Hydrogel for sustained and localized delivery of a regulatory T cell-inducing agent promotes bone regeneration.

Local activation of regulatory T (Treg) cells represents a promising strategy for wound repair and tissue regeneration. However, current Treg-based therapies rely on adoptive cell transfer, which requires ex vivo expansion and reinfusion, posing challenges to donor variability and tissue specificity. Here we engineer a hydrogel-based system for in situ Treg activation at injury sites through the controlled release of AS2863619 (AS), a cyclin-dependent kinase inhibitor. AS is incorporated into collagen hydrogels at concentrations exceeding its solubility limit, reducing burst release and extend

Translational biology of human cardiac fibrosis.

Cardiac fibrosis is a pathological process that arises as a compensatory response to injury or mechanical stress and is a common feature across diverse cardiovascular diseases. Here, we review recent advances in the translational biology of human cardiac fibrosis, highlighting insights from single-cell studies of human cardiac fibroblasts and genetic analyses of fibrotic phenotypes using cardiac imaging. We discuss the heterogeneity and origins of cardiac fibroblasts, the role of immune-fibroblast cross-talk in fibrogenesis, and the utility of imaging and circulating biomarkers for assessing f

Cytokine circuits in CNS homeostasis and disease.

Cytokines are critical regulators of central nervous system (CNS) integrity, acting not only as immune messengers but also as signals that coordinate interactions among glial and neuronal cells, resident macrophages, such as microglia and border-associated macrophages, and infiltrating leukocytes. These circuits establish basal immune tone, shape responses to perturbation, and promote tissue adaptation and recovery. In this Review, we examine selected cytokine pathways across homeostasis, inflammatory or degenerative tissue damage and repair to develop a framework for how cytokine networks pre

Precursor-Encoded Metal-Free Carbon Dots Reshape the Bone-Regenerative Microenvironment: Materials Chemistry, Multiscale Mechanisms, and a Translational Roadmap.

Bone-defect healing is a spatiotemporally coordinated continuum involving inflammatory initiation and resolution, redox re-equilibration, immune-cell state transitions, angiogenesis, osteogenic commitment of mesenchymal stromal/progenitor cells, and osteoblast-osteoclast coupling. Although metal-containing nanomaterials can enhance regeneration through ion release, catalysis, or photothermal effects, their performance is often constrained by local dose, oxidation state, release kinetics, and long-term accumulation. Metal-free carbon dots (CDs) combine nanoscale interfacial activity, programmab

Genetic prioritization of HSDL2 in suicide and intentional self-harm: multi-omics evidence linking lipid metabolism and neuronal vulnerability.

BACKGROUND: Suicide or other intentional self-harm (SSH) represents a critical global public health burden, yet biologically informed targets remain poorly defined due to its complex and incompletely understood etiology. This study aimed to prioritize genetically supported candidate targets for SSH and characterize their biological and translational relevance. METHODS: We developed a multi-omics evidence framework integrating transcriptomic and proteomic Mendelian randomization, colocalization, and sensitivity diagnostics. The primary SSH outcome was from FinnGen R12 (11,538 cases; 488,810 co

Association of Serum GDF-15 with Bone and Muscle Parameters in Community-Dwelling Individuals.

INTRODUCTION: Muscle mass and bone mineral density decrease with aging, although following different patterns among males and females. Growth differentiation factor 15 (GDF-15) is a stress-induced cytokine and has emerged as a key biomarker for inflammation, tissue injury, and metabolic stress. We tested the association between GDF-15 and specific bone and muscle parameters in a large cohort of community-dwelling subjects. METHODS: The SardiNIA survey included 4,842 subjects (2,795 females). All underwent a detailed medical visit and laboratory testing. Peripheral quantitative computed tomogr

A robust lightweight deep learning network with deformable convolutions for efficient segmentation of optical coherence tomography images.

INTRODUCTION: Manual segmentation and evaluation of optical coherence tomography (OCT) images are difficult and rely on specialist training, which limits the output in a busy clinic. While automatic segmentation can ease this burden, the deep models that currently lead the field tend to be too heavy for routine deployment on standard clinical hardware. A further obstacle is the unusual geometry of OCT data: instead of compact objects on a background, the structures of interest form long, undulating tissue strata that extend horizontally across the whole frame, and this layout is poorly matched

SnoRNA Evolution, Genomic Context and Mobility: The Role of Function and Host Gene Association.

