The additional role of contrast-enhanced ultrasound in identifying lesions in pathological nipple discharge patients: a prospective study comparing with DCE-MRI.

OBJECTIVE: To evaluate the auxiliary role of contrast-enhanced ultrasound (CEUS) in the etiological diagnosis of pathological nipple discharge (PND) and compare its effectiveness with dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI). METHODS: In this prospective study conducted from March 2021 to February 2024, PND patients underwent breast ultrasound and DCE-MRI after initial history and physical examination. Lesions were categorized as mass or non-mass-like on baseline US and assessed for vascularity using colour Doppler. CEUS was utilized to refine the breast imaging reportin

Evaluating the association between upstream perceived individual and neighborhood determinants of health and intensity of breast cancer screening.

Exposure to negative perceived individual and neighborhood determinants disproportionately impacts marginalized groups and can profoundly shape health behavior. We assessed the hypothesized relationship between exposure to adverse perceived individual and neighborhood determinants and rates of mammography. In this cohort study, we identified 31 568 female participants aged 40-74 without history of breast cancer in the All of Us Research Program. Participant-reported levels of perceived stress, everyday discrimination, perceived neighborhood physical disorder, perceived neighborhood social cohe

Long-Term Outcomes and Oncologic Implications of Histopathological Findings in Breast Reduction Specimens: A 15-Year Follow-Up Study.

BACKGROUND: Breast reduction surgery provides both functional and aesthetic benefits and enables incidental detection of breast pathology. Although high-risk or malignant lesions are uncommon, their long-term clinical relevance remains unclear. OBJECTIVES: The aim of this study was to evaluate the histopathological findings of breast reduction specimens and assess their relationship with long-term breast outcomes, including the development of new breast disease and the need for subsequent biopsy or mastectomy. METHODS: This retrospective cohort study included 1349 patients (2630 breasts) who

Neoplasms arising at the CIED pocket: a hybrid study combining a case report, scoping review, and clinical survey.

Cardiac implantable electronic device (CIED) implantation is a minor surgical procedure, with a low complication incidence. Neoplasm occurrence at the CIED pocket is a rare event, and sporadic cases are reported without a comprehensive evaluation. This hybrid study aims to fill this gap through a three-part approach: a new case report, a scoping review with individual patient data (IPD) analysis, and a clinical survey. In March 2024, a male patient with a history of high-grade urothelial cancer referred to our Department with a mass at the CIED pocket. Incisional biopsy confirmed the diagnosis

Mutational landscape of normal breast tissues adjacent to invasive breast cancer.

Individuals with a history of breast cancer are at increased risk of developing a new breast cancer during their lifetime. Rare but high-impact somatic mutations in normal breast tissues may contribute to malignant transformation. We analyze mutations in cancer-relevant pathways across matched samples of peripheral blood, cancer-adjacent normal breast, and breast cancer from patients diagnosed before 50 years of age who carry no germline mutations in cancer-predisposing genes. Gene- and pathway-level mutation profiles and single-base substitution (SBS) signatures are compared between tissue ty

Performance of different polygenic risk scores for breast cancer risk prediction: in-depth evaluations across large UK and Australian cohorts.

Polygenic risk scores (PGS) have the potential to support enhanced, risk-based screening for breast cancer. Previous studies for many diseases found that genome-wide PGS (GW-PGS) outperform PGS derived by applying hard GWAS significance thresholds. To support future breast cancer risk predictions, we compared the predictive performance of two existing PGS (including PGS313, a leading hard-thresholding PGS) and five newly developed GW-PGS (applying different methods to recent GWAS). We evaluated the performance of PGS Z-scores and of predicted 5-year absolute breast cancer risks based on age al

Black-White Racial Differences in Background Parenchymal Enhancement at Breast MRI.

