Clinical Nursing in Times of War and a Pandemic.

Nurses who served in the Vietnam War and those who cared for patients during the 2020 COVID-19 pandemic surge may be separated by decades, but they share the experience of caring for large numbers of critically injured, ill, or dying patients while risking their own lives. This article reviews the findings of two studies, one conducted in 1986 based on interviews with Vietnam War nurses, and one conducted in 2020 based on interviews with nurses during COVID-19, and explores the idea that professional nurses from different generations have similar thoughts when their lives are in danger and pat

Prevalence of alpha-synuclein seeding activity in the Olfactory Mucosa following SARS-CoV-2 infection: results from a pilot study.

Olfactory dysfunction (OD) is a hallmark of SARS-CoV-2-infection where it is believed to result from neuroinflammation induced by viral invasion via the olfactory mucosa (OM). Hyposmia also affects the majority of patients with Parkinson's disease (PD) where it is caused by α-synuclein (aSyn) pathology in the olfactory system, and aSyn-seeding-activity can be detected in the OM using aSyn-seeding-amplification-assays (aSyn-SAA). The present pilot study aimed to investigate whether SARS-CoV-2-infection induces local aSyn-misfolding and -aggregation in OM samples using aSyn-SAA. We conducted an

Implementing a First Nation data-sovereign and agile modelling platform for pandemic management: lessons from Manitoba.

This paper describes lessons learned from a partnership between the First Nations Health and Social Secretariat of Manitoba (FNHSSM) and researchers at the University of Manitoba to develop a community-led COVID-19 modelling platform. The envisioned tool would allow First Nations leaders to input local data and generate outputs to support decision-making using a stochastic, age-stratified agent-based model of COVID-19 transmission. Although the platform was not fully operationalized during the project, several important lessons emerged. First, the COVID-19 pandemic placed significant demands o

Serologic Testing for Middle East Respiratory Syndrome Coronavirus and SARS-CoV-2 Cross-Reactive Immune Responses, the Netherlands.

Cross-reactive SARS-CoV-2-specific antibodies could compromise the specificity of MERS-CoV serology, potentially leading to misclassification of infection history. SARS-CoV-2 exposure induced MERS-CoV antibodies primarily targeting the conserved S2 domain, with occasional neutralizing capacity. Combining MERS-CoV S1 binding with neutralization, however, limited the influence of SARS-CoV-2-induced cross-reactivity on MERS-CoV serology.

Post-COVID-19 Expansion of Streptococcus pyogenes Lineages Causing Toxic Shock Syndrome, Japan, 2023-2024.

We analyzed 919 Streptococcus pyogenes isolates from laboratory-confirmed streptococcal toxic shock syndrome cases in Japan during 2023-2024. Post-COVID-19 resurgence was driven by expansion of M1UK (49.8%) and emerging M113SNPs (37.4%) lineages, replacing ancestral M1global. M113SNPs were associated with younger patient age and higher death rates in unadjusted analyses.

Data-driven forecasting of Flu, RSV, and COVID-19 related outcomes in the United States and Canada via Hankel dynamic mode decomposition.

The (large) season-to-season variability and limited dynamical history make the forecasting of infectious diseases a challenging problem. Here, we examine the extent to which advances in data-driven dynamical modeling can provide accurate predictions by benchmarking the performance of one such method, Hankel dynamic mode decomposition (DMD), on the 2024-2025 influenza, respiratory syncytial virus (RSV), and COVID-19 seasons in the United States and Canada. Using Hankel-DMD, we generated weekly forecasts that were submitted to the Center for Disease Control and Prevention's (CDC) FluSight Forec

The detection paradox: How COVID-19 surveillance patterns shifted from lower-income to more affluent neighborhoods in Miami-Dade County.

Passive disease surveillance systems may systematically undercount burden in low-resource minority neighborhoods, yet fine-scale evidence of this phenomenon remains limited. We examined spatiotemporal patterns of reported COVID-19 cases across Miami-Dade County's diverse neighborhoods and investigated which socioeconomic and demographic characteristics predicted areas of elevated and suppressed case rates. In an ecological, cross-sectional analysis of 491,769 geocoded cases across 1,831 census block groups from March 2020 to May 2021, we applied negative binomial regression with spatial eigenv

Permissive versus conventional weaning from venovenous ECMO in patients with ARDS: a retrospective cohort study.

