Burkitt's lymphoma (BL) is common in sub-Saharan Africa, yet diagnosis is often delayed due to limited pathology capacity. Here we evaluated blood-based liquid biopsies from 377 children and young adults with clinically suspected lymphoma at four hospitals in Tanzania and Uganda, assessing diagnostic accuracy and turnaround time (TAT). After extensive pathology capacity building, a gold-standard diagnosis was established using tissue morphology, a limited validated immunohistochemistry panel and independent dual histopathologist review. Using clinical features and circulating tumor DNA markers

OBJECTIVES: The aim of this study was to assess the burden of Burkitt lymphoma (BL) at the global, regional, and national levels from 1990-2021 and to project trends over the next 15 years.
METHODS: Data from the 2021 Global Burden of Disease database was used for analysis.
RESULTS: Both the incidence and prevalence rates of BL increased from 1990-2021. BL demonstrated a bimodal age pattern, with incidence peaking among children aged 5-9 years and again in adults older than 65 years. The disease burden was consistently greater in males than in females. A significant positive correlation was

High-grade B-cell lymphoma with 11q aberration (HGBL-11q) is a recently recognized entity according to the 2022 WHO classification that often morphologically and immunohistochemically resembles Burkitt lymphoma but has distinct molecular characteristics. In HGBCL-11q, the defining abnormality is a characteristic gain/loss pattern of chromosome 11q without MYC rearrangement, whereas Burkitt lymphoma is characterized by a classical MYC rearrangement with one of the three immunoglobulin genes. HGBL-11q is a rare lymphoma that predominantly affects children and adolescents. Clinically, patients ty

Epstein-Barr virus (EBV) is associated with multiple malignancies including Burkitt lymphoma (BL), Hodgkin's lymphomas, nasopharyngeal carcinomas (NPC), and gastric cancers. Canonically, EBV positive tumors display latent gene expression programs that are difficult to target pharmacologically. To overcome this hurdle, lytic reactivation therapies have been developed based on HDAC inhibition with limited mechanistic studies. We therefore characterized the impact of pan-HDAC inhibitor, panobinostat, and class I HDAC inhibitor, nanatinostat, on the growth, survival, and lytic reactivation of four

Epstein-Barr virus (EBV) persistently infects over 95% of adults worldwide and is associated with a range of cancers, including lymphomas and epithelial malignancies. Despite advances in understanding EBV biology, targeted therapies for EBV-associated cancers remain limited. To identify novel dependencies in EBV-infected cancers, we performed genome-wide CRISPR-Cas9 loss-of-function screens in EBV+ lymphoblastoid versus Burkitt lymphoma cells, which differ by EBV latency programs. JunB emerged as a critical LCL-selective host dependency factor. LCL JunB knockout significantly decreased prolife

Epstein-Barr virus (EBV) is associated with 200,000 cancers per year, including Burkitt lymphoma and post-transplant lymphomas. We previously reported that EBV latency oncogene programs dynamically remodel infected B cell metabolism and sensitivity to induction of ferroptosis, a programmed cell death pathway driven by lipid reactive oxygen species. However, much has remained unknown about how EBV remodels key redox defense pathways in support of infected B cell proliferation. Here, we identify EBV latent membrane protein 1 (LMP1), a key viral oncogene necessary for B cell immortalization and w

Epstein-Barr virus is a globally disseminated oncovirus capable of causing various malignancies, including gastric cancer, Burkitt lymphoma, and Hodgkin's lymphoma. The influence of recombination on the EBV genome revealed limitations in the current traditional EBV classification, and the extent of these recombination events across the EBV genome is not fully understood. The nuclear antigen 3C (EBNA3C) is an indispensable gene in the oncogenesis of the virus. Despite its critical role, little is known about EBNA3C sequence variability. We examined 988 EBNA3C gene sequences extracted from EBV g

February, 2026 is the 40th anniversary of the 1st publication of rapid recovery of bone marrow function after high-dose chemotherapy and infusion of an autologous blood- rather than a bone-marrow-derived graft. The recipient, a man with advanced Burkitt lymphoma, is in remission and alive. In the next 40 years transplants of blood-derived progenitor cells have become the most common graft for auto- and allotransplants. The rapid hematopoietic recovery after blood cell-derived grafts is associated with fewer short-term complications and better outcomes in many settings compared with bone marrow

RuvB-like 1 (RUVBL1) and RuvB-like 2 (RUVBL2) are AAA ATPases that form hetero-oligomeric complexes involved in diverse cellular functions. Increasing evidence implicates the RUVBL1/2 complex as an essential cofactor of MYC, with RUVBL1/2 inhibition reducing c-MYC levels in vitro. Herein, we report a potent RUVBL1/2 inhibitor discovered through a Single-Molecule Tracking (SMT)-driven SAR campaign. Compared with a biochemical ADP-Glo assay, which exhibited limited dynamic range and poor reproducibility under our experimental conditions, the live-cell high-throughput RUVBL SMT assay provided rob

