Optimization of a comprehensive metabolomic analysis system for characterizing metabolic alterations in the cauda epididymis of mature and juvenile mice.

The cauda epididymis protects and stores mature sperm in mammals. Recently, comprehensive transcriptomic and proteomic analyses have been conducted to understand its molecular functions; however, fundamental information on its metabolites has not been reported. In this study, we optimized a system for the comprehensive metabolic analysis of the cauda epididymis in mature and juvenile mice. This system identified 116 and 92 metabolites in mature and juvenile mice, respectively. Comparative analysis revealed that 44 and 13 metabolites were upregulated and downregulated, respectively, in the caud

The effect of stromal vascular fraction cells derived from epididymal and inguinal adipose tissues on in vitro spermatogenesis.

Infertility affects 10-20% of sexually active couples and male-related factors contribute to 30-50% of all cases of infertility. The development of three-dimensional in vitro approaches to promote the differentiation of spermatogonial stem cells (SSCs) into functional spermatozoa is an essential step for the treatment of male infertility. Stromal vascular fraction (SVF) cells derived from adipose tissue are known to have a regenerative effect on spermatogenesis and testicular regeneration after testicular damage. The study assessed the effect of SVF cells from epididymal and inguinal adipose t

Exposure to Chlorpyrifos Influences Epididymal Development in Pubescent Rats.

The epididymis is responsible for sperm maturation, but is sensitive to the action of external agents, especially during the peripubertal period. Chlorpyrifos is an organophosphate class pesticide to which humans are constantly exposed. Currently, there are few studies that evaluate the effects of this insecticide on the epididymis; therefore, the objective of the present study was to assess the effects of chlorpyrifos on the postnatal development of the epididymis during the peripubertal period. Thirty Wistar rats were divided into three groups: Control, C5, and C15. The control group receive

A novel inducible Cre mouse model for genetic manipulation in nonciliated cells of efferent ductules†.

The efferent ductules (ED) function as essential conduits for spermatozoa transport from the rete testis to the epididymis. The nonciliated and ciliated cells within the efferent ductal epithelium are responsible for fluid reabsorption and stirring the luminal fluid to prevent sperm agglutination, respectively. Dysfunction in either cell type can result in obstructive azoospermia. To systematically investigate the molecular mechanisms underlying efferent ductal development and function, we successively developed two novel knock-in mouse models via CRISPR/Cas9-mediated insertion of Cre-P2A or C

The role of extracellular vesicles in animal reproduction.

Infertility affects approximately one in six couples worldwide and remains a multifactorial condition influenced by genetic, physiological, and environmental factors. Despite remarkable advances in assisted reproductive technologies (ART), current therapeutic approaches often fail to address the underlying molecular mechanisms contributing to infertility. In recent years, extracellular vesicles (EVs) have emerged as key mediators of intercellular communication, capable of transferring bioactive molecules such as proteins, lipids, and nucleic acids between cells within the reproductive system.

Epididymal transcriptional networks underlying sperm motility differences in Chinese indigenous chicken breeds.

This study characterized phenotypic distinctions between high- and low-motility sperm from 40-week-old Yufen1 roosters and the alteration in epididymal transcriptional networks underlighting sperm motility. Temperature challenge tests (4 °C and 37 °C) revealed that high-viability sperm maintained significantly better survival rates at both temperatures compared to low-viability samples, and high-motility sperm consistently exhibited lower morphological abnormality rates across temperature conditions (P < 0.05), which was further confirmed by extended fertilization windows (P < 0.05) and produc

Potential Proteins Associated with Canine Epididymal Sperm Motility.

The maturation and motility of epididymal sperm (ES) cells are largely driven by changes in protein expression. This study aimed to analyze the proteomic profile of canine (Canis lupus familiaris) ES across groups characterized by different progressive motility (PMOT) values to identify motility-related sperm proteins (MRSPs). ES were obtained from the epididymal semen of 19 dogs. The motility and movement parameters of ejaculated sperm (ES) were evaluated using computer-assisted semen analysis (CASA). Samples were classified into two groups: good sperm motility (GSM), defined as PMOT% ≥ 55%,

Investigation of endocannabinoid 2-AG and its hydrolases for regulation on sperm motility and acrosome reaction in Kunming mice.

CONTEXT: A key endocannabinoid, 2-arachidonoyl glycerol (2-AG), regulates sperm hyperactivation in humans via alpha/beta hydrolase domain-containing protein 2 (ABHD2)-mediated CatSper calcium channel activation. However, the role of ABHD2 in mice is unclear, whereas monoacylglycerol lipase (MAGL), ABHD-containing protein 6 (ABHD6), and ABHD-containing protein 12 (ABHD12) modulate 2-AG in the mouse central nervous system (CNS). AIMS: This study explored the impact of 2-AG on mouse sperm motility, its regulatory mechanisms, and the involvement of MAGL, ABHD6, and ABHD12. METHODS: 2-AG concentr

Morphologic diversity of the epididymis in orchiectomy specimens: a multi-institutional study.

