Cysteine derivatives coordinate photosynthetic performance and osmotic adjustment to enhance drought tolerance in vegetable soybean.

BACKGROUND: Drought stress severely constrains photosynthetic efficiency in vegetable soybean. However, the role of cysteine-based biostimulants in coordinating photosynthetic regulation and osmotic adjustment remains poorly understood. This study evaluated how N-acetylcysteine (NAC) and N-acetylcysteine trisulfide (NAC-TS) modulate photosynthetic function, photoprotection, and osmotic adjustment to enhance growth and reproductive performance under drought in two vegetable soybean (Glycine max L. Merr.) cultivars with different yielding capabilities and drought sensitivities (UVE17 and AGS429)

Tumor-intrinsic execution of interferon-γ-associated immunity in bladder cancer: checkpoints in signaling, recruitment, and recognition.

Interferon-γ (IFN-γ) is a central mediator of antitumor immunity in bladder cancer, but an IFN-γ-rich tumor microenvironment does not necessarily translate into tumor control. Tissue-level inflammatory signatures capture overall immune activity and cannot determine whether malignant urothelial cells remain responsive to immune-derived IFN-γ or preserve the downstream functions required for immune elimination. This Mini Review examines tumor-intrinsic IFN-γ-associated immunity at three functional checkpoints: proximal IFN-γ sensing and signal transduction, chemokine-mediated effector-cell recru

Berberine as a Multitarget Therapeutic Agent: Insights from Molecular Docking Studies.

Natural compounds are crucial in pharmacological research for modulating numerous biological processes. This is particularly helpful in the case of complex diseases and in overcoming challenges (e.g., drug resistance and adverse effects). Berberine, an isoquinoline alkaloid extracted from plants such as Berberis, Coptis, and Hydrastis species, exhibits a wide range of activities, including antidiabetic, anticancer, anti-inflammatory, antimicrobial, and antiviral. However, due to its low oral bioavailability (poor absorption, high first-pass metabolism, along with rapid elimination), further me

Discovery of dihydrothiazolo ring-fused 2-pyridones with extended carboxylic acid as anti-influenza A agents.

Influenza A virus (IAV) infections are a global health concern, aggravated by emerging drug-resistant strains. We report the identification of a series of dihydrothiazolo-ring-fused 2-pyridones bearing an extended carboxylic acid motif as promising antiviral scaffolds. Initial phenotypic screening yielded compound 9 (IC50 = 28 μM), which guided structure-activity optimization at C-6, C-7, and C-8. This yielded compound 35d, which showed moderate potency (IC50 = 21 μM), but with minimal cytotoxicity and a moderate Selectivity Index (∼19). A time-of-addition study indicates that 35d disrupts an

Structural characterization of HCoV-229E and H163A main protease (Mpro) interactions with the inhibitor narlaprevir.

Narlaprevir, originally developed as a clinical-stage inhibitor of the hepatitis C virus (HCV) NS3/4A protease, has recently demonstrated potent inhibitory activity against the SARS-CoV-2 main protease (Mpro), with its co-crystal structure already resolved. To elucidate the mechanistic basis of its cross-genus inhibition and drug resistance profiles, we determined the crystal structures of narlaprevir in complex with HCoV-229E Mpro and the SARS-CoV-2 Mpro(H163A) mutant at resolutions of 2.01 Å and 2.14 Å, respectively. By integrating these crystallographic data with molecular dynamics (MD) sim

Co-infection models of Staphylococcus aureus and respiratory viruses reveal synergistic interactions counteracted by a bithiazole derivative.

Viral-bacterial co-infections represent a major contributor to global morbidity and mortality. In influenza virus infections, secondary bacterial pneumonia occurs in approximately 20-30% of cases and is associated with an increased risk of shock, mechanical ventilation, and even mortality. Bacterial superinfections are frequent causes of mortality in influenza severe infections and have been a major cause of morbidity and mortality also in COVID-19 patients. Staphylococcus aureus has been identified as one of the most prevalent bacterial pathogens among hospitalized and non-hospitalized patien

Evaluating Fc-mediated antibody functions against SARS-CoV-2 in PLHIV: reduced FcγRIIIa-dependent activity and ADCC despite preserved neutralising antibodies.

BACKGROUND: Antibody-dependent cellular cytotoxicity (ADCC) is a critical Fc-mediated immune mechanism contributing to antiviral defence. However, its functionality in people living with HIV (PLHIV) following SARS-CoV-2 exposure remains poorly understood. OBJECTIVE: This study evaluates the anti-Spike ADCC antibody levels, functional ADCC activity, and neutralising antibody responses in HIV-negative and HIV-positive individuals. METHODOLOGY: The study included 30 samples per group. Anti-Spike ADCC-binding antibodies were measured using FcγRIIIa-binding assays, while functional ADCC activity

Association of nucleos(t)ide analogue treatment history with mortality in HBV-DNA-positive HBV-ACLF: a multicenter cohort study.

BACKGROUND AND AIMS: Hepatitis B virus-related acute-on-chronic liver failure (HBV-ACLF) is associated with high short- and long-term mortality. Prior nucleos(t)ide analogue (NA) treatment history, particularly treatment withdrawal and virological resistance, may contribute to ACLF development and influence prognosis after onset. However, the clinical impact of different NA treatment histories in patients with HBV-ACLF remains insufficiently characterized. This study evaluated the associations of baseline HBV-DNA status and prior NA treatment history with mortality in patients with HBV-ACLF.

Emergent HIV-1 Drug Resistance in Randomized Trials of Contemporary Antiretroviral Regimens.

