Blocking ELISA Protocol for Salmonella Infection.

Salmonella is an important zoonotic pathogen that is widely distributed in livestock, poultry, and other animals. Salmonella contamination not only affects the healthy development of the livestock industry but also poses a serious threat to public health. Therefore, establishing detection methods capable of simultaneously identifying different Salmonella serotypes is crucial. Among current detection methods for Salmonella, immunological detection methods using MAbs offer high throughput, shorter processing times, and enhanced specificity, making them useful for rapid Salmonella detection. The

Measure the Biofilm Formation of Salmonella Using Crystal Violet Assay.

Salmonella can form biofilms, which are structured microbial communities composed of cells that adhere irreversibly to surfaces or to other cells. The formation of Salmonella biofilms may enhance the bacterial resistance to drugs, environmental stress, and the host immune system. Crystal violet staining is one of the most classical and widely used methods for measuring biofilms in laboratories. Its principle is primarily based on the binding of crystal violet to various components within the biofilm. Here, we describe the crystal violet assay protocol to measure the formation of Salmonella bio

Construction and characterization of a chimeric Salmonella phage P6-UPF2 via homologous replacement of the phage P6 tail spike protein Gp9.

With over 2,600 Salmonella serotypes identified, no single natural or existing bacteriophage can cover this diversity, making customized phages a promising strategy for precise control. In this study, the previously characterized phage P6 served as the recipient, and the broad-spectrum phage UPF_BP1 (94.91% nucleotide identity with P6) served as the donor. Two fragments (b1 and b2) from the donor's tail spike protein gene were individually recombined into the Gp9 locus of P6, generating two chimeric phages: P6-UPF1 and P6-UPF2. Host range analysis showed that P6-UPF1 exhibited no alteration in

Dual activities of lysis and capsule degradation enable the bacteriophage protein Lys33795 to effectively disrupt Streptococcus suis biofilms.

Streptococcus suis (S. suis) is an encapsulated zoonotic pathogen that can form recalcitrant biofilms. Bacteriophage-derived endolysins and depolymerases represent promising antibiofilm agents, but currently, no report has described protein which displays both of lytic and capsule degradation activities simultaneously. Here, we mined the porcine gut virome and identified Lys33795, a dual-activity antibacterial protein that degrades both capsule and peptidoglycan of S. suis. Lys33795 consists of an N-terminal GH25_muramidase domain and a C-terminal cell wall binding domain (CBD). However, trans

Discovery of Two 17β-Estradiol Metabolic Microbes and Their Metabolic Enzyme from Cecal Contents of Hy-Line Brown Laying Hens.

17β-Estradiol (E2), a reproductive hormone closely associated with the decline in egg production of laying hens, has attracted considerable attention. Although microbial feed additives have significantly alleviated this decline, the direct interactions between the host microbe and E2 remain poorly understood. In the present study, two E2-metabolic strains (Brevibacillus borstelensis NRRL NRS-818 and Acinetobacter radioresistans) were isolated through gradient pressure acclimatization with E2. Further RNA-Seq analysis of these two strains grown in medium with or without E2 identified 14 upregul

Mycobacterium tuberculosis Rv3248c Inhibits Type I Interferon Response and Promotes Intracellular Survival by Mediating Ubiquitin-Proteasome-Dependent Degradation of TBK1.

Mycobacterium tuberculosis (M. tb) is a leading cause of infectious disease mortality worldwide, and M. tb subverts host immunity to persist intracellularly. Type I interferon (IFN-I) modulates host defense during M. tb infection, but the mechanisms controlling their induction remain incompletely defined. In this research, a previously uncharacterized mycobacterial factor, M. tb Rv3248c, which regulates IFN-I response, was identified using pull-down and mass spectrometry. Overexpression of Rv3248c suppresses IFN-β expression in macrophages at both the transcriptional and protein levels. In add

Collateral Sensitivity in Bacteria: Phenomenon, Mechanisms, Modulators and Therapeutic Prospects.

The rapid proliferation and infection of multidrug-resistant bacteria pose a significant threat to global public health. The gradual increase in drug resistance, primarily driven by the inappropriate use of antibiotics, underscores the urgent need for developing novel therapeutic approaches for drug-resistant bacterial infections. Collateral sensitivity (CS) describes the phenomenon whereby bacteria resistance to a specific antibiotic increases susceptibility to other antibiotics. In some cases of antibiotic resistance, CS can provide rapid and effective guidance for clinical drug use by detec

Identification and whole-genome sequencing of blood-brain barrier-penetrating Riemerella anatipestifer isolated from chickens and ducks.

Riemerella anatipestifer (RA) is a major cause of bacterial meningitis, which poses a serious threat to the poultry industry. To identify RA strains with a strong ability to cross the blood-brain barrier (BBB) and analyze their genomic characteristics, a co-infection model was established using multiple RA strains with different serotypes in this study. We found that during co-infection with multiple RA strains, certain RA strains rapidly penetrated the BBB and were recovered from brain tissue as early as 0.25 h post-infection. The pioneer strains were selected to further evaluate their BBB-pe

Epidemiologic determinants of antimicrobial resistance in feline urinary Escherichia coli in the United States (2018-2024).

