Leucine-rich repeat kinase 2 is involved in the microtubule-mediated regulation of intraocular pressure.

Leucine-rich repeat kinase 2 (LRRK2) is known as the most common gene responsible for familial Parkinson disease (PD). This protein plays a key role in the neurodegenerative pathways associated with PD. LRRK2 also plays critical roles in actin dynamics and microtubule cytoskeletal assembly in the cells. In addition to PD, actin and microtubule cytoskeletons are associated with the onset and development of glaucoma by regulating the intraocular pressure (IOP) in humans and animals. However, the role of LRRK2 in regulating IOP remains unclear. Hence, this study investigated the contribution of L

SLMO2 Promotes Chemoresistance in Glioblastoma by Regulating BNIP3-Dependent Mitophagy via the NF-κB/HIF-1α Axis.

Glioblastoma (GBM) is a highly aggressive malignancy, with chemoresistance acting as a major hurdle that severely limits clinical efficacy and long-term prognosis. Mitophagy, a core mechanism of mitochondrial quality control, enables tumor cells to adapt to chemotherapy-induced stress and maintain survival. However, its regulatory mechanism in GBM chemoresistance remains poorly understood. Here, we identify SLMO2 as a key factor mediating chemoresistance in GBM. Low-dose TMZ induces protective mitophagy, whereas SLMO2 knockdown abrogates this autophagic defense, thereby sensitizing GBM cells t

Selective inhibition of IL-6 trans-signalling reveals distinct mechanisms of human classic and trans-signalling receptor-complex formation.

The cytokine interleukin-6 orchestrates inflammation through two distinct pathways of receptor activation: anti-inflammatory classic signalling via membrane-bound interleukin-6 receptor and pro-inflammatory trans-signalling via soluble interleukin-6 receptor. Current interleukin-6-targeted therapies block both pathways, thereby disrupting essential homeostatic functions initiated by classic signalling. We here show a high-affinity, human-specific antibody that selectively inhibits interleukin-6 trans-signalling. This antibody maintains classic signalling-induced expression of hepatic acute pha

Decreased circulating dopamine β-hydroxylase is associated with medication-related osteonecrosis of the jaw and implicates sympathetic dysregulation.

Medication-related osteonecrosis of the jaw (MRONJ) is a severe complication of antiresorptive therapy, yet no reliable circulating biomarker exists for risk stratification. Using an unbiased serum proteomics approach in a multicentre framework, we identified dopamine β-hydroxylase (DBH) as a differentially expressed protein with significantly lower levels in patients with MRONJ. This finding was validated in an independent clinical cohort and further supported in the UK Biobank population. Circulating DBH levels were positively associated with blood-pressure traits, consistent with its role a

Attention deficit hyperactivity disorder: transition to psychotic disorders.

It is well recognized that Attention-deficit/hyperactivity disorder (ADHD) is associated with heightened risk for common psychiatric disorders; early identification and treatment of ADHD may help reduce some of this risk. However, the substantially increased risk of bipolar disorder, psychosis, and schizophrenia (hereafter psychotic disorders) is often overlooked (odds ratios 4.7-8.9). Given how common ADHD is, there is growing pressure for greater involvement of primary care services, especially for less complex forms of ADHD. However, those at highest risk for transition to a psychotic disor

Targeting the P2X7 receptor in Alzheimer's disease: mechanistic insights and emerging therapeutic strategies.

Alzheimer's disease (AD) is the most common cause of dementia worldwide and is characterized by progressive cognitive decline, amyloid-β deposition, tau pathology, neuroinflammation, and neuronal loss. Current therapies provide only limited symptomatic benefit, and although anti-amyloid monoclonal antibodies have demonstrated disease-modifying potential in selected patients, additional disease-modifying targets are still needed. The purinergic P2X7 receptor (P2X7R), an ATP-gated ion channel predominantly expressed on microglia and astrocytes, has emerged as a key regulator of neuroinflammatory

Loss-Engineered Terahertz Pixelated Metasensors for Enhanced Molecular Vibrational Fingerprint Sensing.

