The Rise of Extended-Spectrum β-Lactamase-Producing Ceftriaxone-Resistant Salmonella typhi in Western Rajasthan, India.

INTRODUCTION: Typhoid fever, caused by Salmonella typhi and Paratyphi, remains a significant public health concern, particularly in developing countries. The emergence of antimicrobial resistance, including resistance to first-line drugs, fluoroquinolones, and the development of resistance to ceftriaxone, poses a significant threat to effective treatment. METHODS: This study investigated extended-spectrum β-lactamase (ESBL)-producing Salmonella typhi isolates from blood samples of patients with suspected typhoid fever at a tertiary care hospital in Western Rajasthan, India, between April 2022

Azithromycin resistance in non-typhoidal Salmonella: a review.

Antibiotics are crucial for controlling severe or invasive non-typhoidal Salmonella (NTS) infections. However, the high resistance or toxicity of traditionally first-line antibiotics calls for alternatives. The excellent membrane permeability, long half-life and safe to use in children render azithromycin a promising candidate. However, azithromycin resistance has been observed in NTS isolates recovered from humans, food, animals and the environment, with particularly high rates in Asia. Although azithromycin resistance remains low in Europe, the USA and Australia, an increasing tendency has b

Novel KPC-2 Plasmid in a Clinical Salmonella Rissen Selected by Antibiotic Pressure.

In this work, we present the genomic characterization of a clinical Salmonella enterica serovar Rissen isolate harboring a novel IncN2 plasmid carrying the blaKPC-2 gene. The identified plasmid (pSEay-KPC) also encoded additional resistance genes, including blaACC-1, blaTEM-1, and qnrB. The pSEay-KPC conferred broad-spectrum antimicrobial resistance, allowing the pathogen to survive two consecutive antibiotic therapies with ceftriaxone and ciprofloxacin. Effective treatment was ultimately achieved with meropenem-vaborbactam, a last-resort agent. These findings highlight IncN2 plasmids as poten

Genetic Diversity and Antibiotic Resistance Profiling of Salmonella Isolates from Diarrhea Patients in Chifeng, China.

Salmonella is known to cause intestinal infections in humans, which can result in symptoms such as diarrhea, vomiting, and, in severe cases, sepsis and death. Isolates of Salmonella from specific populations and regions exhibit varying patterns of antibiotic resistance and genetic characteristics. This study aims to evaluate the diversity of 32 strains of Salmonella isolated from fecal samples of diarrhea patients in Chifeng City, China, between 2021 and 2023, through antibiotic resistance and genomic analysis. Microbroth dilution and whole genome sequencing were employed to investigate antibi

An mRNA platform to create isolated, monospecific Th1 responses.

Helper T cells (CD4 T cells) are lynchpins of adaptive immune responses. Each CD4 T cell expresses a single T-cell receptor, recognizing an epitope presented by major histocompatibility complex class II (MHC-II). Due to the enormous diversity of the T-cell repertoire, it is often desirable to study a population that responds to the same epitope. Here, we devised a novel method of selective immunization to produce a robust, monospecific helper T-cell response in mice using a modular mRNA vector. The vector encodes the target epitope attached via flexible linker to MHC-II. Immunization with this

Zero Salmonella prevalence found in common starlings Sturnus vulgaris captured in Danish cattle sheds on farms infected with Salmonella enterica Dublin.

Salmonella enterica serovar Dublin is host-specific to cattle, causing severe symptoms and economically impacting milk herds. Despite an eradication programme in Denmark, levels of infection have not decreased and suspicion has fallen on the common starling Sturnus vulgaris as a potential vector between herds. The number of breeding starlings in Denmark declined by 60% between 1976 and 2015, a trend correlated with decreases in the number of cattle grazing outside. Ironically, more starlings are now coming into Danish cattle sheds to feed on maize silage outside the breeding season, so it is i

Salmonella Typhimurium evades from host immunity and antibiotics in monocytes.

Intracellular bacteria can survive in vivo, evading host immunity and antibiotics. Salmonella persists in organs such as the spleen by invading the phagocytic cells. However, it remains controversial which specific cell populations, e.g. macrophages, monocytes, neutrophils, or dendritic cells, harbor the bacteria during persistent infection. To address this question, we employed a persistent infection mouse model using attenuated Salmonella expressing an acid tolerant fluorescent protein. We found that the bacteria predominantly resided in monocytes. Although these cells expressed Ly-6G, a typ

High indigestible dietary protein impairs growth and health status of nursery pigs with Salmonella Typhimurium more than with enterotoxigenic Escherichia coli F4 challenge.

Indigestible dietary protein content is an emerging dietary concept that is thought to be related to adverse intestinal health outcomes and increased incidence of pathogen-related diarrhea in pigs. The objective of this study was to examine the effect of IDP on growth performance, immune status, and fecal consistency score (FCS) of nursery pigs challenged with enterotoxigenic Escherichia coli F4 (ETEC) or Salmonella Typhimurium (ST). Thirty-two mixed-sex nursery pigs with an average initial body weight of 7.26 ± 0.40 kg were individually housed and randomly assigned to 1 of 2 dietary treatment

Effects of Defined Pig Microbiota on Acute Salmonellosis in Gnotobiotic Piglets.