Background Women with high background parenchymal enhancement (BPE) at contrast-enhanced MRI have up to a fourfold increased risk of breast cancer, even after adjusting for breast density. However, no studies have examined BPE specifically among Black women, despite their higher breast cancer-specific mortality and typically lower breast density. Purpose To examine Black-White racial differences in contrast-enhanced MRI-derived BPE. Materials and Methods This retrospective cross-sectional study examined the data of Black and White women without a history of breast cancer who underwent both mam

European Code Against Cancer, 5th edition - organised cancer screening programmes.

The 5th edition of the European Code Against Cancer (ECAC5) recommends sustainable, organised screening programmes for: (a) colorectal cancer using biennial quantitative faecal immunochemical test (FIT) for individuals aged 50-74 years. As an alternative strategy, once-only endoscopy may be considered within the same age range; (b) breast cancer using biennial digital mammography for women aged 50-69 years. Implementing this strategy for women aged 45-49 years and 70-74 years can be considered. Other screening strategies or additional examinations could be considered for women with high mammog

Deep learning-based multimodal pathogenomics integration for precision cancer prognosis.

BACKGROUND: Recent studies have revealed valuable prognostic insights in haematoxylin and eosin (H&E)-stained histological sections and transcriptomic profiles, suggesting potential applications in machine learning. However, existing methods lack sufficient intra- and inter-modal interactions, and face challenges in clinical validation due to incomplete multimodal data. METHODS: We proposed PathoGems (PathoGenomics-based integrative survival prediction), a weakly-supervised, interpretable multimodal learning framework that integrates histology and genomic profiles for precise cancer prognosis

Diabetes and psychological distress posed greater risks than model-inferred risk of radiation-induced cancer after the Fukushima disaster: An application of the loss of happy life expectancy indicator.

After disasters, direct deaths and secondary health effects, such as diabetes and psychological distress, can occur. After the 2011 Fukushima disaster, radiation exposure was extremely limited; however, secondary health effects increased significantly. It is critical to compare the magnitudes of these model-inferred risks to promote lifespan development and effective public health measures. Here, we used the loss of happy life expectancy (LHpLE) to compare the model-inferred risks associated with diabetes, psychological distress, and radiation-induced cancer after the disaster. Two questionnai

Causative Role for a BRCA2 Germline Pathogenic Variant in External Auditory Canal Squamous Cell Carcinoma.

External auditory canal squamous cell carcinoma (EACSCC) is rare, affecting 1.6 in a million individuals. We report a case of EACSCC in a 66-year-old woman carrying a heterozygous BRCA2 germline pathogenic variant (GPV) (c.8537_8538del), with prior history of breast cancer. Tumor copy-number analysis showed loss of heterozygosity at the BRCA2 locus. Genomic scar analysis supported homologous recombination repair deficiency (HRD), with mutational signatures showing the predominance of APOBEC activity and lower contributions of HRD-associated single base (SBS3, SBS8) and INDEL (ID6) signatures.

Associations of smoking with the risk of second smoking-associated primary cancer among adults with a history of cancer.

Associations between smoking and smoking-associated second primary cancers (SSPC) have been documented predominantly among survivors of smoking-associated first primary cancers (FPC), with limited evidence among survivors of non-smoking-associated FPCs. We examined survivors of FPCs diagnosed from 1992 to 2015 in the Cancer Prevention Study II Nutrition Cohort, followed through June 30, 2017. Self-reported smoking status, intensity, and years since quitting were collected a mean of 1.6 years before FPC diagnosis. Among 28,272 survivors (58.8%, male; mean age at FPC diagnosis, 72.2 years [SD=7.

Evaluating the Test-Retest Reliability and Measurement Error of a Sit-Stand-Power Test in Individuals Treated for Cancer.

INTRODUCTION: Muscular strength and physical function are commonly assessed in exercise oncology trials, yet muscular power is often overlooked despite being an important determinant of morbidity and mortality. The sit-to-stand power (STSp) test offers a rapid, low-cost method to evaluate lower-body power. The purpose of this study was to assess the test-retest reliability, measurement error, and minimal detectable change (MDC) of the STSp test assessed using a linear power transducer in individuals treated for cancer. METHODS: Adults with a history of cancer completed the STSp test on 2 occa