Lower urinary tract infections are less common in cats than in dogs, with Escherichia coli being the predominant bacterial pathogen responsible for these infections. Although antimicrobial therapy is used for optimal treatment, repeated or prolonged exposure may increase the risk of antimicrobial resistance (AMR). This study examined risk factors for AMR in urinary E. coli isolates from cats across the United States, focusing on prior positive cultures and other host, socioeconomic, and geographic factors, using multilevel logistic regression models, accounting for clustering at the patient, c

Lactobacillus derived small RNAs protect against Klebsiella pneumoniae infection.

Carbapenem-resistant Klebsiella pneumoniae (CRKP) is a major multidrug-resistant pathogen with limited treatment options. Plant-derived natural products have broad biological activities, but their protective effects against extraintestinal multidrug-resistant bacterial infections remain poorly understood. Herein, we show that selected plant-derived natural products protect mice against CRKP pulmonary infection by remodeling the gut microbiota. Fecal microbiota transplantation and microbiota-depletion experiments demonstrated that this protection was microbiota dependent, with Lactobacillus eme

Monoclonal Antibodies Targeting Bacillus Calmette-Guérin (BCG) Outer Membrane Proteins Confer Protection Against Tuberculosis.

Tuberculosis (TB), caused by the Mycobacterium tuberculosis complex (MTBC), is a persistent global health crisis, remaining the world's leading infectious killer. There were abundant outer membrane proteins (OMPs) distributed in the bacterial cell wall, while fewer proteins were identified and characterized as antigens that could induce protective antibodies against TB. The OMPs of BCG were extracted with the detergent N-octyl-β-D-glucopyranoside, and the extracted proteins were used to immunize mice to generate an antibody library. Subsequently, the library underwent HAT/HT screening, and fiv

Experimental and Monte Carlo Insights into the Biofilm Control of Klebsiella pneumoniae Using Green-Synthesized Ag/Ni(OH)2/Clove Nanocomposites.

One of the most significant challenges with disease control in humans and animals is biofilm formation by Klebsiella pneumoniae (K. pneumoniae). The current study investigated the prevalence of K. pneumoniae in fecal and milk samples from cattle and humans, along with their antimicrobial susceptibility patterns and biofilm formation, using conventional methods. In addition, the efficacy of Ag/Ni(OH)2/Clove and Ag/Ni(OH)2/C nanocomposites was comparatively evaluated to combat resistance and biofilm-associated traits. K. pneumoniae was recovered from 48.0%, 38.0% and 43.3% of the fecal, milk and

Cat scratch disease with fever and lymphadenopathy without definite cat exposure diagnosed via tissue metagenomic next-generation sequencing technology: a case report.

BACKGROUND: Cat scratch disease (CSD), a disease caused by Bartonella henselae (B. henselae), is often associated with exposure to cats. However, in rare cases, a history of cat exposure is absent, and thus achieving a definitive diagnosis may be challenging. Metagenomic next-generation sequencing (mNGS) is a relatively new technology that can analyze pathogens without introducing inherent biases. In this report, we describe a case of CSD diagnosed via mNGS, which supports the use of mNGS in identifying rare types of pathogenic infection. CASE DESCRIPTION: A 54-year-old male exhibited fever a

Case Report: Atypical cat-scratch disease presenting as referred shoulder pain and deep lymphadenopathy in a pediatric patient.

BACKGROUND: Cat-scratch disease (CSD) is a zoonotic infection attributable to Bartonella henselae. Typical clinical manifestations include regional lymphadenopathy accompanied by primary cutaneous lesions that develop following exposure to cats. Although most patients present with typical regional lymphadenopathy, a subset of patients may develop atypical manifestations, which can complicate diagnosis. Owing to the nonspecific clinical features, CSD is prone to misdiagnosis or diagnostic omission. This report describes a pediatric case of atypical cat-scratch disease characterized by initial s

Case Report: Dasatinib-associated chylothorax and occult Bartonella henselae infection in pediatric leukemia: a case report and systematic review.

Dasatinib is a second-generation tyrosine kinase inhibitor (TKI) used in Philadelphia chromosome-positive (Ph+) acute lymphoblastic leukemia (ALL), and chylothorax is one of its rare but serious lymphatic complications. We encountered a 14-year-old boy with Ph + acute lymphoblastic leukemia (ALL) who developed bilateral chylothorax while on dasatinib, with concurrent B. henselae infection (cat-scratch disease) confirmed in the pleural fluid. Prompted by this index case, we performed a systematic review of the literature to explore the potential association between dasatinib-associated chylotho

The evolution of Brucella as a model intracellular pathogen.

Brucella species are intracellular bacterial pathogens that cause brucellosis, one of the most common zoonoses worldwide. This review traces how Brucella, first discovered in the 19th century through investigations of human and bovine disease, became a modern experimental model system. Work on Brucella has illuminated how intracellular bacteria evade and adapt to host defenses and how they remodel host cell biology to establish a replicative niche. It has also yielded insights that extend beyond pathogenesis into bacterial cell envelope architecture and development. Though long regarded as a s

The fecal microbiota of healthy dogs fed extruded and raw meat-based diets.

Conventional dog diets, such as extruded diets, remain common. The popularity of unconventional options, particularly raw diets, has increased over time and may differentially influence the gut microbiota. Raw diets inherently harbor higher bacterial loads, including potentially pathogenic microorganisms, raising concerns from a One Health perspective due to the close interaction among pets, humans, and the environment. The study aims were to characterize the nutritional profiles of extruded and raw diets, compare the fecal microbial composition of dogs fed extruded diets with that of dogs fed