Salmonella enterica subsp. enterica serovar Typhimurium (S. Typhimurium) is a ubiquitous Salmonella serovar that causes enterocolitis in humans, livestock, poultry, and some pets. Gnotobiotic animals are suitable for studying the development of initial colonization in the gastrointestinal tract, host-microbiota cross-talk, and interactions among microbiota strains. To clarify the impact of pre-colonization of the germ-free piglets with defined pig microbiota (DPM) on the subsequent infection with S. Typhimurium LT2 (LT2), we focused on the acute host immune response. Colonization with DPM did

The contrary short-term influence of heatwaves on Salmonellosis and Campylobacteriosis in Singapore - A national time-series analysis.

BACKGROUND: Salmonellosis and Campylobacteriosis are driven by climate variability and are among the most important causes of foodborne disease globally. Studies have largely reported positive associations between ambient air temperature and these diseases. However, those investigating how heatwaves influence these bacterial infections are few and inconsistent. I examined the short-term influence of heatwaves on these bacterial infections in the tropical city-state of Singapore. METHODS: All national reports of Salmonellosis and Campylobacteriosis in Singapore were analysed in negative binomi

Endovascular management of a non-typhoidal Salmonella-associated mycotic pseudoaneurysm of the iliac artery: a case report.

Non-typhoidal Salmonella (NTS) usually causes self-limiting gastroenteritis, but can lead to severe invasive infections, such as mycotic pseudoaneurysms, in immunocompromised patients. Here, we present the case of a 62-year-old male with NTS bacteraemia, which was complicated by an iliac artery mycotic pseudoaneurysm. The patient underwent successful endovascular management with a balloon-expandable graft stent, followed by targeted antimicrobial therapy against Salmonella enterica serotype Enteritidis. The patient achieved favourable clinical and radiological outcomes. This case highlights th

Salmonella meningitis due to horizontal transmission in an extremely premature neonate: A case report.

Salmonellosis is commonly associated with foodborne illnesses in adults. Meningitis, although rare, is a severe complication of a Salmonella infection, associated with high rates of mortality and significant neurological complications and sequelae. Premature infants are particularly susceptible for invasive infections due to their immature immune system. In this case report, we present an unusual case of Salmonella group B, serotype Typhimurium sepsis and meningitis in a 27-week premature neonate on the fourth day of life, resulting from horizontal transmission from mother to child, with fatal

Evaluation of resistance and virulence factors in Salmonella enterica subsp. enterica serovar Thompson isolates in the poultry chain.

1.This study investigated antimicrobial resistance and virulence factors in Salmonella enterica subsp. enterica serovar Thompson (SET) isolates from broilers in the Brazilian poultry industry.2. Among 44 isolates, 18.18% showed phenotypic resistance, with tetA, sul1 and sul2 genes frequently detected, while sul3 was absent. Two isolates carried clinically relevant resistance genes: blaCTX-M-1 (ESBL) and OXA-48 (carbapenemase).3. The PFGE analysis revealed five distinct clusters with low genetic diversity, which suggested a clonal dissemination pattern. One strain from cluster E showed the high

Determination of Effective Prophylactic Responses of Truncated Flagellin Protein as a Vaccine Candidate Against Uropathogenic Escherichia coli.

Uropathogenic Escherichia coli (UPEC) is a major cause of more than 80% of urinary tract infections (UTIs), a global health problem, and the second most common infectious disease. UTIs are responsible for approximately 40% of all nosocomial infections and 50% of all bacteremia. Since an approved vaccine against UTIs is not yet approved, evaluating different antigens, injection routes, and adjuvants is necessary to assess the ideal vaccines. In this study, we constructed a fusion protein composed of a truncated form of FimH antigen, the key virulence factor of UPEC, and the FliC antigen of Salm

Multivalent vaccines for invasive Salmonella disease: need, rationale, and immunological foundations.

Salmonella enterica infections are a major cause of morbidity and mortality worldwide, especially in sub-Saharan Africa and in the Asian continent, and are increasingly associated with antimicrobial resistance. Salmonella enterica serovars Typhi and Paratyphi A, B, and C cause enteric fever, while non-typhoidal Salmonella serovars (usually Typhimurium and Enteritidis) cause mainly gastroenteritis which can lead to systemic infections. Vaccines are only licensed against S. Typhi, but different combinations are in clinical development to prevent S. Typhi and S. Paratyphi A or S. Typhi and non-ty

Pathogens without borders: a review on cross-kingdom transmission strategies and pathogenicity of plant and human pathogens.

Emerging evidence indicates that certain plant pathogens-once thought to be restricted to the plant kingdom-can opportunistically infect humans, particularly those who are immunocompromised. This paradigm-shifting recognition of cross-kingdom pathogenicity underscores complex microbial adaptability and highlights an underexplored public health concern. This review is a compilation of current knowledge on bacterial (Pantoea, Burkholderia, Agrobacterium), fungal (Fusarium, Aspergillus, Curvularia), and viral (e.g., TMV, PMMoV) plant pathogens implicated in human disease. Shared eukaryotic cellul

PagN: a multifunctional outer membrane protein in Salmonella virulence and host interaction.

Salmonella utilizes multiple virulence factors to adhere to, invade, and replicate within host cells, ensuring systemic dissemination. Among these, the outer membrane protein PagN functions as both an adhesin and invasin, playing a central role in early infection stages and in establishing persistent intracellular populations. PagN expression is controlled by the PhoP/PhoQ two-component system and is specifically induced under host-related environmental conditions such as acidic pH, low magnesium concentration, and the presence of antimicrobial peptides—conditions typically encountered within
