Rapidly Progressive Buschke-Löwenstein Tumor Managed With Wide Local Excision and Healing by Secondary Intention: A Case Report and Literature Review.

Buschke-Löwenstein tumor (BLT) is a rare, locally aggressive anogenital tumor associated predominantly with low-risk human papillomavirus (HPV) types 6 and 11. BLT is classically described as a slow-growing lesion predominantly affecting middle-aged men who are immunocompromised; cases of rapid progression are rarely reported. We present the case of a 23-year-old male with a progressively enlarging perianal mass over 1 year that was accompanied by rectal pain and intermittent bleeding. He reported prior unprotected heterosexual intercourse, though serologic testing for HIV, syphilis, and hepat

Recurrent TFE3-Rearranged Renal Cell Carcinoma Responding to Pembrolizumab Plus Lenvatinib After Postoperative Pembrolizumab: A Case Report.

INTRODUCTION: TFE3-rearranged renal cell carcinoma (RCC) is rare, and evidence guiding systemic therapy is limited. CASE PRESENTATION: A 25-year-old man with gross hematuria had a left renal tumor with hilar lymph node metastasis. Robot-assisted radical nephrectomy and hilar lymph node dissection were performed. Histology, immunohistochemistry, and fluorescence in situ hybridization confirmed TFE3-rearranged RCC, pT3aN1. Postoperative pembrolizumab was started because of the high risk of recurrence. Eight months after surgery, lung and retroperitoneal metastases developed. Lenvatinib was adde

LncRNA AP003469.2 promotes hepatocellular carcinoma progression by regulating cuproptosis via the miR-4732-3p/LIPT1 axis.

BACKGROUND: Hepatocellular carcinoma (HCC) is a malignant tumor characterized by high recurrence and metastasis rates, as well as a poor prognosis. Cuproptosis is a form of programmed cell death driven by abnormal copper-ion metabolism. Existing studies have shown that its occurrence is closely regulated and associated with the progression of HCC. However, it is still unclear how long non-coding RNAs (lncRNAs) control the process in HCC. The purpose of this work is to clarify how the lncRNA AP003469.2 influences cuproptosis and promotes the development of HCC. METHODS: This study utilized RNA

CircUGP2 inhibits tumor progression via PI3K/AKT/mTOR signaling through sponging miR-137-3p and interacting with UPF1 in hepatocellular carcinoma.

BACKGROUND: Circular RNA (circRNA) has been shown to have a pivotal role in the initiation and evolution of multiple tumors, but its function in hepatocellular carcinoma (HCC) requires further elucidation. METHODS: circUGP2 (hsa_circ_0001020) was identified in the GEO database by HCC circRNA microarray. Subsequently, an assessment was conducted to determine the biological effects of circUGP2, employing a series of analytical methods, including CCK8, EdU, colony formation, transwell, scratch assays, and in vivo experimentation. To explore the functions of circUGP2 as a sponge, including RNA pu

Integrating scRNA-Seq and Spatial Transcriptomics Reveal Novel Risk Stratification Signatures in Cervical Carcinoma.

Cervical cancer is the fourth most common malignancy in women worldwide. Biomarker-assisted screening may improve diagnostic accuracy, but biomarkers predicting progression of low-grade squamous intraepithelial lesions (LSIL) remain limited. We integrated single-cell RNA sequencing and spatial transcriptomics to identify prognostic biomarkers associated with LSIL outcomes, followed by validation using improved in situ sequencing (IISS). Analysis of 341,978 single cells identified eight major cell populations and twelve epithelial subclusters, including a previously unrecognized subcluster, epi

Spatial Transcriptomics in Thyroid Cancer: A PRISMA-Style Systematic Review of Platforms, Architecture, and Clinical Applications.

Background/Objectives: Thyroid cancer spans indolent differentiated tumors to lethal poorly differentiated and anaplastic carcinomas, and histopathology and molecular testing do not capture the spatial organization of tumor, stromal, and immune compartments that underlies invasion, dedifferentiation, and treatment resistance. Spatial transcriptomics profiles gene expression in situ. We systematically reviewed its platforms, applications, and findings in thyroid cancer. Methods: PubMed/MEDLINE, Scopus, and Web of Science were searched, following PRISMA 2020 guidance and supplemented by citation

High-Frequency Ultrasound in High-Risk Cutaneous Squamous Cell Carcinoma.

INTRODUCTION: High-frequency ultrasound (HFUS) is a valuable tool for the preoperative tumor assessment of cutaneous squamous cell carcinoma (cSCC). A subgroup of patients with cSCC is at an increased risk of tumor recurrence, metastasis, and disease-specific mortality - high-risk cSCC (HRcSCC). The use of HFUS can provide early knowledge of tumor thickness and depth of invasion, allowing for more appropriate surgical planning and staging in this subset of patients. MATERIALS AND METHODS: We designed a prospective observational cohort study on patients diagnosed with cSCC and surgically treat

Dying cells initiate DLL4-Notch signaling to drive tumor repopulation after insufficient thermal ablation of papillary thyroid carcinoma.

