The advent of single-molecule nanopore sequencing established a powerful platform for modern genomics by using static biological pores to report the translocation of canonical nucleic acids, enabling rapid, accessible nucleic acid analysis. However, extending this strategy to single-molecule proteomics has stalled against a fundamental biophysical bottleneck. Current efforts in nanopore proteomics attempt to retrofit these static, spatial "caliper" biological nanopores (e.g., α-hemolysin, MspA, CsgG, aerolysin) for protein sequencing despite the immense steric, charge, and conformational heter

Physical activity (PA) is a modifiable lifestyle behaviour associated with lower dementia risk; however, molecular pathways bridging PA-related dementia prevention are poorly understood. We leveraged large-scale plasma proteomics to identify biological signatures of objectively monitored PA and cognitive ageing in functionally intact older adults, cross-validated these signatures in independent exercise cohorts and tested associations with both symptomatic and presymptomatic stages of neurodegeneration across multiple Alzheimer's disease and related dementias (ADRD) cohorts. We analysed large-

Disease outbreaks are non-random public health phenomena attributed to known or idiopathic human, animal, and animal-based factors; a realm referred to as the One Health concept; a new global public health order for control and prevention of epidemics, endemics, and pandemics. These are diseases of viral, prion, bacterial, fungal, and parasitic origin, transmitted between vertebrate animals and humans and vice versa. Emergence and re-emergence of zoonotic diseases such as rabies, Ebola, COVID-19, monkeypox, avian influenza, anthrax, brucellosis, leptospirosis, and Lyme disease continue to affe

We report a rare survival case of anthrax meningoencephalitis in a 56-year-old male from a low-incidence region. The patient presented with nasal discharge, fever, headache, and rapid onset of coma following the slaughter of a diseased cow. Physical examination revealed a characteristic ulcerative eschar on the right index finger, while laboratory investigations showed significant leukocytosis and hemorrhagic cerebrospinal fluid (CSF) characterized by elevated protein and decreased glucose levels. Although initial microscopy misidentified the pathogen as Bacillus cereus, metagenomic next-gener

Anthrax, caused by Bacillus anthracis (BA), is a prominent neglected zoonosis with major impacts on human, livestock, and wildlife health. Despite this, limited genomic investigation at the One Health interface constrains current understanding of BA transmission and of the ecological and host factors shaping its diversity and population structure. This includes the possibility of host-specific BA lineages, given that anthrax outbreaks often disproportionally affect individual species. This study characterises the genomic diversity of BA in an endemic area, the Ngorongoro Conservation Area (NCA

BACKGROUND: Anthrax remains a life-threatening zoonotic disease in resource-limited settings. Adsorbed anthrax vaccine (AVA, BioThrax) is the only United States-authorized preexposure prophylaxis for high-risk Bacillus anthracis exposure. Although AVA safety has been described, depression-related adverse events (AEs) remain insufficiently characterized. This study evaluated real-world depression-related safety signals associated with AVA and anthrax-related antibacterial agents.
METHODS: AVA-related AE reports were extracted from the Vaccine Adverse Event Reporting System (VAERS), and depress

Anthrax lethal toxin is a key virulence factor of Bacillus anthracis. However, non-canonical epitopes of protective antigen (PA) and alternative neutralization mechanisms beyond receptor blockade or prevention of proteolytic cleavage remain underexplored. Here, we characterize a fully human monoclonal antibody, 22F1, with ultrapotent toxin neutralization in vitro (IC50 = 0.027 nM) and in vivo (molar ratio 1:7.5-15). Unlike known PA antibodies, 22F1 selectively binds the post-cleavage form PA63, not full-length PA83, due to shielding of the epitope by PA20. Cryo-EM reveals a novel conformationa

Environmental persistence of Bacillus anthracis spores sustains anthrax transmission, necessitating rapid and field-deployable detection tools. This study aimed to develop and quantitatively evaluate a LAMP-assisted CRISPR-Cas12a assay for sensitive and specific detection of B. anthracis spores in soil and meat meal matrices. Two B. anthracis-specific target genes plasmid-encoded lef and chromosomal SNP locus (CR5)- were selected. Target-specific LAMP primers and CRISPR crRNAs were designed using Primer Explorer, CRISPOR, and RNAfold platforms. The LAMP-CRISPR/Cas12a assay was optimised for re

