Women in their 40s responding to the 2024 United States Preventive Services Task Force mammography guideline update: a focus group study.

OBJECTIVE: Focus groups involving US women aged 40-49 were conducted to identify women's attitudes towards the 2024 United States Preventive Services Task Force (USPSTF) mammography guideline, particularly the change in emphasis away from informed choice. Focus groups explored what information women feel they need to receive about screening benefits and harms prior to getting a mammogram. DESIGN: Seven focus groups of 4-6 women were conducted 3-4 months following the USPSTF guideline announcement. SETTING: Participants were recruited from two healthcare systems in the USA and participated in

Challenges in Achieving US Population Awareness of Alcohol as a Carcinogen: Identifying Predictors of Continued Lack of Awareness after Educational Video Exposure.

BACKGROUND: Alcohol use increases the risk of seven cancers, including breast and colorectal cancers. Awareness of this is low in the US population (34%), which impedes risk reduction efforts, support for alcohol regulation policies, and physician and family communication about this topic. Efforts to improve awareness of the cancer risks of alcohol will require multiple approaches, including the use of educational messages tailored to audience needs. There is a lack of evidence about how existing messaging influences awareness and characteristics associated with awareness that could guide futu

Results of the E-intervention for Protein Intake and Resistance Training to Optimize Function (E-PROOF) study among older cancer survivors.

INTRODUCTION: Many older adults with a history of cancer do not meet diet or exercise recommendations, leading to suboptimal physical function. The E-PROOF study is the first synchronous, online, protein-focused dietary and resistance training randomized controlled trial among older cancer survivors. This study determined the feasibility, acceptability, and exploratory intervention effects (improving physical function, diet quality, and exercise) over the 12-week intervention. MATERIALS AND METHODS: Eligibility criteria included adults age ≥ 65 years, with breast, colorectal, or prostate canc

Diagnostic and Risk Stratification Value of Serum 8-Hydroxy-2-Deoxyguanosine in Women with Suspected Breast Carcinoma in Bangladesh: A Cross-Sectional Analytical Study.

Breast carcinoma is a leading cause of carcinoma-related morbidity and mortality among women worldwide, with a rising burden in low- and middle-income countries. Early differentiation between malignant and benign breast lesions remains a major challenge in resource-constrained settings, where access to imaging and histopathology is often limited. Oxidative DNA damage plays a key role in carcinogenesis and serum 8-hydroxy-2-deoxyguanosine (8-OHdG) has emerged as a potential non-invasive biomarker; however, evidence from Bangladesh is scarce. The objective of this study was to evaluate the diagn

Risk-reducing mastectomy and salpingo-oophorectomy in women with hereditary breast and ovarian cancer: a single-institute experience following coverage by Japanese national medical insurance.

BACKGROUND: Risk-reducing mastectomy (RRM) and risk-reducing salpingo-oophorectomy (RRSO) are preventive options for women with hereditary breast and ovarian cancer (HBOC). The Japanese national medical insurance began covering RRM and RRSO for patients with HBOC in April 2020. METHODS: We retrospectively analyzed 59 individuals with pathogenic germline variants (PGVs) from 55 families diagnosed with HBOC between 2010 and 2024 to assess the prevalence of RRM and RRSO after April 2020. RESULTS: The median age at diagnosis was 50 years. Ten individuals (16.9%) were diagnosed before April 2020,

Decision-making surrounding ovarian cancer risk-reducing surgery: perspectives from a diverse population.

OBJECTIVE: Individuals with BRCA1/2 pathogenic variants (PV) are recommended risk-reducing salpingo-oophorectomy between 35 and 45 years of age to prevent ovarian cancer. Trials evaluating the oncologic safety of risk-reducing salpingectomy are ongoing. One's decision on the type and timing of surgery has implications for fertility, quality of life, and long-term health. Published literature on patient preferences has focused on White women from North America and Europe; thus, findings are not necessarily generalizable to patients from other backgrounds. To address this gap, we sought to asses

PROMs: Patient related or patient centred? A bridge between Patient-Reported Outcome Measures (PROMs) and clinical ethics in the context of a cohort of gynaeco-oncological patients analysed by qualitative semi- structured interviews.

BACKGROUND: Patient-reported outcome measures (PROMs) are widely used in oncologic care, yet it remains unclear to what extent they capture the depth and contextual meaning of patient experience in gynecologic oncology. OBJECTIVE: To explore gynecologic oncology patients’ narratives of treatment experience, needs, and psychosocial impacts, and to examine how these narratives align with domains covered by commonly used PROMs. METHODS: We conducted semi-structured, in-depth interviews with 20 female participants (31–72 years) across different therapy phases. Interviews were analysed using a cons

Uterine serous carcinoma and germline genetic testing: patterns of referral, completion and pathogenic variant detection.

