COVID-19 pleiotropy with brain imaging-derived phenotypes highlights novel insights into the biology shared between immune function and brain variation.

This study investigated the pleiotropy and the underlying biology linking COVID-19 outcomes to brain variation, leveraging genome-wide data from the COVID-19 Host Genetics Initiative and multimodal brain-wide magnetic resonance imaging from the UK Biobank. In our analyses, COVID-19 outcomes showed evidence of polygenic overlap with respect to 11 imaging-derived phenotypes (IDPs) related to brain structure (e.g. parahippocampal and entorhinal cortical thickness) and connectivity (e.g. Broca's area and dorsolateral prefrontal cortex). Among these, the genetic liability to COVID-19 hospitalizatio

Multi-omics integration reveals vaccine-specific molecular signatures and temporal dynamics of COVID-19 immune responses.

Vaccination elicits systemic immune responses that extend beyond antibody and lymphocyte activation, engaging coordinated metabolic and lipid pathways that modulate immune function. Nevertheless, how systemic molecular responses differ across COVID-19 vaccine platforms, including closely related mRNA platforms, remains poorly understood. Here, we performed integrated plasma proteomic, lipidomic, and metabolomic profiling of 53 SARS-CoV-2-naïve adults vaccinated with Pfizer-BioNTech, Moderna, or AstraZeneca COVID-19 vaccines. Samples were analysed across multiple timepoints using differential a

A multimodal in situ framework for spatial characterization applied on SARS-CoV-2 live-attenuated vaccines in the nasal mucosa.

Respiratory viruses frequently initiate infection in the nasal mucosa, where virus-host interactions shape disease progression and transmission. Yet this site remains undercharacterized due to anatomical and technical challenges. Spatially resolved technologies are limited by cost and complexity, leaving most studies reliant on bulk or descriptive approaches lacking spatial context. We present a framework for spatially resolved investigation of infections in the nasal cavity. Using wild-type SARS-CoV-2 and two live-attenuated vaccines (OTS-206, OTS-228) as model viruses in Syrian hamsters, we

Prevalence of respiratory viruses among pilgrims in Saudi Arabia during the Hajj 2026: a cross-sectional study.

BACKGROUND: Hajj is one of the world's largest annual mass gatherings and provides favorable conditions for respiratory virus transmission because of intense crowding and extensive international travel. Contemporary post-pandemic molecular surveillance data remain limited. This study investigated the prevalence, determinants, and characteristics of respiratory virus detections among pilgrims during the 2026 Hajj season. METHODS: A cross-sectional study was conducted among adult domestic and international pilgrims over an extended peri-Hajj period (before, during and after Hajj) in the year 20

Post-translational modifications in the insoluble fraction of platelet-poor plasma distinguish pre-COVID-19 Postural Orthostatic Tachycardia Syndrome, Long COVID, and Long COVID-POTS: a preliminary study.

BACKGROUND: Pre-COVID-19 Postural Orthostatic Tachycardia Syndrome (PC-POTS), Long COVID with initial orthostatic hypotension (LC-IOH), and Long COVID-POTS (LC-POTS) are primarily chronic post-viral conditions with debilitating symptoms, despite routine laboratory findings often being normal. We previously identified amyloidogenic fibrinaloid microclot complexes (FMCs) in the insoluble fraction of platelet-poor plasma (PPP-IF) in Long COVID, but it remains unclear whether similar abnormalities occur in PC-POTS and LC-POTS, or whether post-translational modifications (PTMs) contribute to diseas

Immunosuppression is associated with loss of airway microbiome resilience in severe COVID-19.

BACKGROUND: The role of the lower airway microbiome in COVID-19-associated mortality remains poorly understood despite evidence implicating secondary pathogens and non-airway commensals. The influence of prior immunosuppression on the lower airway microbiome and its contribution to mortality in mechanically ventilated patients has been largely unexplored. Given that host-microbiome interactions are dynamic, we assessed longitudinal patterns in the respiratory microbiome associated with mortality and immunosuppression status. RESULTS: We conducted longitudinal forecasting, differential abundan

Jet-injected combinational adjuvants potentiate naked mRNA vaccines for protection against respiratory viral infections in mice.

The use of lipid nanoparticles (LNPs) to enhance delivery and immunogenicity of mRNA vaccines raises safety concerns due to strong reactogenicity. Developing naked mRNA vaccines, which can be delivered through liquid jet-injection to the skin, may offer a safer alternative, but their immunogenicity has been limited. To address this challenge, we screened clinically relevant adjuvants to enhance the efficacy of N1-methyl-pseudouridine-modified naked mRNA jet-injection vaccines in mouse models. Although many adjuvants showed little or inhibitory effects on inducing humoral or cellular immunity-d

Respiratory Viral Infections in Febrile Children With Lymphoreticular Malignancies: A Single-Centre Descriptive Study.

