Establishment of a blocking ELISA using a newly generated monoclonal antibody against Salmonella flagellin for detecting antibodies in infected chickens.

BACKGROUND: Salmonella is an important zoonotic pathogen that is widely distributed in every link between livestock and poultry production. Salmonella contamination not only affects the healthy development of the livestock industry but also poses a serious threat to public health, as it can infect humans through farming, processing, and consumption of meat and egg products. Therefore, establishing detection methods capable of simultaneously identifying different Salmonella serotypes is crucial. RESULTS: This study focused on using Salmonella flagellin as an immunogen to screen and prepare mon

Targeting bacterial phosphotransferase system to prevent the dissemination of antibiotic resistance genes.

Antimicrobial resistance (AMR) poses a significant challenge to public health and human security, with plasmid-mediated horizontal gene transfer (HGT) being a primary driver for its dissemination. Here, we identify indole analogs containing electron-withdrawing groups as a new category of HGT inhibitors. Using indole-3-acetic acid (IAA) as a representative scaffold, we demonstrate that IAA targets glycolytic enolase to deplete phosphoenolpyruvate (PEP) in donor bacteria; this suppresses phosphotransferase system (PTS) activity, blocks PtsI phosphorylation, reduces cAMP synthesis and prevents c

Effects of the ecpA gene on the biological characteristics and pathogenicity of avian pathogenic Escherichia coli strain FJLY68.

Avian pathogenic Escherichia coli (APEC) is a major cause of colibacillosis in poultry, yet the role of the ecpA gene, which encodes the major structural subunit of the Escherichia coli common pilus (ECP), remains incompletely defined in APEC pathogenesis. To investigate the role of ecpA in the biological characteristics and pathogenicity of Avian Pathogenic Escherichia coli (APEC) strain FJLY68, an ecpA deletion mutant (ΔecpA) and its corresponding complemented strain (CΔecpA) were constructed using the CRISPR/Cas9 system and verified by PCR and Sanger sequencing. Phenotypic analyses revealed

Silver nanoparticle hydrogen peroxide composite mitigates resistant Escherichia coli dissemination driven by poultry waste biosecurity failures.

This two-phase study evaluated biosecurity vulnerabilities across 100 commercial poultry farms via a field survey and an in vitro assessment of hydrogen peroxide (H2O2) versus a silver nanoparticle-hydrogen peroxide (AgNPs- H2O2) composite. Survey metrics revealed critical baseline deficiencies: 80% of farmers lacked antimicrobial resistance (AMR) awareness, 90% practiced no litter treatment, and 70% of untreated waste was sold directly to aquaculture. Molecular analysis of 192 litter samples verified a 72.4% E. coli prevalence, with Multiple Antibiotic Resistance (MAR) indices peaking at 0.90

SH3PXD2B is the critical host factor promoting the entry of mycobacteria.

Bovine tuberculosis (bTB), caused by Mycobacterium bovis, causes major economic losses in the global livestock industry and threatens public health. Macrophages are essential for pathogen clearance, yet serve as the primary intracellular niche for mycobacterial survival. These characteristics emphasize the importance of identifying host factors that regulate macrophage functions during infection. To systematically uncover such factors, we constructed a genome-wide CRISPR-KO library in Bomac cells (the bovine macrophage cell line) and performed iterative infection screening under high selective

Current and emerging therapeutic strategies targeting adherent-invasive Escherichia coli in Crohn's disease: a review.

Considerable evidence has accumulated over the past two decades demonstrating that Escherichia coli, specifically adherent-invasive E. coli, contributes to the pathogenesis of Crohn's disease. Adherent-invasive E. coli can adhere to and invade intestinal epithelial cells, survive, and replicate within macrophages, thereby enabling a key mechanism that induces chronic inflammation. Despite extensive knowledge of the molecular interactions between adherent-invasive E. coli and the host from these studies, translating this knowledge into targeted therapy remains limited. In this review, we summari

The two-component system histidine kinase LiaS contributes to the stress resistance and virulence of zoonotic Listeria monocytogenes.

LiaS is a histidine kinase receptor in two-component systems that senses environmental stress signals. This study investigated the roles of LiaS in Listeria monocytogenes stress resistance and virulence. Phenotypic assays showed that deletion of liaS (ΔliaS) resulted in significantly impaired growth under acidic, alkaline, and osmotic stresses, as well as reduced survival under strong acidic conditions. These defects were partially restored in the complemented strain (CΔliaS). The growth defects were also observed in the ΔliaS under Cu2+ or H2O2 exposure. Furthermore, the ΔliaS strain exhibite

Chelerythrine potentiates meropenem activity against NDM-producing carbapenem-resistant Enterobacteriaceae.