SUMMARYIn 1876, Robert Koch published "The Etiology of Anthrax Disease, based on the Developmental History of Bacillus anthracis." This seminal work laid the foundation for microbiology as a discipline. To commemorate the 150th anniversary of its publication, this article will review the history of B. anthracis, from its evolutionary origins to recent advancements in understanding its pathogenesis. For millions of years, B. anthracis existed in the environment as a unique pathogen whose lifestyle was heavily reliant on causing lethal disease in mammalian hosts. Beginning around 10,000 years ag

A combined molecular strategy was developed to accurately differentiate wild-type and vaccine strains of Bacillus anthracis. This approach integrates a multiplex TaqMan qPCR assay targeting the BclA, sap, and cap genes with conventional PCR analysis of BclA fragment length polymorphism. The qPCR assay demonstrated high specificity, with no cross-reactivity against other common pathogens, and exhibited a consistent limit of detection of 2.11 × 101 copies/μL (Cq < 35) for all three targets. Excellent repeatability was confirmed, with intra- and inter-assay coefficients of variation below 1.96% a

This review examines the historical development of biological warfare from premodern contamination practices to twentieth-century state programs, post-9/11 bioterrorism, and the renewed prominence of biological risk in the COVID-19 era. Drawing on historical, policy, and public-health scholarship, it shows how disease came to be understood not only as a natural threat but also as a strategic instrument shaped by scientific knowledge, military organization, and administrative secrecy. The review traces the transition from empirical uses of contamination in siege warfare and colonial settings to

INTRODUCTION: The incidence of anthrax in China has declined since 1990 but has resurged in recent years. The purpose of this study was to describe the epidemiology of anthrax and Bacillus anthracis (B. anthracis) in China from 2020 to 2024 to inform control strategies.
METHODS: A descriptive analysis was performed of case characteristics. B. anthracis strains underwent whole-genome sequencing, and phylogenetic relationships were determined by core-genome single nucleotide polymorphism (cgSNP) and canonical single nucleotide polymorphism (canSNP) analysis.
RESULTS: Between 2020 and 2024, 1,8

WHAT IS ALREADY KNOWN ABOUT THIS TOPIC?: Anthrax is a zoonotic disease caused by the bacterium Bacillus anthracis (B. anthracis). Inhalation anthrax, caused by inhalation of B. anthracis spores, is the rarest and most fatal form of anthrax. Historically, naturally acquired inhalation anthrax cases have mainly been associated with occupational exposure to infected animals or animal products, such as in wool mills or tanneries.
WHAT IS ADDED BY THIS REPORT?: This report documents the first case of inhalation anthrax in China that was linked to environmental exposure in a historically endemic ar

WHAT IS ALREADY KNOWN ABOUT THIS TOPIC?: Inhaling Bacillus anthracis spore-laden dust or aerosols can cause inhalational anthrax; such direct-inhalation infection is uncommon and usually affects people who handle diseased animals or animal products.
WHAT IS ADDED BY THIS REPORT?: On May 2, 2026, a Beijing hospital reported confirmed inhalational anthrax in a Kaifeng resident. Exposure to a previously contaminated environment was the probable route of infection, and targeted next-generation sequencing (tNGS) was key to the diagnosis.
WHAT ARE THE IMPLICATIONS FOR PUBLIC HEALTH PRACTICE?: This

PURPOSE: Retrograde peri-implantitis (RPI) has been hypothesized to be associated with apical periodontitis (AP). This retrospective study evaluated whether adjacent AP was associated with RPI and whether implant-to-lesion distance modified this association.
METHODS: A total of 1,047 patients with 1,819 implants were included; 56 implants in 56 patients were placed near teeth with AP. RPI was diagnosed during follow-up using radiographic and clinical examinations. Firth's penalized logistic regression was first used to assess the association between adjacent AP and RPI development. Implant-to

BACKGROUND: Infectious Medical Waste (IMW) from both human and animal healthcare sources have the potential to transmit diseases in human populations due to its pathogenic composition. Conversely, a paucity of data regarding cause-and-effect relationships inhibits our understanding of the extent to which disease transmission occurs. In parts of Africa where the management of IMW is not prioritized, the risks to public health, along with its global implications cannot be ignored.
METHODOLOGY: A systematic literature search following the PRISMA guidelines was conducted across five electronic da