BACKGROUND: Criteria for weaning from venovenous extracorporeal membrane oxygenation (V-V ECMO) in acute respiratory distress syndrome (ARDS) remain poorly standardized. Some patients are decannulated despite persistent abnormalities in respiratory mechanics once oxygenation and carbon dioxide clearance are considered adequate. We aimed to compare outcomes associated with permissive versus conventional V-V ECMO weaning. METHODS: We conducted a retrospective single-center cohort study including adult patients with refractory ARDS treated with V-V ECMO between January 2015 and December 2022 who

Preliminary stability studies of a β-SARS-CoV-2 virus-like particle vaccine.

We aimed to study the stability of a Beta (β)-SARS-CoV-2 virus-like particle (VLP) vaccine in a series of preliminary experiments using selective stabilizing excipients. β-SARS-CoV-2 VLPs were produced and purified using established methodologies. The thermostability of VLPs was tested at 4°C and -30°C in the presence or absence of stabilizers polysorbate 80, sorbitol, or L-histidine in the presence of a physiological NaCl concentration of 137 mM. The integrity of VLPs was assessed using ELISA, Western immunoblot, and dynamic light scatter (DLS). β-SARS-CoV-2 VLPs were stable at 4°C for 14 day

Cell DiffErential Expression by Pooling (CellDEEP) highlights issues in differential gene expression in scRNA-seq.

Accurate identification of differentially expressed genes (DEGs) in single-cell RNA sequencing data remains challenging. Single-cell-specific statistical models often report large numbers of candidate genes but can exhibit inflated false positive rates, whereas pseudobulk approaches improve false discovery control at the cost of reduced sensitivity. To overcome the noise and bias that other tools have, and allow the user to have more control of the DEG process, we present CellDEEP, which uses a cell aggregation (metacell) approach. This tool provides a framework for flexible selection of pooli

Viral Infection-Induced Epilepsy and the Gut Microbiome Crosstalk.

Viral infections of the central nervous system (CNS) pose a significant risk for seizures, and in some cases, long-term epileptogenesis. Seizure development following viral infection involves diverse CNS mechanisms, with emerging evidence that peripheral factors, including gut microbiome composition and immune-inflammatory responses, also contribute. The gut microbiome is a multifaceted ecosystem with important roles in CNS health, and advances in sequencing have enabled investigation of host and microbial genomic changes associated with seizure development. While both animal and human studies

Prolonged Active SARS-CoV-2 Infection in a Patient With Chronic Lymphocytic Leukemia on Venetoclax.

We report a case of prolonged active SARS-CoV-2 infection in a patient with chronic lymphocytic leukemia (CLL) receiving venetoclax. The patient demonstrated persistent symptoms over a span of 105 days and repeatedly positive SARS-CoV-2 polymerase chain reaction (PCR) testing. The case highlights the diagnostic and therapeutic challenges in immunocompromised hosts, including limited guidance and drug availability. Management required multiple antiviral regimens and consideration of alternative therapies. This case underscores the need for further research that can inform more robust guidelines

Intranasal gold nanoparticle-antigen induces mucosal and systemic immunity against β-coronaviruses.

The continued emergence of diverse beta-coronaviruses (β-CoVs) from animal reservoirs underscores the urgent need for broad-spectrum mucosal vaccines, yet their development is hindered by physiological barriers, rapid antigen clearance, and suboptimal multivalent display strategies. To address this, we intranasally immunized BALB/c mice (n = 3 or 4 per group) with 5 or 15 nm gold nanoparticles (AuNPs) displaying the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) receptor-binding domain (RBD, 5 μg per mouse) and adjuvanted with the STING agonist cGAMP (20 μg per mouse). Evaluating a multi-vaccine respiratory immunization strategy involving SARS-CoV-2, pneumococcal, and influenza vaccines in adults: A randomized, open-label, multicenter, controlled, non-inferiority Phase IV trial in China.

Co-administration of vaccines targeting multiple respiratory pathogens may enhance coverage and streamline adult immunization programs. We conducted a randomized, multicentre, open-label Phase IV trial registered at Chinese Clinical Trial Registry (ChiCTR2100046227) in China to evaluate the immunogenicity and safety of co-administering an inactivated SARS-CoV-2 vaccine (WIBP-CorV) with the 23-valent pneumococcal polysaccharide vaccine (PPV23) and a quadrivalent inactivated influenza vaccine (IIV4). A total of 1,404 adults aged ≥ 18 year, including equal numbers of adults aged 18-59 year and ≥6
