Berberine as a Multitarget Therapeutic Agent: Insights from Molecular Docking Studies.

Natural compounds are crucial in pharmacological research for modulating numerous biological processes. This is particularly helpful in the case of complex diseases and in overcoming challenges (e.g., drug resistance and adverse effects). Berberine, an isoquinoline alkaloid extracted from plants such as Berberis, Coptis, and Hydrastis species, exhibits a wide range of activities, including antidiabetic, anticancer, anti-inflammatory, antimicrobial, and antiviral. However, due to its low oral bioavailability (poor absorption, high first-pass metabolism, along with rapid elimination), further me

Biochemical and molecular insights into the antibiofilm and antivirulence effects of a B-phycoerythrin-rich extract from Porphyridium cruentum against Pseudomonas aeruginosa.

The growing resistance of Pseudomonas aeruginosa to conventional antibiotics has drawn increasing attention to antivirulence strategies that inhibit biofilm development and interfere with Quorum Sensing (QS) mechanisms. This study evaluated the biological activity of B-phycoerythrin (B-PE), a phycobiliprotein extracted from the red microalga Porphyridium cruentum, against Pseudomonas aeruginosa PAO1. This phycobiliprotein significantly inhibited biofilm formation by up to 62.4% and reduced performed biofilm by over 63% at 1/2 MIC, as quantified by crystal violet staining, causing notable morph

Adaptive landscape of the metallo-β-lactamase NDM-1 to emerging β-lactam-based therapies.

The New Delhi metallo-β-lactamase (NDM) is a major determinant of carbapenem resistance. This has prompted the development of novel therapeutic strategies to treat NDM producers. Since these therapies impose new selective pressures, their clinical deployment may favor NDM variants carrying escape mutations. To anticipate these events and inform therapeutic decisions, we explored the evolutionary landscape of NDM-1 under different selective constraints. To this end, we generated a diverse library of blaNDM variants and challenged it with zinc starvation, antibiotics, or β-lactam/β-lactamase inh

Cefepime-Zidebactam: Leveraging β-Lactam Enhancement Against Difficult-to-Treat and Metallo-β-Lactamase-Producing Gram-Negative Pathogens.

Carbapenem resistance among Gram-negative bacteria remains a serious public health threat, with metallo-β-lactamase (MBL)-producing organisms increasing substantially in prevalence and few therapeutic options available to adequately address these difficult-to-treat pathogens. Cefepime-zidebactam (FEP-ZID) is a novel β-lactam/β-lactam enhancer combination that offers a mechanistically distinct approach to overcoming resistance in Gram-negative organisms. Zidebactam contributes three complementary mechanisms: inhibition of Class A and C β-lactamases, direct antibacterial activity via PBP2 inhibi

[Pythium keratitis: a case report].

A 31-year-old female presented with a 3-week history of right eye redness, ocular pain, photophobia and decreased vision after tap water exposure. Slit-lamp examination revealed corneal lesions with pseudopodia and hill-shaped hypopyon. In vivo confocal microscopy (IVCM) detected filament-like structures in the corneal stroma. The patient was initially suspected of having fungal keratitis, but the condition deteriorated despite standardized antifungal treatment. Repeated corneal scraping culture and targeted DNA sequencing confirmed the infection of Pythium insidiosum. After switching to linez

Sigma-1 Receptor-Targeting Compounds Show Antiviral Activity Across Multiple Coronavirus Models.

Over the past two decades, coronaviruses have caused three major public health crises, including the epidemics of SARS-CoV, MERS-CoV, and SARS-CoV-2. Following the outbreak of SARS-CoV-2, several antiviral drugs have been approved for the treatment of COVID-19. However, the high mutation rate of SARS-CoV-2 has led to the emergence of multiple variants, some of which can escape vaccine-induced immunity and develop resistance to virus-targeted drugs. Viruses rely on host cell mechanisms to complete replication and transmission. Host-directed therapy is an emerging strategy that inhibits infectio

Transient receptor potential channel activation targets multiple stages of the schistosome parasite.