BACKGROUND: Current guidelines recommend consideration of germline genetic testing for patients with uterine serous carcinoma (USC) but real-world data are limited on completion rates of testing and pathogenic variant (PV) identification. This study aimed to evaluate the rate of genetic testing referral and completion in a cohort of patients with USC, determine the prevalence of clinically meaningful PVs found on testing and explore factors associated with genetics referral and testing completion. METHODS: We retrospectively examined the medical records of all individuals diagnosed with USC b

Evolving Cancer Characteristics Among World Trade Center Survivors: An Updated Analysis from the WTC Environmental Health Center.

Local community populations ("survivors") exposed to the World Trade Center (WTC) disaster experienced complex exposures to mixtures of dust and combustion products with potential carcinogenic effects. Survivors with certifiable WTC-related conditions are eligible for inclusion in the federally funded WTC Health Program. We provide an updated description of cancers in the WTC Environmental Health Center (EHC), a program for WTC survivors, through 31 December 2024. Using data from the WTC EHC Pan Cancer Database, we summarized demographics, exposure history, and tumor characteristics among enro

Comprehensive Germline Profiling of High-Grade Serous Ovarian Cancer Using Whole-Exome Sequencing.

While ovarian cancer screening is not recommended in the general population, attention has shifted to screening women with elevated hereditary risks. Although germline BRCA 1/2 pathogenic variants account for 40% of inherited ovarian cancer risk and family history (FH) remains important, known germline variants alone do not fully explain familial ovarian cancer risk. Whole-exome sequencing (WES) was performed on blood samples taken from 231 individuals, including 39 patients with high-grade serous ovarian cancer (HGSOC) and 192 healthy controls (HCs) stratified by FH. We analyzed pathogenic or

Bilateral Vortex Keratopathy in a Patient Receiving Long-Term Ribociclib Therapy.

We report a case of bilateral vortex keratopathy associated with long-term ribociclib therapy in a 67-year-old woman with metastatic breast cancer. The corneal deposits were identified during routine ophthalmic examination after 4 years of continuous treatment. Slit-lamp biomicroscopy revealed bilateral whorl-like epithelial deposits with a subtle morphological appearance compared with classic amiodarone-associated cornea verticillata. The patient had no history of tamoxifen, amiodarone, or chloroquine use, and there were no clinical features suggestive of Fabry disease. The combined severity

Hereditary Endometrial Cancer: Lynch Syndrome, Mismatch Repair Deficiency, and Emerging Genetic Predispositions-A Comprehensive Review with Clinical and Laboratory Guidelines.

Endometrial cancer is the most common gynaecologic malignancy in high-income countries, with a rising incidence largely driven by reproductive factors, obesity, and prolonged exposure to unopposed oestrogens. Although most cases are sporadic, approximately 2-5% are associated with hereditary cancer syndromes, of which Lynch syndrome represents the most important contributor. Lynch syndrome results from germline mutations in DNA mismatch repair (MMR) genes and is associated with a substantially increased lifetime risk of endometrial cancer, reaching up to 71% in carriers of MutS homologue 6 (MS

BRCA Testing in Asian Patients with Ovarian Cancer: Standard Clinical Practice or Mutation Prediction Model?

BACKGROUND: Germline BRCA1/2 testing is recommended for all patients with ovarian cancer, as identifying pathogenic variants (PV) informs treatment and enables family cascade testing. However, in resource-limited settings, high testing costs often limit feasibility. An alternative approach is to use predictive models to prioritize patients at risk, optimizing resource allocation. Existing models are largely Western-derived; in Asian populations, models exist for breast cancer-not ovarian cancer. METHODS: Using data from a multicenter study of 1,126 Asian patients with ovarian cancer (includin

Characterization of ESR1 alterations in patients with breast and gynecologic cancers.

BACKGROUND: ESR1 alterations present a common mechanism of resistance to endocrine therapy (ET) in hormonally driven tumors. The clinical significance of these alterations continues to evolve with newly approved targeted therapies and a range of ongoing investigational trials. METHODS: A retrospective study of 2574 breast cancer (BC) and 1110 gynecologic cancer samples that underwent whole exome and whole transcriptome profiling was conducted to assess the distribution of ESR1 and associated co-alterations in local (primary breast or regional lymph node) versus metastatic BC samples and in th

Tyrer-Cuzick Lifetime Risk Is Not Associated With Non-BRCA1/2 Pathogenic Variants for Breast Carcinoma.

BACKGROUND: The Tyrer-Cuzick (TC) or IBIS risk calculator is a widely used tool to estimate the probability of developing breast cancer. The latest version incorporates various factors to assess the risk of breast cancer, including family history, personal history, breast density, and past medical history. The TC is commonly used to guide patients toward further diagnostic imaging, genetic testing, chemoprevention, or risk-reducing surgery. However, it is unclear whether the TC is associated with non-BRCA1/2 pathogenic variants (PVs) in breast cancer susceptibility genes. METHODS: A populatio
