Protein translation regulation is critical for cellular responses and development, yet how elongation stage disruptions shape these processes remains incompletely understood. Here, we identify a single amino acid substitution (P55Q) in the ribosomal protein RPL-36A of Caenorhabditis elegans that confers complete resistance to the elongation inhibitor cycloheximide (CHX). Heterozygous animals carrying both wild-type RPL-36A and RPL-36A(P55Q) develop normally but show intermediate CHX resistance, indicating a partial dominant effect. Leveraging RPL-36A(P55Q) as a single-copy positive selection m

Necrotrophic Alternaria alternata induces EXECUTER 1(EX1)/2-dependent singlet oxygen (1O2) bursts, leading to plant cell death, with jasmonic acid (JA) acting as a key signal transducer downstream of EX1/2-mediated signaling. Salicylic acid (SA), a crucial defense hormone, is known to respond to pathogen invasion and activate defense gene expression. Previous studies emphasized the importance of SA in A. alternata-induced necrosis in the light of the increased susceptibility of SA-deficient transgenic Arabidopsis NahG to A. alternata. In this study, we investigated the role of SA in A. alterna

Fresh Japanese plums are highly perishable and susceptible to postharvest diseases caused by fungal pathogens. Current management strategies rely on synthetic fungicides, raising concerns about resistance and sustainability. Immature-thinned apples (ITA), an underutilized agricultural by-product, are rich in polyphenols with potential antifungal properties. This study evaluated the potential of ITA ultrasound-assisted extracts of the cultivars 'Gala', 'Golden Delicious', and 'Opal' for controlling plum postharvest diseases. Extracts were chemically characterized for total polyphenol content an

The MYB transcription factor family is one of the largest gene families in plants and plays critical roles in how plants respond to biotic and abiotic stresses. However, the functions of MYB members in high-temperature seedling plant (HTSP) resistance in wheat to Puccinia striiformis f. sp. tritici (Pst) are not well understood. Previously, TaMYB73, an MYB transcription factor, was identified through an RNA-sequencing study of the wheat cultivar Xiaoyan 6 under infection by Pst. In this work, we characterized the molecular function of TaMYB73. The expression TaMYB73 was upregulated following P

Wheat sharp eyespot, mainly caused by Rhizoctonia cerealis, is a destructive soilborne disease that significantly threatens global wheat production, necessitating the development of sustainable control strategies. In this study, we isolated and identified an endophytic fungus, Penicillium oxalicum G11, from the roots of healthy wheat plants and comprehensively evaluated its antagonistic mechanisms and biocontrol potential against R. cerealis. In vitro assays demonstrated that G11 effectively inhibited R. cerealis growth through multiple mechanisms, including competition, antibiosis, and mycopa

Pomegranate and carob extracts have been characterized as potential bioprotectors against Verticillium wilt of olive caused by Verticillium dahliae, suggesting they may act as host resistance inducers. This study aimed to elucidate whether these extracts activate metabolic pathways involved in olive defense against V. dahliae infection through marker gene expression analysis. The relative expression of seven resistance-related marker genes was evaluated by RT-qPCR in noninoculated and treated olive plants. Subsequently, the effect of the extracts as inducers of resistance against Verticillium

The synergistic combination of two antimicrobial drugs is a promising therapeutic modality for many infectious diseases. However, systemic fungal infections still have a high mortality rate because of distinct in vivo distributions of the two drugs. Here we address this challenge by designing an antifungal polymer that forms micelles suitable for delivering a second antifungal agent to achieve temporal and spatial consistency of delivery. We show that the polymer, which mimics host defense peptides, exerts a synergistic effect with the antifungal amphotericin B (AmB). The AmB-encapsulated mice

Heterochromatin, marked by histone H3 lysine 9 methylation, can be epigenetically inherited through cell division1-3, maintaining gene repression that preserves cell identity and enables adaptation to environmental challenges2-6. Studies on Schizosaccharomyces pombe have shown that heterochromatin propagation depends on the read-write mechanism, wherein a sufficient density of H3K9me3-modified nucleosomes, stabilized by histone deacetylases, concentrates Clr4SUV39H on chromatin to promote further deposition of H3K9 methylation7-9. Whether other mechanisms control heterochromatin propagation by