INTRODUCTION: NDM-driven carbapenem-resistant Enterobacteriaceae (CRE) are spreading globally, posing a serious One Health threat that extends to veterinary medicine through transmission among food-producing animals, companion animals, and the farm-to-food continuum. Antibiotic adjuvants that restore or potentiate the activity of existing antibiotics represent a promising strategy to address this challenge. METHODS: In this study, we evaluated the ability of the natural benzophenanthridine alkaloid chelerythrine to potentiate meropenem activity against NDM-producing Enterobacteriaceae. Antiba

Longitudinal study of a rodent invasion reveals shifts in host pathogen interactions.

Invasive species can disrupt native epidemiological processes, potentially leading to the emergence of zoonotic pathogens. The relatively recent appearance and spread of the bank vole (Clethrionomys glareolus) in Ireland provides a unique model system to study these phenomena. The depauperate small mammal community in Ireland, combined with baseline data, allows us to identify the invasion gradient of C. glareolus and its effects on rodent-borne pathogens over time. A total of 498 C. glareolus and 584 Apodemus sylvaticus were sampled, via removal trapping, across nine sites in Ireland in 2016

Pangenomic and genomic plasticity analyses of the genus Rickettsia.

The Rickettsia genus comprises obligate intracellular bacteria transmitted by arthropods and responsible for clinically relevant zoonoses, rickettsioses, such as spotted fever and typhus. The difficulty of cultivating these bacteria in vitro reinforces the importance of in silico approaches, such as pangenomic and genomic plasticity analyses. This study analyzed 165 genomes from 31 Rickettsia species available in the REFSEQ (NCBI) database. Tools such as Orthofinder, ANIclustermap, Gegenees, and Mauve were used to classify genes into core, shared, and singletons, assess genomic similarity, and

The lipid A acylation pattern of Coxiella burnetii prevents detection and clearance by the non-canonical inflammasome in primary murine macrophages.

C. burnetii is a Gram-negative, obligate intracellular bacterium and the causative agent of Q fever. The disease is either asymptomatic or manifests as a mild flu-like illness, but pneumonia or hepatitis might also occur. In most cases, the infection is self-limiting and the pathogen is cleared. In a small percentage of patients, the host immune system fails to eliminate the pathogen, potentially allowing the development of chronic Q fever months or even years after primary infection. The elimination of the bacteria, and thereby prevention of disease onset, would require an inflammatory respon

Accelerating Campylobacter zoonosis in the Anthropocene.

Intensive poultry farming has transformed global bird populations, concentrating billions of chickens into dense industrial systems that fundamentally reshape ecological space. As birds are important pathogen reservoirs, these changes can have important consequences for the spread of zoonoses including Campylobacter jejuni-the leading cause of bacterial gastroenteritis. Analyzing 2,747 genomes from chickens and wild birds, we show that poultry intensification has eroded historic host-strain associations and created a new and expanding ecological niche. Phylogenetic reconstructions reveal an es

Persistent Intestinal Colonisation and Systemic Immune Activation Following Experimental Campylobacter jejuni Infection in Broiler Chickens.

Campylobacteriosis is one of the leading foodborne bacterial illnesses worldwide, with poultry meat representing its principal source of human infection. Broiler chickens are a key reservoir for Campylobacter jejuni, where the bacterium persists in the gastrointestinal tract without disease. Understanding this host-pathogen interaction is essential to clarify mechanisms of persistence and food safety implications. This study investigated persistent C. jejuni colonisation and systemic immune responses in experimentally infected broiler chickens. Following oral challenge with the virulent ST-403

The Co-Evolutionary Arms Race Between Salmonella and the NLRC4 Inflammasome: Immune Recognition and Evasion Strategies.

Salmonella is a globally significant foodborne intracellular pathogen, and invasive salmonellosis poses a major global public health threat. The NLR family CARD-containing protein 4 (NLRC4) inflammasome, a pivotal cytosolic innate immune sensor, specifically recognizes Salmonella flagellin and type III secretion system (T3SS) components via the NAIP (NLR family apoptosis inhibitory protein) family. Upon activation, it triggers pyroptosis, pro-inflammatory cytokine release, and infected intestinal epithelial cell extrusion, serving as a central pathway for host defense against Salmonella coloni
