INTRODUCTION: Epigenetic modifications, particularly DNA methylation, play a key role in oropharyngeal squamous cell carcinoma (OPSCC). This is the first study to apply genome-wide methylated DNA sequencing (MeD-seq) to identify differentially methylated regions (DMRs) in OPSCC. By comparing HPV-positive and HPV-negative tonsillar carcinomas, we evaluate MeD-seq's applicability to validate known and identify novel DMRs. These findings may support the identification of potential biomarkers for risk stratification and prognostic assessment in OPSCC.
METHODS: Methylated DNA sequencing (MeD-seq)

BACKGROUND: This study aims to compare the parafoveal retinal microcirculation in children with severe adenotonsillar hypertrophy (ATH) and healthy controls using optical coherence tomography angiography (OCTA).
METHODS: Sixty-six eyes (31 children with ATH and 35 healthy controls) were analyzed. Parafoveal OCTA images (6 × 6 mm) were obtained using a swept-source OCTA device (DRI OCT Triton; Topcon Corporation, Tokyo, Japan). Vessel density (VD) of the superficial capillary plexus (SCP) and deep capillary plexus (DCP) was measured in the central and four parafoveal quadrants. The foveal avas

Circulating T follicular regulatory (cTfr) cells are measured as blood biomarkers after vaccination, in autoimmune disease (e.g., lupus, rheumatoid arthritis) and cancer immunotherapy, yet whether they originate from natural Tregs (yielding nTfr) or germinal center T follicular helper (GC Tfh) cells (yielding iTfr) remains unknown. This distinction is critical for interpreting cTfr expansion as Treg-derived regulation versus Tfh-derived negative feedback. To characterize tonsillar follicular T cell heterogeneity and resolve the origins of cTfr cells we applied single-cell multiomics and bulk T

Trans-cervical ultrasound of the palatine tonsils provides a non-invasive technique to image the adult tonsils and the associated peritonsilar space to investigate intratonsillar and peritonsillar abscesses. The peritonsilar space, where peritonsillar cellulitis and abscesses are located is readily identified sonographically between the palatine tonsil and the superior constrictor muscle, which can be used as a helpful sonographic landmark. An understanding of the sonographic anatomy, technique, landmarks and imaging appearances is essential to correctly perform and interpret ultrasound imagin

Tonsillectomy is one of the most common surgical procedures in pediatric populations, indicated primarily for recurrent tonsillitis and upper airway obstruction. Palatine tonsils represent lymphoepithelial structures crucial for local and systemic immune surveillance, with a key role in lymphocyte maturation and the production of immunoglobulins, particularly secretory IgA. This review summarizes the available evidence on the immunological impact of tonsillectomy, including short- and long-term studies. Data show that tonsillar removal can result in transient changes in immunoglobulin and lymp

OBJECTIVE: Peritonsillar abscess (PTA) after surgical tonsil removal is an infrequent complication. Residual lymphoid tissue after tonsillotomy may increase this risk compared to tonsillectomy. This study aimed to identify cases of PTA following tonsillotomy or tonsillectomy in a pediatric population and compare the incidence and case details.
STUDY DESIGN: Retrospective comparative case series.
SETTING: Tertiary pediatric hospital.
METHODS: A review was conducted of patients ≤18 years of age who underwent tonsillotomy or tonsillectomy from January 2010 to January 2025 and later presented w

CD22 (Siglec-2) is a key regulator of B-cell receptor (BCR) signaling during affinity maturation in the germinal center (GC) by setting the BCR signaling threshold, yet its candidate ligands on B cells and their dynamics during the light zone-dark zone (LZ-DZ) transition remain unclear. Here, we combined liquid chromatography-electrospray ionization mass spectrometry N-glycomics with recombinant CD22 (rCD22) pulldown and cell-surface binding assays in human tonsil B-cell subsets. rCD22 pulldown enriched α2,6-sialylated bisected and non-bisected N-glycan compositions, identifying these as candi