The occurrence of virological failure in a subset of individuals is an inevitable aspect of antiretroviral treatment, and historically has been primarily influenced by suboptimal adherence to oral therapies. The risk of selecting 1- or 2-class human immunodeficiency virus (HIV) drug resistance is influenced by the composition of the regimen, differing significantly depending on the intrinsic barrier to resistance of the regimen. HIV resistance emergence during treatment can be viewed as a regimen-related adverse effect that warrants equal consideration in clinical trials alongside virological

Real-world Prevalence of Nonintegrase INSTI Resistance-Associated Mutations and Virological Outcomes in People Who Have Recently Acquired HIV-1 in the United Kingdom.

BACKGROUND: Integrase strand transfer inhibitors (INSTIs) are the mainstay of antiretroviral therapy (ART) globally. Virological breakthrough is uncommon but often manifests as low-level viremia, and only 50% of cases have identified drug resistance mutations in the integrase gene. Nonintegrase mutations in the Gag-nucleocapsid protein (NC), envelope glycoprotein (Env), and 3' polypurine tract (3'PPT) have been identified in vitro. METHODS: Between 2015 and 2021, human immunodeficiency virus type 1 (HIV-1) whole genome sequencing was performed on samples from people with recently acquired HIV

Synergistic antibacterial and antibiofilm activity of N-acetylcysteine combined with niosomal recombinant lysostaphin against MRSA Staphylococcus aureus.

Wound infections significantly impair healing and often result in chronic wounds, burdening healthcare systems substantially. The principal pathogen in such infections is Staphylococcus aureus (S. aureus), which forms biofilms that resist host defences and conventional therapies. To overcome these challenges, recombinant lysostaphin (rLyso) was encapsulated in niosomes using thin-film hydration and evaluated with N-acetylcysteine (NAC) against standard and clinical S. aureus strains. Antibacterial and antibiofilm activities were assessed by determining the minimum inhibitory concentration (MIC

A highly potent, stable, and safe dePEGylated lipopeptide against Nipah virus and related henipaviruses.

Nipah virus (NiV) poses a significant threat owing to its high mortality and the lack of approved therapeutics. Targeting the conserved heptad repeat 1 (HR1) domain of the viral fusion (F) protein constitutes a promising antiviral strategy. Also, lipopeptides derived from the human parainfluenza virus 3 (HPIV3) heptad repeat 2 (HR2) regions inhibit NiV fusion by blocking formation of the critical six-helix bundle (6-HB); however, their efficacy has been impeded by the controversial use of PEGylation. To resolve these limitations, we employed our proprietary heptad repeat 2 C-terminal fragment

Seneca Valley virus 3C protease targets the Nrf2/HO-1 pathway to antagonize its antiviral activity.

UNLABELLED: Seneca Valley virus (SVV) infection gives rise to severe vesicular diseases in pigs, presenting a substantial threat to the global swine industry. The redox imbalance resulting from oxidative stress is an essential pathogenic mechanism during viral infections. Nevertheless, the regulatory mechanisms of oxidative stress by viral and host factors during SVV infection remain elusive. In this study, we discovered that SVV elicited cellular oxidative stress through the induction of reactive oxygen species production and the suppression of the nuclear factor erythroid 2-related factor 2

Non-Coding RNAs as key regulators of interferon signaling in cancer immunotherapy: mechanistic insights and clinical prospects.

Non-coding RNAs (ncRNAs), including microRNAs (miRNAs), long non-coding RNAs (lncRNAs), and circular RNAs (circRNAs), play vital, context-specific roles in cancer, functioning as either tumor suppressors or oncogenic factors. Preliminary evidence suggests that a primary mechanism of ncRNA influence is through the interferon (IFN) signaling pathway, with a focus on how they may regulate IFN-induced immune surveillance and immunotherapy outcomes. Types I, II, and III IFNs regulate antitumor immunity, antigen presentation, and immune checkpoint activity, but can also support tumor growth in certa

Effect of pH on small-molecule inhibitor binding to influenza virus hemagglutinin.

Influenza A viruses (IAVs) impose a tremendous socioeconomic burden, and the mainstay preventative strategy of using vaccines faces challenges related to annual reformulation and variable efficacy (30-70%). The occurrence of antiviral resistance to the current Food and Drug Administration-approved anti-influenza drugs further highlights the urgent need for novel therapeutics. Our research group previously identified and optimized potent small-molecule inhibitors targeting IAV's hemagglutinin (HA), a surface glycoprotein crucial for viral entry and membrane fusion. Fusion occurs after the virus

Artesunate nanoliposome-hydrogel: a dual-modal therapy for post-surgical melanoma.

Background: Melanoma management faces the dual challenge of preventing tumor recurrence while ensuring optimal post-surgical wound healing, particularly problematic given melanoma's high recurrence rates and therapeutic resistance. Artesunate (ARS) emerges as a promising multimodal agent with concurrent anticancer, anti-inflammatory, and tissue-regenerative properties. However, its anti-melanoma mechanisms remain incompletely characterized, and clinical translation has been limited by suboptimal pharmacokinetics. Methods: We employed transcriptomic profiling (RNA-seq) to identify novel ARS-reg

Mutation rate variability in viral populations: Implications for lethal mutagenesis.

Lethal mutagenesis is a strategy to achieve viral extinction by drugging viral mutation rates beyond an extinction threshold. Accurate estimation of the extinction threshold is critical, as elevating viral mutation rates near, but not past this threshold increases the likelihood of mutations that could result in drug resistance, vaccine escape, or increased pathogenesis. Traditional models of lethal mutagenesis rely on the Poisson distribution, which assumes a uniform mutation rate across individuals. Yet, RNA viruses like influenza A virus (IAV) can have varied mutation rates due to mutations
