CircBRWD3 promotes non-small cell lung cancer progression through enhancing the stability of TRIM25 to increase p53 ubiquitin degradation and upregulating SPTBN2.

Circular RNAs (circRNAs) play crucial roles in cancer progression and metastasis. However, the expression profiles and biological roles of circRNAs in non-small cell lung cancer (NSCLC) remain unclear. In this study, we found a novel circRNA, hsa_circ_0001936(circBRWD3), in NSCLC and investigated the biological role of circBRWD3 in NSCLC progression in vitro and in vivo. qRT-PCR was used to quantify the expression of circBRWD3 in NSCLC tissues, serum and cells. Cell growth curves, colony formation, Edu assay and transwell assays were conducted to study the function of circBRWD3. Finally, the m

HPV detection by in situ hybridisation in sinonasal inverted papilloma and squamous cell carcinoma.

Dear Editor: Sinonasal inverted papilloma (SNIP) is a rare benign but locally aggressive tumour with an annual incidence of approximately 0.5-1.5/100 000, accounting for 4.7% of all sinonasal tumours. SNIP may recur and can transform into sinonasal squamous cell carcinoma (SNSCC) (1). Human papillomavirus (HPV) has been linked to SNIP recurrence and malignant progression, with a meta-analysis reporting the incidence of HPV in SNIP as 16.9% (2). This aligns with the results of our previous prospective study (17.8%), showing that the presence of HPV DNA was associated with SNIP recurrence in 90

Digital analysis of spatial expression of stromal fibroblast activation protein alpha (FAP) in DCIS.

INTRODUCTION: The presence of non-progressing ductal carcinoma in situ (DCIS) cases has raised concerns of overtreatment, necessitating comprehensive risk prediction. The tumour microenvironment, especially cancer-associated fibroblasts (CAFs), plays a key role in DCIS progression. Fibroblast activation protein alpha (FAP), a CAF biomarker, has shown prognostic potential, but its clinical significance and spatial organization in DCIS remain rarely discussed. METHODS: In this study, we employed a digital pathology pipeline to quantify stromal FAP expression and map its spatial organization. A

Preoperative MRI for pleomorphic lobular carcinoma in situ: Implications for surgical management.

BACKGROUND: Pleomorphic lobular carcinoma in situ (PLCIS), a more aggressive variant of classic LCIS (CLCIS), demonstrates higher rates of upgrade to invasive carcinoma at surgical excision. Mammography may underestimate disease extent, whereas MRI may delineate lesions due to sensitivity. METHODS: Our institutional pathology database was queried for patients diagnosed with PLCIS on core needle biopsy (CNB) from 2002 to 2025 who underwent surgical excision. Preoperative MRI and MRI-guided biopsies were reviewed. Radiologic, pathologic, surgical, and outcome data were collected. Proportions we

Invasive interval breast cancer rates after digital breast tomosynthesis plus synthesised mammography versus digital mammography screening (TOSYMA): 24-month follow-up of the multicentre, open-label, randomised, controlled, superiority trial.

BACKGROUND: Despite high invasive breast cancer detection rates, the effectiveness of digital breast tomosynthesis screening is still uncertain. We aimed to compare the invasive interval cancer rates in women screened with digital breast tomosynthesis plus synthesised two-dimensional (s2D) mammography with those screened with digital mammography. METHODS: TOSYMA is a multicentre, open-label, parallel-group, randomised, controlled, superiority trial done at 17 screening sites across two federal states in Germany. Women aged 50-70 years were randomly allocated (1:1) to digital breast tomosynthe

Channel attention of the image for distinguishing ductal carcinoma in situ from benign mimics based on ultrasound images.

BACKGROUND: Ductal carcinoma in situ (DCIS) is widely recognized as a precursor to invasive breast cancer, with an estimated 20-50% of cases progressing to invasive disease. The early detection of DCIS could save doctors and patients more time to deal with the breast cancer, which has posed a great threat to the life safety of women worldwide. This study aimed to develop a deep learning algorithm based on the image-level attention to distinguish DCIS from the benign mimics on ultrasound images, assisting doctors in screening for benign breast lesions. METHODS: By screening the biopsy results

Early-stage HER2-positive breast cancer in neurofibromatosis type 1: a case report with literature review.

Neurofibromatosis type 1 (NF1) is an autosomal dominant disorder associated with an increased risk of malignancies, particularly breast cancer at a younger age. We report the case of a 48-year-old woman with clinically diagnosed NF1 who presented with a left breast mass detected during routine screening. Physical examination revealed numerous café-au-lait macules and cutaneous neurofibromas distributed over the trunk and extremities. Imaging findings were classified as BI-RADS 4B-4C. Following intraoperative confirmation of malignancy, total mastectomy and sentinel lymph node biopsy were perfo

Spatial Atlas of Small Cell Lung Carcinoma Reveals Intratumoral Tumor-State Heterogeneity and an MMP12+ Macrophage Niche.

Small-cell lung carcinoma (SCLC) is a highly aggressive tumor with a dismal prognosis among lung cancers. Treatment is failing because of primary or rapidly developing secondary resistance. Recent molecular studies have suggested four SCLC subtypes with unique therapeutic vulnerabilities-ASCL1, NEUROD1, POU2F3, and YAP. These subtypes still distinguish themselves by considerable intra-tumoral heterogeneity and the influence of the microenvironmental context, which are both considered to be responsible for treatment resistance. We integrated in situ hybridization-based spatial transcriptomics w

Nonmuscle-invasive bladder cancer after solid organ transplantation: Clinical outcomes and recurrence patterns.

BACKGROUND: Solid organ transplant recipients face an increased risk of bladder cancer and require lifelong immunosuppression, which may alter the natural history and treatment response of nonmuscle-invasive bladder cancer (NMIBC). Balancing cancer control with allograft preservation complicates clinical decision-making, yet outcome data in this population remain scarce. METHODS: A retrospective study of transplant recipients diagnosed with NMIBC post-transplant was conducted. The index date was NMIBC diagnosis. NMIBC recurrence was the primary endpoint, with death before recurrence as a comp

TFEB-Amplified Renal Cell Carcinoma With Focal Rhabdoid Features, Hyaline Globules, Giant Tumor Cells, and Osseous Metaplasia.

TFEB-amplified renal cell carcinoma (RCC) is a rare, molecularly defined RCC, characterized by amplification of the TFEB gene on chromosome 6p21.1, with fewer than 100 tumors reported to date. Herein, we report a TFEB-amplified RCC with focal rhabdoid features, hyaline globules, giant tumor cells, and osseous metaplasia. This tumor further expands the histological spectrum of TFEB-amplified RCC. A 59-year-old man underwent radical nephrectomy for a 7.6-cm left renal mass. Histologically, the tumor demonstrated predominantly papillary architecture (90%) with focal solid areas (10%). Tumor cells