Ageing profoundly impairs vaccine efficacy, leaving older individuals highly susceptible to infectious diseases despite vaccination. Although B cells retain the intrinsic capacity to generate germinal centre (GC) responses in older people, age-associated defects in T cells, dendritic cells and follicular dendritic cells (FDCs) limit the development of high-affinity, class-switched antibody responses. Here, we hypothesised that senescent fibroblasts disrupt B cell activation and expansion, thus contributing to the diminution of the humoral response in ageing. To test this hypothesis, we establi

BACKGROUND: Primary small cell neuroendocrine carcinoma (SCNEC) of the tonsils is extremely rare and is often misdiagnosed as squamous cell carcinoma. Cases coexisting with hypopharyngeal squamous cell carcinoma (HPSCC) have not been reported. The presence of concurrent aortic arch aneurysm further complicates treatment.
CASE DESCRIPTION: A 71-year-old male presented with pharyngeal pain lasting over one month. Imaging and pathological examinations confirmed right tonsillar SCNEC (with approximately 80% Ki-67 positivity) and left HPSCC, along with concurrent aortic arch aneurysm and stage III

Simulated microgravity provides a controlled perturbation for testing how loss of mechanical load affects human tissues. After 24 hours in rotating-wall vessels, neural, cardiac and tonsil organoids developed tissue-specific transcriptional responses that overlapped published spaceflight datasets, accompanied by higher transcriptomic-age estimates in neural and cardiac models, altered cardiac-organoid contraction dynamics and modified tonsil-organoid antiviral gene-expression signatures. These findings establish a multi-organoid framework for studying acute responses to gravitational unloading

BACKGROUND: Brown adipose tissue (BAT) dissipates energy via non-shivering thermogenesis, making it a promising therapeutic target for metabolic diseases. However, effective strategies for directing human stem cells toward brown adipocyte lineage remain to be fully established. Here, as an early commitment modulation study, we investigated whether stage-specific exposure to bone morphogenetic protein 7 (BMP7) during initial induction enhances brown adipocyte-like features and β-adrenergic responsiveness in tonsil-derived mesenchymal stem cells (TMSCs).
METHODS: TMSCs were induced to different

PURPOSE: Delays in time to treatment initiation (TTI) have been associated with inferior outcomes in head and neck cancer, yet prior studies have typically evaluated tumors collectively, assuming uniform time sensitivity across diseases. This study quantified subsite- and stage-specific associations between TTI and overall survival (OS) and derived groups of tumors with similar TTI-mortality associations.
METHODS: We conducted a retrospective cohort study using the National Cancer Database (2010-2017), including adults with head and neck malignancies treated with curative intent. Multivariabl

BACKGROUND: Adenoid hypertrophy (AH) is a common childhood condition frequently associated with multiple comorbidities. However, its specific clinical phenotype and the independent risk factors for key complications, particularly in children requiring surgery, remain inadequately elucidated.
METHODS: This retrospective cohort study included 161 children undergoing surgery for AH. Demographic data, disease severity indicators, comorbidities, and laboratory results were collected. Descriptive statistics, chi-square tests, and multivariate logistic regression models were used for data analysis.

T follicular helper (Tfh) cells provide help to B cells for antibody class switching and affinity maturation and are critical for germinal center (GC) formation. The frequency of Tfh cells has proven to be a valuable marker for predicting durable neutralizing antibody responses; conversely, the expansion of dysfunctional Tfh cells and increased T follicular regulatory (Tfr) cells are associated with suboptimal antibody responses. Given that the pig is becoming an appealing preclinical model for vaccine evaluation, a better understanding of porcine Tfh and Tfr cell populations is required. Nove

Classically, B and T cells are differentiated by their mutually exclusive expression of surface markers such as CD3 and CD20, but it is now established that a few percent of T cells in human blood display also CD20. We analyzed CD3+CD20+ cells in detail by combining single-cell proteogenomics, FACS, and ImageStream. Thereby, we identified CD3+ B cells and found that about 40% of plasmablasts displayed CD3 in both blood and tonsils. In tonsils, where T-B interactions occur, about 80% of follicular helper T (Tfh) cells displayed CD20. In vitro, coactivation of T and B cells induced marker exchan