Schistosomiasis, a neglected tropical disease caused by infection with parasitic blood flukes of the genus Schistosoma, affects more than 250 million people worldwide. Currently, patients rely on a single medicine, praziquantel (PZQ), discovered half a century ago. To combat drug resistance and reach elimination of the schistosome parasite, alternative drugs to PZQ are required that treat and prevent disease. Here, we describe a drug candidate, M5493, with potent activity against multiple parasite life cycle stages. The target is a parasite transient receptor potential ion channel, distinct fr

Sulfathiazole scaffold remodeling into benzothiazole-thiazole Schiff bases: Antibacterial activity against Staphylococcus aureus, structure-activity relationships, and integrated DFT, docking and molecular dynamics studies.

The growing challenge of antimicrobial resistance highlights the need for rational modification of established antibacterial scaffolds. In this study, sulfathiazole (St) was structurally remodeled into a series of benzothiazole-thiazole Schiff-base derivatives (B1-B10) to enhance its antibacterial activity. The synthesized compounds were evaluated by antibacterial assays, SAR, DFT, MEP, molecular docking, ADMET prediction, and 100-ns molecular dynamics simulations. Within the narrow-spectrum screening panel, the derivatives showed preferential activity against the tested clinical Staphylococcu

Streptococcal mutanobactin mediates interspecies competition with gram-positive bacteria.

Polymicrobial communities where bacteria must compete with each other to persist can serve as a source of uncharacterized antibacterial compounds to develop drugs for the treatment of drug-resistant infections. This study investigates interbacterial competition between bacteria found in the oral cavity, where Streptococcus species comprise a large portion of the resident oral microbiota, and Enterococcus faecalis is a pathobiont that is commonly found in root canal infections. We used co-cultures to determine whether oral streptococci and E. faecalis compete with each other. Our experiments re

Understanding how bacterial cell wall peptidoglycan metabolism can be used to develop antimicrobial strategies.

INTRODUCTION: Despite the fact that the progress in developing new antibiotics with novel mechanism of action is limited owing to the considerable challenges involved and the modest financial returns, we are urgently needing new molecules with new mode of actions to overcome the increasing burden of multidrug resistance. Half of all antibiotics prescribed for human use target the bacterial cell wall. Targeting the cell wall has many advantages as it is essential for bacterial survival, with many steps readily accessible from the environment to small molecules and despite the success of beta-la

NIR Light-Driven Photocatalytic Antibacterial Hydrogels for Synergistic MRSA Biofilm Eradication and Wound Regeneration.

The treatment of drug-resistant bacterial biofilm infections remains a significant challenge in clinical practice. To address this challenge, 3D oxygen vacancy (OV)-rich iron-doped Bi2O2S (Fe-Bi2O2-XS, Fe-BOS) nanoflowers (NFs) are synthesized for the first time via an ion-exchange method. The resulting material exhibits a small band gap, abundant OVs, and favorable charge-transfer properties. It also shows robust photothermal performance and strong photocatalytic reactive oxygen species (ROS)-generation ability. Fe-BOS@C/H Gel is subsequently prepared by crosslinking hydrazide-modified chondr

Effectiveness and safety of meropenem-vaborbactam versus ceftazidime-avibactam in multidrug-resistant Gram-negative infections: a systematic review and meta-analysis with trial sequential analysis.

Antimicrobial resistance driven by multidrug-resistant (MDR) Gram-negative pathogens poses a major global threat, contributing to substantial morbidity and mortality. Novel β-lactam/β-lactamase inhibitor combinations, particularly meropenem-vaborbactam (M/V) and ceftazidime-avibactam (C/A), have expanded therapeutic options; however, their comparative efficacy and safety remain uncertain. This meta-analysis compared M/V and C/A in adult patients with MDR Gram-negative infections. MEDLINE, Embase, and Cochrane Central were searched for studies evaluating M/V versus C/A in hospitalized adults. O

A review on novel strategies to combat multidrug resistance in pathogenic bacteria exploiting synergism between essential oil and antibiotics.

The rapid rise of multidrug-resistant (MDR) bacteria has rendered many conventional antibiotics ineffective, therefore urging the need for novel treatment approaches. In the last few years, essential oils have been studied for their potential to enhance the effectiveness of conventional antibiotics. Essential oils contain several bioactive compounds, such as terpenes and phenols. In this review we discuss how the combination of essential oils with antibiotics can be used to improve their action against antibiotic resistant bacteria, including Staphylococcus aureus, Escherichia coli, Klebsiella