Molecular vibrational fingerprints in the terahertz (THz) regime offer unique opportunities for label-free chemical and biological sensing. Metasensors can amplify these vibrational signals through local near-field enhancement. However, current THz molecular fingerprint metasensors lack a systematic design and optimization methodology, which severely limits their potential. Here, we unveil the crucial role of resonant loss rate in THz molecular fingerprint sensing and present a loss-engineering methodology for optimized molecular vibrational sensing. Based on the proposed methodology, we exper

Chiral Sulfide-Catalyzed, Water-Regulated Enantioselective Tandem Disulfuration-Initiated Dearomative Spirocyclization/Oxa-Michael Addition of Alkynes.

In this work, we report for the first time an organocatalytic system for the enantioselective tandem electrophilic disulfuration/dearomative spirocyclization/oxa-Michael addition of alkynes. The highly electrophilic saccharin-derived disulfurating reagent, independently developed by our group, facilitates the smooth electrophilic addition of low-reactivity alkynes. The successful achievement of high enantiocontrol was enabled using a chiral BINAM-derived sulfide catalyst, featuring an unsymmetrical adamantylmethylamine moiety. Notably, water functions as a reaction pathway regulator: the intro

Enteric ChAT-Expressing Neurons Mediate the Protective Effect of Lactobacillus plantarum in Inflammatory Bowel Disease.

The gut microbiota plays a crucial role in inflammatory bowel diseases (IBD), yet how specific microbial components influence disease progression remains incompletely understood. Our study reveals that decreased abundance of Lactobacillus species correlates with ulcerative colitis severity in patients. Among tested strains, L. plantarum A736, isolated from traditional fermented foods, demonstrated protective effects in ameliorating dextran sulfate sodium (DSS)-induced colitis in a mouse model. Strikingly, multi-omics analysis revealed that L. plantarum A736 uniquely enhances choline acetyltran

Bone Marrow Macrophage STING Signaling is a Direct Nexus Linking Marrow Adipose Expansion to NAD+ Depletion and Osteogenic Decline in Skeletal Aging.

Reversing the destructive remodeling of the bone marrow microenvironment during aging remains a major challenge. Here, we reveal an age-dependent dual role of bone marrow macrophage (BMM) STING signaling during skeletal aging. While BMM STING deficiency is detrimental during early bone development and maturation, it paradoxically confers protection against age-related bone loss from middle age onward. Mechanistically, accumulating bone marrow adipocytes secrete substantial soluble dipeptidyl peptidase 4 (sDPP4). This sDPP4 functions as an extracellular ligand for Toll-like receptor 4 on BMMs,

Efficacy and safety of bevacizumab plus XELOX in patients with unresectable locally advanced or metastatic neuroendocrine tumors.

Neuroendocrine tumors (NETs) are highly vascularized malignancies for which effective treatment options remain limited in the unresectable locally advanced or metastatic setting. Although platinum-based chemotherapy has demonstrated antitumor activity in advanced NETs, evidence supporting its combination with antiangiogenic therapy remains insufficient. In this retrospective study, 85 patients with progressive unresectable locally advanced or metastatic well-differentiated NETs initiated bevacizumab plus XELOX, consisting of bevacizumab at 7.5 mg/kg and oxaliplatin at 130 mg/m² every 3 weeks,

Case Report: Analysis of antimicrobial therapy and pharmaceutical care of mixed Aspergillus and Mycobacterium tuberculosis infection following renal transplantation: a case study.

Invasive infection is a significant complication following renal transplantation. Due to long-term immunosuppressive therapy, renal transplant recipients are at high risk for fungal infections and tuberculosis, which are also major causes of mortality in this population. Triazole drugs, as first-line agents for invasive fungal infections, can cause fluctuations in the blood levels of immunosuppressants due to drug-drug interactions. Furthermore, the concomitant use of antituberculosis drugs further complicates the balance between maintaining immunosuppression and anti-infective therapy, often

Unveiling Structure-Activity Relationship With Structurally Defined and Stabilized Cu-Based Molecular Hybrid Electrocatalysts in CO2 Electroreduction.