Background and Clinical Significance: Burkitt lymphoma is an aggressive form of non-Hodgkin lymphoma of B-cell origin, caused by a MYC gene translocation on chromosome 8. There are three clinical subtypes, of which the sporadic subtype is most prevalent in the United States. Sporadic Burkitt lymphoma is diagnosed at a median age of 30 years and commonly manifests as bulky abdominal lesions, most often involving the ileocecal region. Pancreatic involvement is uncommon, and presentation as acute pancreatitis secondary to Burkitt lymphoma is exceedingly rare. Case Presentation: We present a case

BACKGROUND: Chimeric antigen receptor (CAR) T cells are highly potent therapies approved by the U.S. Food and Drug Administration (FDA) for several hematological malignancies. However, efficacy remains variable due to resistance, antigen modulation, systemic toxicities, and relapse. Developing improved CAR T cell therapies relies on preclinical models that accurately predict clinical outcomes. For this purpose, immortalized tumor cell lines, such as the Burkitt's lymphoma line Raji, are commonly used.
METHODS: CD19-targeting CAR (CAR19) and non-transduced (NT) human T cells were co-cultured w

Survival outcomes for pediatric Burkitt lymphoma (BL) substantially vary depending on geography (50-90%), which also serves as a proxy for the prevalence of Epstein-Barr virus (EBV) within the tumors. Although BL is considered an immunologically "cold" tumor with few tumor-infiltrating lymphocytes (TILs), their functional status has not been fully evaluated, especially for EBV-positive disease. Here, we characterize the exhaustion and activation profiles of T cells in the tumor microenvironment (TME) of EBV-positive BL using orthogonal methods, single-cell gene expression analysis, spectral fl

Burkitt lymphoma is a highly aggressive mature B-cell non-Hodgkin lymphoma characterized by MYC dysregulation and a high proliferative index. Although the sporadic form commonly presents with abdominal involvement, maxillofacial manifestations in children may mimic odontogenic infections, leading to delayed diagnosis. We report a nine-year-old boy with a rapidly progressive unilateral mandibular swelling initially treated as a dental abscess without improvement. Imaging revealed an ill-defined osteolytic mandibular lesion with cortical erosion and soft tissue extension, raising suspicion for m

Burkitt lymphoma is a very fast-growing non-Hodgkin lymphoma (NHL) characterized by rapid tumor progression and increased lymphocyte proliferation. It often involves the jaw and facial bones. The current case report represented a 37-year-old male patient with Burkitt's lymphoma who sought dental care for persistent and dull pain and paresthesia on the left side of his mandible extending to the periauricular region. The initial diagnosis suggested a dental origin, leading to the lower left third molar (LL8) extraction. Despite this, the patient continued to experience worsening pain, which radi

Burkitt lymphoma (BL) is a rare, aggressive B-cell non-Hodgkin lymphoma with rapid proliferation. Typical manifestations include abdominal disease, B-symptoms and bone marrow infiltration, while rare presentations may include nodal disease, facial palsy and breast involvement. This case report presents a 17-year-old girl with sporadic BL who developed peripheral facial palsy and rapid bilateral breast enlargement. This case highlights the importance of systematic follow-up after facial palsy in young patients and attention to associated symptoms and biochemical abnormalities suggestive of mali

Cyto-morphological analysis, combined with the study of genetic rearrangements, is central to the classification of aggressive B-cell lymphomas in WHO-5. Cytology distinguishes large-cell morphology from Burkitt-type or blastic cells, the latter two defining high-grade morphology. DLBCL-NOS is defined by large-cell morphology and may harbor isolated or combined MYC and BCL6 rearrangements. In contrast, the association of MYC and BCL2 excludes the diagnosis of diffuse large B cell lymphoma NOS (DLBCL-NOS) and defines a high-grade B-cell lymphoma. An isolated MYC rearrangement is associated with

BACKGROUND AIMS: CD19 chimeric antigen receptor (CAR) T-cell therapy has shown promise in treating post-transplant lymphoproliferative disorder, but the risk of graft rejection is acknowledged and there is no consensus on the use of calcineurin inhibitors with CAR T-cell therapy.
METHODS: We report a kidney transplant recipient with refractory Burkitt lymphoma who received cyclosporine A (CsA) following CD19 CAR T-cell infusion. Serial peripheral blood sampling was performed before and after CsA administration, and samples were analyzed via single-cell RNA sequencing.
RESULTS: The patient ex
