experiment. The Con group was subcutaneously injected with 0.2 mL of saline, while the GoAstV group was subcutaneo