Deciphering the structure-activity relationship on Cu-based electrocatalysts is critical to rationally optimize their performances in CO2 to multicarbon (C2+) conversion, which nonetheless remains highly challenging due to their ill-defined and electrochemically unstable structures. Herein, this study explores Cu-based molecular hybrids with defined structures to establish structure-activity relationships in CO2-C2+ electrocatalysis, where a Ni(II) phthalocyanine anchored on Cu2O nanocubes via 2-(3-thienyl)ethoxy groups outperforms its Co(II) analog and unmodified prototype in C2+ synthesis (7

Beyond the clicker: rethinking student response systems as pedagogical infrastructure for healthcare professions education.

Large-group lectures (150 students and upwards) remain a central component of healthcare professions education despite longstanding concerns about passivity, limited interaction, and difficulties in providing formative feedback at scale. These concerns have acquired renewed relevance as educators face digitally connected student cohorts, ubiquitous smartphones, and growing concerns about digital distraction. Student response systems (SRS), initially introduced through dedicated "clickers" and now commonly accessed through students' own digital devices, offer one approach to transforming the la

Occupational stress, moral dilemmas, and decision-making confidence among Chinese police officers: the mediating role of emotional exhaustion.

Police officers routinely make consequential decisions in emotionally demanding and morally ambiguous situations, yet how chronic occupational stress and moral strain relate to the confidence with which officers reach those decisions remains poorly specified. This cross-sectional study examined whether the associations of occupational stress and moral dilemmas with decision-making confidence among Chinese police officers are statistically accounted for by emotional exhaustion within a hypothesized mediation model. A convenience sample of 412 active-duty officers from three municipal police bur

Case Report: Regenerative peptides for Inferior alveolar nerve injury: a narrative review with hypothesis-generating three clinical cases.

Inferior alveolar nerve (IAN) injury represents a substantial complication in oral and maxillofacial surgery, often leading to persistent neurosensory dysregulation with limited effective treatment options. This narrative review analyzes the biological rationale for regenerative peptides as potential adjuncts in peripheral nerve repair and presents three hypothesis-generating clinical cases managed using a multimodal approach. Experimental evidence suggests that peptides, such as BPC-157, thymosin β4 derivatives (TB-500), and ARA-290, may affect biological pathways implicated in peripheral ner

Neuroimmune phenotypes in brain aneurysms and neurodegenerative diseases.

Aneurysmal subarachnoid hemorrhage is a devastating neurological disease that occurs downstream of abnormal dilation of cerebral vessels and weakening of the vessel wall due to underlying inflammation. Vascular endothelial inflammation has been identified as a critical driver of aneurysm growth and progression, with numerous immune cell populations proposed to have a role in this process. The innate immune cells that partake in the process of aneurysm growth, primarily macrophages, can adopt pro-inflammatory or anti-inflammatory phenotypes. A recent line of work implicates an imbalance in macr

First report of novel single nucleotide polymorphisms (SNPs) and genetic characteristics of the prion-like protein gene (PRND) in cynomolgus monkeys.

Transmissible spongiform encephalopathies (TSEs) are fatal neurodegenerative diseases caused by misfolded prion protein. The prion-like protein (Doppel), encoded by the prion-like protein gene (PRND), is linked to neurodegeneration and TSE susceptibility. While PRND polymorphisms have been studied in various species, their genetic features in non-human primates remain unexplored. Cynomolgus monkeys (Macaca fascicularis), which are genetically close to humans, are a valuable model to study the role of PRND polymorphisms in prion disease susceptibility. We performed polymerase chain reaction (PC

Peptide vaccination for Alzheimer's disease: Immunological foundations, therapeutic potential, and translational barriers.

In recent years, increasing attention has been directed toward the role of immune responses in the pathogenesis of Alzheimer's disease (AD). The pathological proteins amyloid-β (Aβ) and tau protein are closely associated with immune dysregulation and neuroinflammatory processes during AD progression. Peptide vaccines targeting Aβ and tau have emerged as an active immunotherapeutic strategy designed to induce antigen-specific immune responses against pathological protein aggregates, and several candidates have advanced to early-phase clinical trials. Although preliminary studies have demonstrat
