BACKGROUND: Hepatic fibrosis (HF) is a key pathological process in the progression of chronic liver diseases to cirrhosis and hepatocellular carcinoma (HCC), accompanied by abnormal remodeling of the intrahepatic vascular network. Evidence suggests that hepatocyte cytokine leukocyte cell-derived chemotaxin 2 (LECT2) and macrophage reprogramming exert crucial effects in angiogenesis. Jiawei Taohe Chengqi Decoction (JTCD) can reduce abnormal angiogenesis in HF, but the specific mechanism remains unclear.
PURPOSE: To clarify whether JTCD attenuates HF by inhibiting LECT2 to modulate macrophage r

ETHNOPHARMACOLOGICAL RELEVANCE: Persicaria capitata, a vital Miao medicine, is traditionally employed to "clear heat and promote diuresis" and "detoxify". In traditional theory, alcoholic liver injury (ALI) is categorized as a "Damp-Heat accumulation" syndrome, often manifesting as damp-heat jaundice. The traditional application of P. capitata in treating jaundice and inflammatory conditions provides a rationale for its hepatoprotective use.
AIM OF THE STUDY: To provide a scientific basis for the traditional use of P. capitata by systematically investigating the protective effects of its wate

Chronic intermittent hypoxia (CIH) is an alternating state of recurrent hypoxia and reoxygenation triggered by disorders such as sleep apnea, leading to systemic oxidative stress and organ damage. In this study, we investigate the protective effect and mechanism of atraric acid (AA), a secondary metabolite produced by lichens, against CIH-induced liver injury. By establishing a CIH mouse model (10 h/d, oxygen cycling 5%-21% for 4 weeks) and an in vitro model of AML-12 cell hypoxia/reoxygenation (H/R), we perform H&E staining, biochemical assays (ALT/AST/MDA/CAT/T-AOC/GSH-Px), qPCR, western blo

Biomarkers with potential for use in clinical diagnosis have been widely studied using biochemical techniques. In particular, endogenous and exogenous metabolites have been targeted in clinical settings. Both types of metabolites are either unmetabolized, metabolized with human enzymes, or metabolized with host-resident bacterial enzymes. In standard clinical chemistry, a specimen must be collected following overnight fasting to minimize the effect of dietary input and the subsequent host and microbial metabolism. Current technology allows the detection of nanomolar concentrations of exogenous

Follicular cholangitis is a rare cause of bile duct inflammation and stenosis, mimicking stricturing malignancies such as cholangiocarcinoma. This condition is considered distinct from follicular cholecystitis, which involves chronic inflammatory and lymphoplasmacytic changes in the gallbladder. We present a case of a woman in her 70s who was incidentally found to have an intrahepatic bile duct stricture in the left lobe of the liver. After surgical management, she was found to have concurrent follicular cholangitis and follicular cholecystitis. This case highlights the similar histopathologic

BACKGROUND: White adipose tissue (WAT) communicates with distal metabolic organs and regulates their functions by secreting adipokines. Subcutaneous WAT (sWAT) is thought to be metabolically protective but prone to dysfunction with aging and obesity. Chronic activation of p53 is commonly observed in dysfunctional WAT at different anatomical locations in metabolic diseases. However, the impact of MDM2 deletion and its associated p53 activation in sWAT on adipokine profile and systemic metabolism is unknown.
METHODS AND RESULTS: To investigate the effects of selective MDM2 deletion and its asso

Non-alcoholic fatty liver disease (NAFLD) is closely associated with gut microbial dysbiosis and metabolic dysfunction, yet effective and sustainable therapeutic options remain limited. Wuqing Decoction (WQT), a multi-herb botanical formula containing Pueraria lobata and other medicinal and edible components, has shown metabolic regulatory potential, but its mechanistic basis in NAFLD remains unclear. Here, we evaluated the preventive and therapeutic effects of WQT in high-fat diet (HFD)-induced NAFLD mice and examined its microbiota-dependent mechanisms. WQT significantly alleviated hepatic s

BACKGROUND: Congenital hepatic fibrosis (CHF) is a rare disease associated with the polycystic kidney gene, and the initial symptoms are often associated with portal hypertension, such as splenomegaly and esophageal varices. While the basic clinical features and pathogenesis of isolated CHF have been thoroughly studied, patients frequently present variable combined phenotypes including autosomal dominant polycystic kidney disease (ADPKD) and Von Meyenburg complex (VMC). The pathogenic mechanisms driving these composite phenotypes require further exploration.
METHODS AND RESULTS: We report a c

