The changing face of dermatophytosis: from common skin disease to emerging resistant epidemic.

Dermatophytosis is a commonly encountered superficial fungal infection with an affinity for keratin-rich structures, such as skin, hair and nails. The recently emerging recalcitrant dermatophytosis is a recurrent, relapsing, chronic or persistent disease, which has a significant impact on the social, emotional and financial aspects of a patient's life. Trichophyton indotineae, a newly identified species, has become resistant to conventional therapies, unlike its predecessor. This review highlights the shift of this commonly encountered skin disease from an easily manageable clinical entity to

The lichen secondary metabolite lichesterinic acid exhibits antibiofilm activity against fungal pathogens.

Lichens are well known for producing unique secondary metabolites, some of which have been shown to exhibit medically relevant bioactivities, including antimicrobial effects. With the increasing prevalence of fungal infections and the growing resistance to commonly used antimycotics, there is an urgent need for new antifungal agents, especially for aged and/or immunocompromised individuals. In this study, we screened a collection of lichen-derived metabolites for antifungal properties in two medically relevant fungal pathogens, Candida albicans and Nakaseomyces glabratus. Several compounds exh

Arabica coffee SBP transcription factor family members respond to brown leaf spot stress by regulating their expression.

Coffee brown spot is a prevalent foliar fungal disease that seriously threatens the survival and yield of Coffea arabica L. To explore the molecular basis of disease resistance, we conducted a genome-wide identification and analysis of the SBP gene family in C. arabica L. A total of 22 CaSBP genes were identified and classified into three phylogenetic subfamilies, distributed across 12 chromosomes and including nine segmentally duplicated gene pairs. Cis-element analysis revealed a high proportion (55.37%) of hormone-related regulatory elements in CaSBP promoters. Integrated transcriptomic and

Epimutations driven by RNAi or heterochromatin evoke transient antimicrobial drug resistance in pathogenic Mucor fungi.

Antimicrobial resistance (AMR) is a global health threat emerging through microbe adaptation, driven by genetic variation, genome plasticity or epigenetic processes. In this study, we investigated how the Mucor circinelloides species complex adapts to the antifungal natural product FK506, which binds to FKBP12 and inhibits calcineurin-dependent hyphal growth. In Mucor bainieri, most FK506-resistant isolates (90%) were found to be unstable and transient, readily reverting to being drug sensitive when passaged without drug, and with no associated DNA mutations. In half of the isolates (50%), FK5

Boosting phenazine-1-carboxylic acid (PCA) yield in Pseudomonas aeruginosa and assessing antifungal performance of PCA-loaded mesoporous carriers.

Phenazine-1-carboxylic acid (PCA) is a potent antifungal metabolite from Pseudomonas aeruginosa, but its application is limited by low production yields and stability issues. This study aimed to enhance PCA production and evaluate its efficacy using a nano-delivery system. We employed a two-stage statistical optimization strategy. First, a Plackett-Burman (PB) design identified pH, temperature, and glycerol concentration as critical factors. Subsequently, Response Surface Methodology (RSM) based on a Central Composite Design (CCD) was used to fine-tune these parameters. The optimal conditions

Novel aminoguanidine derivatives containing alkynyl moiety: synthesis and evaluation of antibacterial and antifungal activities.

The urgent global health threat of antimicrobial resistance demands innovative therapeutic strategies. Herein, we report the design, synthesis, and biological evaluation (antibacterial and antifungal activities) of two series of alkynyl-linked aminoguanidine derivatives. A critical structure-activity relationship (SAR) was revealed: the exposure of the aminoguanidine group is paramount for activity. In vitro antibacterial and antifungal activities revealed that the imidazol-2-hydrazine series exhibited inhibition against both Gram-positive and Gram-negative bacteria, with minimum inhibitory co

Inhibition mechanism of the fungal β-1,3-glucan synthases by triterpenoid antifungal drugs.

β-1,3-glucan synthase is the molecular target for triterpenoid and echinocandin antifungal drugs in clinical. It catalyzes the formation of β-1,3-glucan, which is the primary component of the fungal cell wall. However, the inhibition mechanism of β-1,3-glucan synthase by triterpenoid drugs remains unclear. In this study, we report cryo-electron microscopy (cryo-EM) structures of Saccharomyces cerevisiae β-1,3-glucan synthase Fks1 and Fks2 in the apo state, the triterpenoid drug enfumafungin-bound state, and an open state. Structural analysis along with mutagenesis reveals the enfumafungin bind

Mutations in ERG11, TAC1B, and CDR1 reduce fluconazole accumulation in drug-resistant Candidozyma auris isolates.

Fluconazole (FLC)-resistant Candidozyma auris isolates have reduced drug accumulation compared to azole-susceptible isolates. Of 119 C. auris isolates, 83 out of 87 resistant isolates (~95%) had extremely low fluconazole uptake, whereas 30 out of 32 susceptible isolates (~93%) had high fluconazole uptake. In search of a genetic explanation for this phenomenon, we compared metadata for TAC1B and CDR1 single-nucleotide polymorphisms (SNPs) and found overlap with many but not all isolates that are FLC resistant. We found that CDR1 mutations are common in resistant isolates from Clade 1, and TAC1B

iAFP-fLRM: Accurate identification of antifungal peptides via hybrid deep learning architecture and multi-modal feature fusion.

Fungal infections (mycoses) represent a significant and increasing global health concern, particularly in immunocompromised populations. The emergence of antifungal drug resistance and the limited efficacy of conventional treatments necessitate the development of novel therapeutic strategies. Antifungal peptides (AFPs), due to their broad-spectrum activity, low toxicity, and reduced likelihood of resistance development, have garnered considerable attention as potential alternatives. However, the experimental identification of AFPs remains costly, labor-intensive, and time-consuming. To address

Integrative Structural Characterization of Candida glabrata Phosphoglycerate Kinase by Small-Angle X‑ray Scattering and AlphaFold: Implications for Therapeutic Targeting in Candidiasis.

Candida glabrata is the second leading cause of mortality in immunocompromised patients hospitalized for invasive candidiasis (IC). Several drugs have been available to treat this disease for decades, such as polyenes, azoles, echinocandins, flucytosine, and, in critical cases, amphotericin B. However, these antifungals' constant and routine use have led to the development of resistance mechanisms, making the design and development of new drugs indispensable. The first step for the design and subsequent synthesis of a new chemical molecule as a potential antifungal is the identification of new