Small nucleolar RNAs (snoRNAs) are best known for guiding chemical modifications of ribosomal and spliceosomal RNAs. Most vertebrate snoRNAs are encoded within introns of host genes and therefore depend on the production and processing of host precursor RNAs for their biogenesis. Despite this close genomic association, how host-gene context is associated with snoRNA abundance and evolution remains poorly understood. Here, we combined human snoRNA annotations from snoDB, Ensembl gene annotations, snoRNA-sensitive abundance data across multiple tissues, and vertebrate orthology analyses to syste

The evolution of direct transcranial photobiomodulation: a history of dose, depth, and delivery.

Direct transcranial photobiomodulation (dtPBM) delivers red or near-infrared light through the scalp and skull with the aim of affecting brain tissue. The field has produced encouraging findings but inconsistent clinical results. We reconstruct its development as a history of scientific and engineering decisions in which optical access, target depth, and dose repeatedly shaped what could be tested. The clinical lineage began with light therapy for ischemic myocardium and moved to stroke, where cadaver measurements established that near-infrared light could traverse the human scalp and skull an

Synthetic activation of gibberellin signalling reveals spatial coordination of root growth.

Gibberellins (GAs) influence cell division and elongation, profoundly shaping plant architecture and yield. GA perception occurs when bioactive GAs bind the receptor GID1, promoting DELLA degradation and activating transcriptional programs. While GA signalling in the root endodermis is essential for promoting root elongation, the contributions of other layers have remained unclear. Here we developed a synthetic GA (sGA) that exhibits little or no binding to endogenous GID1, together with a modified GID1 (mGID1) engineered to selectively recognize sGA, enabling cell-specific activation of GA si

[Analysis of the response of the OsLFNR2 gene in rice to shading treatment].

In a previous study, quantitative trait locus (QTL) mapping identified the gene LOC_Os06g01850, which is associated with oxidative damage resistance in rice, and it was further confirmed to encode a chloroplast-localized ferredoxin-NADP+ oxidoreductase, namely OsLFNR2. To gain deeper insight into the function of OsLFNR2, bioinformatics approaches were employed in the present study to determine its evolutionary conservation and to predict cis-acting elements related to stress and light responses in its promoter region. Using the japonica rice cultivar Nipponbare (Oryza sativa L. ssp. japonica)

[Genome-wide identification, expression profiling, and natural allelic variation analysis of the ckx genes in sorghum (Sorghum bicolor)].

Cytokinin (CK) is one of the essential plant hormones, and cytokinin oxidase (CKX) is the key enzyme for CK degradation. Currently, the ckx gene family of rice (Oryza sativa) and maize (Zea mays) have been characterized, while a genome-wide analysis of the ckx gene family in sorghum (Sorghum bicolor) has not been reported. This study aims to identify the Sbckx gene family in sorghum, analyze its evolutionary characteristics and expression patterns, and screen for key genes. A combined approach of bioinformatics analysis and experimental validation was utilized to examine sorghum subjected to a

Pesticides commonly used in aquaculture: Interactive and repetitive effect of azamethiphos and deltamethrin on the physiology and growth of the mollusc Mytilus chilensis.

The intensive use of antiparasitic pesticides in salmon aquaculture has raised concerns regarding their impacts on non-target marine organisms. In Chile, the organophosphate azamethiphos and the pyrethroid deltamethrin are widely applied to control infestations of the sea louse Caligus rogercresseyi, yet their combined and repetitive effects on ecologically and economically important species remain poorly understood. This study evaluated the physiological and growth responses of juveniles Mytilus chilensis exposed to concentrations of azamethiphos (15 and 100 μg L-1) and deltamethrin (1 and 2

Influence of yoga on the epigenetics of obesity: a study on its short- and medium-term benefits in the treatment of patients with severe/morbid obesity eligible for bariatric surgery.

BACKGROUND: Obesity is a growing global health problem characterised by chronic low-grade inflammation and epigenetic dysregulation. Epigenetic mechanisms-potentially reversible changes in gene expression occurring without alteration of the DNA sequence, most notably DNA methylation-are increasingly implicated in obesity-related inflammation. Yoga may act as complementary therapy. OBJECTIVE: To explore whether participation in a structured yoga program is associated with changes in DNA methylation of TNF-α, and with clinical, hormonal, metabolic and psychological parameters, in patients with