Fungal infections are mainly caused by opportunistic species of the Candida genus. These yeasts can form polymicrobial biofilms with bacteria on living or inert surfaces, including medical devices. The synergistic or antagonistic interactions within these biofilms promote virulence and resistance to antimicrobials as well as to the host's immune defenses. In response to this resistance, considerable efforts are being made to limit treatment failures. The use of antifungal-antibiotic-natural compound combinations, as well as the modification of medical device surfaces, offer promising solutions

This study compares the ability of the cytokinin (CK) trans-zeatin (tZ) and the CK sugar conjugate 6-(3-methoxybenzylamino)purine-9-arabinoside (BAPA) to induce resistance against the bacterial pathogen Pseudomonas syringae in Arabidopsis thaliana. Treatment with either tZ or BAPA significantly reduced bacterial growth after a later infection. This chemically induced resistance (IR) required the CK receptor AHK3, highlighting its critical role in mediating resistance by tZ and BAPA. This is remarkable as these compounds show either high or no affinity for this CK receptor, respectively. Surpri

The alarming rise of drug resistance has created an urgent need for novel antifungal agents. On the other hand, growing environmental concerns necessitate the development of sustainable alternatives to conventional synthetic methods for active pharmaceutical ingredients (APIs). β-Aminosulfones represent a potent yet underexplored functionality in biologically active compounds. In this work, a series of β-aminosulfone derivatives, designed as potential antifungal agents, were synthesized via a one-step, reagent-free, catalyst-free double aza-Michael addition of 2-aminobenzothiazoles, biogenic a

The emergence of antimicrobial resistance necessitates the exploration of novel bioactive compounds. We conducted this study to explore the biological capabilities of R6, an endophytic actinomycete isolated from Ocimum tenuiflorum. The strain exhibited significant antibacterial activity, particularly against ESBL-producing Escherichia coli (20 mm) and Klebsiella pneumoniae (25 mm). Moderate antifungal activity was also observed against Aspergillus fumigatus (9–10 mm). Cytotoxicity screening using the MTT assay revealed 75% cell mortality at 100 mg/mL against Baby Hamster Kidney fibroblast (BHK

Infections caused by Candida albicans pose a serious challenge in healthcare, particularly due to increasing resistance to conventional antifungal agents. Photodynamic therapy (PDT), which combines light, photosensitizers, and oxygen, offers a promising alternative solution. This study aimed to evaluate the antifungal effectiveness of gold nanoparticles (AuNPs) synthesized via laser ablation, as well as the effect of photodynamic activation using a blue diode laser (403 nm) on the growth of C. albicans. Samples were divided into four groups: (1) control, (2) Candida albicans treated with three

Hosts deploy mechanosensory systems to intercept herbivory threats, yet how plants decode insect-derived mechanical force remains unclear. Here, we demonstrate that PIEZO-type mechanosensitive ion channel component 1 (PIEZO1) serves as a mechanoimmune hub converting whitefly probing into calcium-dependent defense activation. Mechanical force during feeding activates PIEZO1-dependent Ca2+ influx, triggering CBP60g phosphorylation that amplifies both PIEZO1 transcription and salicylic acid (SA) biosynthesis, forming a self-amplifying defense loop. Furthermore, PIEZO1 positively modulates the cor

Ocimum gratissimum L. (African basil) is widely used in ethnomedicine to treat infectious and inflammatory conditions, yet its antifungal mechanisms remain unclear. In this study, we evaluated the essential oil of O. gratissimum L. (OGEO) against Candida albicans biofilms, a clinically relevant driver of antifungal resistance in device-associated infections. Gas chromatography-mass spectrometry identified eugenol as the predominant constituent (59.5%), supported by terpenoid and sesquiterpene derivatives. Functional assays demonstrated potent antibiofilm activity, as exposure to 1% and 2% OGEO