BACKGROUND: Although lacking high-quality clinical evidence, the use of thermal ablation for papillary thyroid carcinoma (PTC) is increasing. Emerging evidence suggests that thermal ablation may accelerate tumor progression in some patients, but the underlying biological mechanisms remain unclear. This study aims to explore the mechanisms of tumor repopulation following thermal ablation and identify possible intervention strategies. MATERIALS AND METHODS: In vitro and in vivo models were employed to simulate tumor repopulation after thermal ablation and investigate mechanisms. Clinicopatholog

Breast cancer overdiagnosis due to mammography screening-A long-term follow-up population study of BreastScreen Norway.

ObjectivesEstimates of breast cancer overdiagnosis related to mammography screening vary substantially. Overdiagnosis is commonly defined as cases that would not have been detected during the person's remaining lifetime in the absence of screening. We here aim to quantify overdiagnosis in the population-based BreastScreen Norway mammography screening program using long-term follow-up and more detailed modeling than previous studies.SettingWe applied data on Norwegian screening patterns and breast carcinoma incidence for the period 1987-2019, covering women aged 49-84, leveraging the gradual im

The State of Mohs Micrographic Surgery in 2025-2026: Current Challenges, Pressures, and the Path Forward.

BACKGROUND: Mohs micrographic surgery (MMS) unites staged extirpation, comprehensive intraoperative histopathologic assessment, and immediate reconstruction to deliver durable local control with tissue conservation in anatomically critical sites. Contemporary practice, however, is shaped by intersecting forces: stewardship of indications, reimbursement trends, vertical consolidation, expanding use of intraoperative immunohistochemistry (IHC), and the emergence of complementary modalities (superficial radiation therapy; hedgehog and programmed cell death-1 [PD-1] inhibitors) alongside digital t

Extremely rare primary tracheal hyalinizing clear cell carcinoma: two case reports.

BACKGROUND: Primary tracheal hyalinizing clear cell carcinoma (HCCC) is an exceptionally rare, low-grade malignant epithelial tumor. Its nonspecific respiratory manifestations and overlapping histopathological features with other clear cell neoplasms can delay diagnosis. Molecular alterations involving the Ewing sarcoma breakpoint region 1 (EWSR1) gene are diagnostically valuable; however, the clinicopathological features, optimal management, and long-term outcomes of tracheal HCCC remain insufficiently characterized. We present two cases to expand the limited evidence regarding its diagnosis

Aberrant HER2 Expression as a Specific and Feasible Diagnostic Marker for Fumarate Hydratase-Deficient Renal Cell Carcinoma.

The presence of human epidermal receptor 2 (HER2) in fumarate hydratase-deficient renal cell carcinoma (FH-deficient RCC) remains unexplored. We investigated immunohistochemical staining for HER2 (clone 4B5), FH, 2SC, and AKR1B10 on 72 cases of FH-deficient RCC and 440 cases of other RCC subtypes. HER2 positivity was observed in 88.9% (64/72) of FH-deficient RCCs, manifesting as cytoplasmic only (6.9%), cytoplasmic and nuclear (47.2%), cytoplasmic and membranous (2.8%), or cytoplasmic, nuclear, and membranous (31.9%) patterns. Notably, all four cases of low-grade FH-deficient RCC exhibited cyt

CircBRWD3 promotes non-small cell lung cancer progression through enhancing the stability of TRIM25 to increase p53 ubiquitin degradation and upregulating SPTBN2.

Circular RNAs (circRNAs) play crucial roles in cancer progression and metastasis. However, the expression profiles and biological roles of circRNAs in non-small cell lung cancer (NSCLC) remain unclear. In this study, we found a novel circRNA, hsa_circ_0001936(circBRWD3), in NSCLC and investigated the biological role of circBRWD3 in NSCLC progression in vitro and in vivo. qRT-PCR was used to quantify the expression of circBRWD3 in NSCLC tissues, serum and cells. Cell growth curves, colony formation, Edu assay and transwell assays were conducted to study the function of circBRWD3. Finally, the m

HPV detection by in situ hybridisation in sinonasal inverted papilloma and squamous cell carcinoma.

Dear Editor: Sinonasal inverted papilloma (SNIP) is a rare benign but locally aggressive tumour with an annual incidence of approximately 0.5-1.5/100 000, accounting for 4.7% of all sinonasal tumours. SNIP may recur and can transform into sinonasal squamous cell carcinoma (SNSCC) (1). Human papillomavirus (HPV) has been linked to SNIP recurrence and malignant progression, with a meta-analysis reporting the incidence of HPV in SNIP as 16.9% (2). This aligns with the results of our previous prospective study (17.8%), showing that the presence of HPV DNA was associated with SNIP recurrence in 90
