BACKGROUND: Microdeletions and microduplications are chromosomal variants ranging up to 3 Mb in size. These abnormalities often arise spontaneously and have significant clinical implications, including developmental delay and congenital anomalies. Given the resolution of conventional preimplantation genetic testing for aneuploidy is > 5-10 Mb, these clinically significant abnormalities are missed. This study presents two cases where preimplantation genetic testing-whole genome sequencing successfully identified small, clinically significant abnormalities.
CASE PRESENTATION: In the first case,

Scientific advancements offer new possibilities for developing novel treatments for children with monogenic neurodevelopmental disorders (mNDDs), targeting the underlying effects of their condition. Although, in qualitative studies, parents of children with mNDDs have predominantly positive attitudes towards the development of novel treatments, a few parents express concerns about whether treatments could change who their child is. Existing literature on personal identity, including literature on how personal identity is affected by medical (neuro-)interventions, can assist in clarifying these

Congenital anophthalmia is the complete absence of the eye resulting from a failure of optic vesicle formation during early embryogenesis. It may present as a unilateral or bilateral condition and can occur as an isolated anomaly or as part of a syndromic disorder. We report the case of a full-term newborn admitted on the second day of life for the management of bilateral congenital anophthalmia. Family history was notable for anophthalmia in three cousins belonging to the shared parental lineage, suggesting a possible genetic predisposition. The diagnosis is primarily clinical and is confirme

OBJECTIVE: To explore the genetic etiology for a pregnant woman with a history of multiple adverse pregnancies and assess the phenotype-genotype correlation of 22q11.2 microduplication syndrome in her family.
METHODS: Amniotic fluid sample was taken from a pregnant woman for whom non-invasive prenatal screening indicated chromosome 22 abnormalities in the fetus. Peripheral blood samples from the woman, her brother and parents were collected for high-throughput low-depth whole genome sequencing (CNV-seq). A pedigree traceability analysis of the results was conducted in conjunction with analysi

Maternal uniparental disomy of chromosome 15 [UPD(15)mat] and imprinting defects account for about 30% of cases of Prader-Willi syndrome (PWS). Mosaic UPD(15)mat is rare and may escape routine testing. We describe a 45-year-old male patient in whom persistent clinical suspicion of PWS was not genetically confirmed by repeated methylation-based analyses. Clinical manifestations included neonatal hypotonia with low birth weight, early hyperphagia, severe obesity, short stature, growth hormone deficiency, type 2 diabetes mellitus, dyslipidemia, and MASLD/MASH with compensated cirrhosis. He presen

BACKGROUND: 22q11.2 deletion syndrome (22q11.2DS) may be overlooked in adults when neurological care is prompted by pyramidal, gait, or parkinsonism-mimicking presentations rather than by congenital, developmental, psychiatric, or systemic features.
METHODS: We retrospectively reviewed the clinical, imaging, and genetic findings of two unrelated adults whose neurological presentations led to whole-exome sequencing-based copy-number variant (CNV) detection of 22q11.21 deletions. Orthogonal confirmation by chromosomal microarray, multiplex ligation-dependent probe amplification (MLPA), quantita

BACKGROUND: Waardenburg syndrome (WS) is a group of autosomal dominant hereditary disorders characterized by auditory-pigmentary abnormalities. Haploinsufficiency of paired box 3 (PAX3) gene is one of the known pathogenic mechanisms. However, clinical phenotypes are difficult to predict precisely in fetuses harboring PAX3 haploinsufficiency. In this study, we report a family with a PAX3 deletion encompassing exons 1-4, in which clinical manifestations ranged from normal to mild abnormalities.

Chromosome 11q13 microdeletion syndrome, or otodental syndrome, involves dental, auditory, and ocular anomalies linked to deletions in the 11q13.2q13.4 region. We report a 1-year-old girl with a 43 kb deletion of the FGF3 gene on chromosome 11q13.3, exhibiting otodental dysplasia, hearing difficulty, and developmental delay. Her family history includes permanent childhood hearing loss and otodental syndrome. Chromosomal microarray analysis (CMA) and sequencing confirmed a complete heterozygous deletion of FGF3. This case suggests that FGF3 haploinsufficiency is sufficient to cause the syndrome

Terminal deletions of chromosome 14q are characterized by a spectrum of phenotypes that can include microcephaly, growth deficiency, intellectual disability, characteristic facial features, and various congenital anomalies. The rarity of this syndrome, together with the broad spectrum of phenotypes, has made genotype-phenotype correlations difficult. Herein, we describe a family with the core features of the condition and a heterozygous 3.7 Mb deletion at 14q32.32qter. To our knowledge, this is the first case of vertical transmission of a terminal 14q deletion. In addition to this family, we r

Kleefstra Syndrome (KLEFS1) is a monogenic neurodevelopmental disorder characterized by developmental delays, somatic issues, and (neuro)psychiatric symptoms. Individuals with KLEFS1 often require complex, ongoing support, which significantly impacts family life. To effectively improve care and support for families, a better understanding is needed of the impacts of raising a child with KLEFS1 on the family and families' support needs. We conducted a semi-structured interview study among parents of children with KLEFS1 to investigate these impacts. The interviews were audio-recorded, transcrib

Background Congenital anomalies (CA) are a major contributor to infant mortality worldwide, and the risk of CA differs by maternal and fetal characteristics. Moreover, studies establishing the etiology of CA are methodologically heterogeneous, and its investigation among the Hispanic population is limited. This study aims to investigate the potential etiologies for CA and compare their etiologic profile across pregnancy outcome, fetal/infant sex, and maternal age in a Mexican population-based setting. Methods Potential causes of CA from 2022 to 2024 at Durango General Hospital (northwest Mexic

INTRODUCTION: Orofacial clefts are common congenital malformations that may occur in isolation or as part of a syndrome. Early prenatal diagnosis provides crucial information for parental counseling, delivery planning, and multidisciplinary neonatal care. This study aimed to review orofacial cleft cases diagnosed during the prenatal period and to assess the relationship between cleft type, associated anomalies, genetic findings and pregnancy outcomes.
METHODS: This retrospective study included all fetuses with a prenatal diagnosis of cleft lip and/or cleft palate who received obstetric care a