Optimizing murine models for studying Metabolic Dysfunction-Associated Steatohepatitis (MASH) and fibrosis is crucial for understanding disease mechanisms and evaluating therapies. In this study, we characterized diet-induced male murine models of MASH and liver fibrosis using an AI-digital pathology (DP) pipeline for zone-specific analysis and spatial (co)-localization of key MASH features within the liver microarchitecture, providing insights beyond standard histology. Model characterization included an integrative omics-based approach, standard blood-chemistry, and traditional histology. AI

Hepatocellular carcinoma (HCC) is a common and lethal malignancy, with an increasing incidence globally, which is partially attributed to the rising prevalence of metabolic dysfunction-associated steatotic liver disease (MASLD). Complex metabolic rearrangements occur in MASLD-related HCC (MASLD-HCC) to promote the initiation and development of cancer. Nevertheless, comprehensive understanding of the metabolic transformations during the progression from MASLD to HCC remains limited. In this review, we introduce recent advances characterizing the metabolic changes in MASLD-HCC, thereby offering

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease frequently coexists with type 2 diabetes mellitus (T2DM). Because the gold standard for assessing insulin resistance (IR) is difficult to implement in primary care settings, this cross-sectional study aimed to compare the associations and discriminatory ability of 12 surrogate IR indexes for prevalent T2DM among community-dwelling older adults with metabolic dysfunction-associated fatty liver disease.
METHODS: This study was based on a health examination cohort from a community in northwestern China and included 2641 eligibl

INTRODUCTION: Hepatocellular carcinoma is the sixth most common malignancy worldwide, with immune checkpoint inhibitors transforming treatment paradigms despite modest response rates of approximately 30%. Emotional distress, encompassing depression and anxiety symptoms, affects 30-50% of cancer patients and may influence immunotherapy efficacy through immune system modulation. Recent studies in lung cancer, gastric cancer and melanoma demonstrate associations between pretreatment emotional distress and worse immunotherapy outcomes. However, this relationship remains unexplored in patients with

Amodiaquine (AQ), an antimalarial drug, is metabolized by cytochrome P450 3A4 (CYP3A4) to quinoneimine forms, which are proposed to cause liver injury; however, the underlying mechanism remains unclear. Ferroptosis, a type of regulated cell death driven by iron-dependent lipid peroxidation, has been implicated in certain cases of drug-induced liver injury. Arylacetamide deacetylase (AADAC), a drug-metabolizing hydrolase, regulates hepatic iron homeostasis through non-enzymatic mechanisms and may protect against ferroptosis-associated drug-induced liver injury. This study aimed to elucidate the

BACKGROUND: The worldwide burden of metabolic dysfunction-associated steatotic liver disease (MASLD) is rising sharply, powered by high prevalence of obesity and its associated metabolic insults. Though urgent, definitive treatment is not yet available. Empagliflozin, a sodium glucose co-transporter inhibitor has been suggested in both human and animal studies to have diverse beneficial effects in MASLD by anti-inflammation and anti-oxidative stress effects, but many of the underlying mechanisms remain unexplained. The main aim of this systematic review and meta-analysis was to evaluate the ef

This study investigates the genotoxicity and developmental toxicity of phenol-branched fatty acid (PhBC-FA) monomer and its derivatives, PhBC-FA methyl ester (PhBC-FAME) and PhBC-FA-amide (PhBC-FAAM), which have been used in the development of antimicrobial coatings. Given the potential risks of phenol (Ph) and the need for safer alternatives to traditional antimicrobial products, in silico toxicity tools, in vivo experiments for developmental toxicity, and DNA damage in chicken embryos were used. In the in silico simulation, among three test compounds, PhBC-FAME was identified as the most tox

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a progressive hepatic metabolic disorder characterized by excessive fat accumulation in hepatocytes. MASLD can advance to metabolic dysfunction-associated steatohepatitis (MASH). These conditions often lead to hepatic fibrosis, cirrhosis and hepatocellular carcinoma (HCC) if not timely diagnosed and managed; thus, molecular phenotyping of MASLD and MASH conditions is highly desirable. The analyses were performed to identify distinct, differentially expressed genes, particularly noncoding ones, in MASLD and

Candidiasis caused by Candida albicans represents a major opportunistic infection, particularly in hospitalized individuals, with high morbidity and mortality rates. Experimental models that reproduce natural routes of exposure and allow integrated analyses of host responses remain limited. In this study, we established and characterized an immersion-based infection model of C. albicans in adult zebrafish (Danio rerio) to investigate systemic, behavioral, immunological, oxidative, and histopathological outcomes. Adult zebrafish were exposed by immersion to different fungal concentrations (1 ×