The epididymis frequently exhibits a broad spectrum of non-neoplastic epithelial and stromal alterations that may mimic neoplastic or obstructive processes in orchiectomy specimens. Existing data are mostly derived from single-institution series. This multi-institutional study aimed to provide a comprehensive, contemporary, multi-institutional analysis of the prevalence, spectrum, and clinicopathological associations of epididymal morphological variations in a large orchiectomy cohort. This retrospective study included 1,528 orchiectomy specimens from multiple academic centers. All hematoxylin

A Review on the Effects of Atrazine on Male Rat Reproductive System Cytoarchitecture, Steroidogenesis and Oxidative Pathway.

Atrazine (2-chloro-4-ethylamino-6-isopropylamino-s-triazine) is an active component of herbicides used worldwide in agriculture. Atrazine is an endocrine disruptor, capable of interfering with the delicate hormonal balance necessary for normal reproductive function. Despite numerous studies on atrazine's impact on male reproduction, significant gaps remain, particularly concerning the male genital tract beyond the testes. Therefore, we reviewed the effects of atrazine on the entire male reproductive system to understand the complexity of its actions. Adverse effects of atrazine on the morphoph

Paternal immune activation-induced alteration of 28S rRNA-derived small RNAs in sperm reprograms offspring phenotypes.

Parental environmental exposures can induce transgenerational effects through epigenetic modifications in germ cells. Although paternal immune activation is implicated in transgenerational metabolic and neuropsychiatric disorders, the germline-encoded molecular vectors mediating this inheritance remain poorly understood. Here, we demonstrated that lipopolysaccharide-induced immune activation dynamically upregulated the abundance of 28S ribosomal RNA-derived small RNAs (28S-rsRNAs) in mouse sperm in a time-dependent manner. Furthermore, epididymal sperm maturation exhibited heightened susceptib

Activation of TRPM8 promotes K+ secretion in rat epididymal epithelium.

The epididymis establishes a unique hyper-potassium luminal microenvironment essential for sperm maturation and storage, which is largely orchestrated by epididymal epithelial ion transport. Although the transient receptor potential melastatin 8 (TRPM8) is broadly expressed across various organ systems, its physiological role in male reproduction has remained largely unexplored. This study demonstrated that TRPM8 was predominantly expressed in rat epididymal epithelial cells. Activation of TRPM8 by either the exogenous agonist WS-12 or the endogenous hormone testosterone triggered a decrease i

Engineered bacterial S-layer enhanced high electron mobility transistor sensors for ultrasensitive detection of tumor antigen.

Ultrasensitive and specific detection of low-abundance tumor biomarkers remains a major challenge for early and minimally invasive cancer diagnosis. Here, we present a high-performance biosensing platform that integrates a genetically engineered bacterial S-layer with an AlGaN/GaN high-electron-mobility transistor (HEMT) sensor for label-free detection of tumor antigens. As a proof-of-concept, the ovarian cancer antigen human epididymis protein 4 (HE4) was selected. Specifically, the S-layer protein rSbpA was fused with HE4-specific nanobody 1G8 to construct a bifunctional membrane capable of

Control of offspring sex ratio by injection of Toll-like Receptor 7/8 agonist Resiquimod into father mice†.

Sex control technologies hold great promise for enhancing animal production efficiency. X chromosome-linked toll-like receptor 7/8 (TLR7/8) genes are specifically expressed in mammalian X sperm but not Y sperm. In vitro treatment with the TLR7/8 ligand Resiquimod (R848) can separate X and Y sperm by selectively reducing X sperm motility via inhibition of energy production. Fertilization using such treated sperm leads to sex-skewed embryos or offspring. However, whether R848 can be used in vivo to control offspring sex remains unclear. This study tested this hypothesis. We found that the X/Y sp

Morphological and proteomic characterization of sperm in a beluga whale (Delphinapterus leucas):a case report.

The study of cetacean reproductive physiology is important for their conservation; however, limited information is available on the reproductive characteristics of these species. The present study aims to characterize beluga whale (Delphinapterus leucas) sperm by analyzing their morphology, dimensions and shape, and proteome. Epididymal fluid samples (from each epididymis) were collected from a beluga whale housed in an aquarium that died (~ 60 years old). The results indicated that, in most cases, the sperm head has an ovoid shape with a slightly tapered anterior end. However, significant div

The Functional Epididymal Amyloid Cystatin-Related Epididymal Spermatogenic (CRES) is a Component of the Mammalian Brain Extracellular Matrix.

CRES is the defining member of a reproductive subgroup of family 2 cystatins of cysteine protease inhibitors. We previously showed that CRES and other subgroup members are part of a highly plastic amyloid-containing extracellular matrix (ECM) with host defense functions in the mouse epididymal lumen. Based on parallels between the epididymis and the brain, we hypothesized that CRES and CRES amyloids might also function within the brain including the ECM. Here we show that CRES is produced by hippocampal neurons and astrocytes in the male and female mouse and human brain. Further, approximately