Children with lymphoreticular malignancies are highly vulnerable to respiratory viral infections (RVIs) because of disease- and chemotherapy-related immunosuppression, yet data on their epidemiology, clinical spectrum, and outcomes from resource-limited settings remain scarce. This study aimed to characterize the seasonal distribution, clinical presentation, and outcomes of RVIs other than SARS-CoV-2 in febrile children with lymphoreticular malignancies at a tertiary care centre in India. In this prospective, descriptive, cross-sectional study (May 2023-November 2024), 33 children under 12 yea

D-Dimer in Viral Respiratory Infections: Established Evidence in COVID-19 and Emerging Evidence in Other Respiratory Viruses.

Viral respiratory infections remain a major cause of global morbidity and mortality, with severe disease increasingly recognized to involve profound interactions between inflammation, endothelial dysfunction, coagulation activation, and immunothrombosis. D-dimer, a fibrin degradation product and established biomarker of fibrin turnover, has gained considerable attention because of its strong association with adverse outcomes in coronavirus disease 2019 (COVID-19). This narrative review compares the evidence for D-dimer in COVID-19 with that available for other respiratory viruses and deliberat

Herpes zoster epidemiology during and after the COVID-19 pandemic: A 5-year retrospective study from primary care in Poland.

BACKGROUND: Herpes zoster (HZ) is caused by reactivation of latent varicella-zoster virus and affects 25-30% of individuals, with the highest incidence in adults aged ≥50 years and a lifetime risk exceeding 50% after the age of 85. Its most common complication is postherpetic neuralgia (PHN), i.e., persistent neuropathic pain. Reports of increased HZ incidence during or after SARS-CoV-2 infection suggest potential epidemiological associations requiring further investigation. OBJECTIVES: Our goals were to evaluate the incidence of HZ and its complications during the pandemic and the post-pande

Hematological indices as predictors of in-hospital adverse outcomes in patients with COVID-19.

The SARS-CoV-2 pandemic required medical teams to make rational decisions with limited resources, especially in Brazil. Various diagnostic tools, clinical guidelines, and existing indices were adopted to predict the risk of death and other adverse outcomes. The objective of this study was to validate three hematological indices [neutrophil-lymphocyte ratio (NLR), platelet-lymphocyte ratio (PLR), and lymphocyte-monocyte ratio (LMR)] for predicting adverse hospital outcomes in patients with COVID-19 in an observational cohort at a hospital in northeastern Brazil between April 2020 and October 20

The impact of COVID-19 on all-cause mortality and life expectancy: findings from a population-based cohort in uMkhanyakude district, northern KwaZulu-natal.

BACKGROUND: Despite the comprehensive death reporting system in South Africa, significant gaps persist in our understanding of the pandemic's impact in rural areas. METHODS: We computed monthly death counts stratified by demographic factors from a population-based demographic surveillance cohort in rural KwaZulu-Natal. We fit seasonally adjusted negative binomial regression models to estimate excess mortality during the COVID-19 pandemic period (5 March 2020 to 31 December 2021) compared to the pre-pandemic period (1 January 2015 to 4 March 2020). We then assessed the relative risk of death b

Spatial transcriptomics unveils the in situ cellular and molecular hallmarks of the lung in fatal COVID-19.

Severe Coronavirus disease 2019 (COVID-19) can induce progressive diffuse alveolar damage (DAD) leading to fatal outcomes. Here, we leveraged data from Visium spatial transcriptomics for COVID-19 and control lung samples to unravel insights into cellular and molecular events driving lethal COVID-19. We report a progressive loss of endothelial cell types, pneumocytes type I and natural killer cells coupled with a continuous increase of myeloid and stromal cells, mostly peribronchial fibroblasts, across acute and proliferative DAD. Spatial organization analysis also identified specific compartme

The Water-Soluble [60]Fullerene Derivatives as Binders of SARS-CoV‑2 Proteins.

Water-soluble fullerene derivatives represent emerging molecular nanomaterials for antiviral drug discovery, yet their target selectivity against SARS-CoV-2 proteins remains insufficiently defined. Here, four functionalized [60]-fullerenes, C60(OH)24, C60ser, C60ALA, and C60Trp, were evaluated as binders of key SARS-CoV-2 targets involved in viral entry and replication: spike glycoprotein, papain-like protease, 3CL main protease, and RNA-dependent RNA polymerase. Microscale thermophoresis revealed distinct target-dependent binding profiles governed by fullerene functionalization. The C60Trp fu
