Prophylactic Antibiotic Use in the Setting of Non-Operative, Low-Velocity Gunshot Wounds.

The management of low-velocity gunshot-induced fractures (LGF), particularly regarding prophylactic antibiotic use, is widely debated. Recent studies have suggested no significant difference in infection rates between patients receiving prophylactic antibiotics for LGF and those who do not. This retrospective study compared infection rates between patients receiving prophylactic antibiotics and those who did not for non-operative, LGF between 2015 and 2020. Data were analyzed for variables including demographics, injury characteristics, and infection within 30 days. Of the 74 patients included

Dual-functional quaternized chitosan-modified carbon dots within a polymeric fiber-gel scaffold for infected wound healing.

While polysaccharide-based biomaterials show great promise in wound management, endowing them with potent, antibiotic-free antibacterial and immunomodulatory properties remains a profound challenge. Herein, we report the functionalization of aspirin-derived carbon dots with a bioactive carbohydrate derivative, quaternized chitosan, via a one-step hydrothermal synthesis, yielding a novel dual-functional macromolecular conjugate (CS-ACD). Leveraging the strong polycationic properties of the modified carbohydrate, CS-ACD achieves a remarkable 97.3% sterilization rate against E. coli through physi

The contribution of LepA to virulence and host cell invasion in Salmonella Typhimurium.

BACKGROUND: LepA is one of the most conserved proteins and is found in nearly all bacteria as well as in eukaryotes. Mutation of lepA increases antibiotic tolerance in Escherichia coli, Salmonella, and rifampicin-resistant clinical mycobacterial isolates. However, the physiological function and its role in virulence of LepA remain elusive, particularly considering that LepA is non-essential under laboratory conditions. METHODS: We investigated the lepA gene of Salmonella Typhimurium for its role in bacterial invasion of host cells. Invasion assays using lepA mutants were performed in macropha

Biotics-based modulation of gastric precancerous states: microbial, metabolic, and epithelial mechanisms.

Gastric precancerous states are important intermediate stages in gastric carcinogenesis, yet strategies that directly modify their persistent biological drivers remain limited. Current management mainly includes Helicobacter pylori eradication, endoscopic surveillance, and mucosal protective strategies. Biotics, including probiotics, synbiotics, postbiotics, and next-generation microbial products, have been investigated as potential strategies to modulate persistent microbial, metabolic, inflammatory, and barrier alterations. However, existing evidence derives from studies of H. pylori-associa

Genomic diversity, host associations and ecology of Campylobacter jejuni recovered from wild birds in Michigan crop production systems.

Campylobacter jejuni is a leading cause of foodborne illness worldwide, yet infection sources are not always clear. We used whole-genome sequencing to compare C. jejuni from bird faeces collected in cherry orchards in Michigan to 214 clinical isolates from Michigan patients and 1,286 published bird-derived C. jejuni genomes. The bird isolates from Michigan possess class D β-lactamases and multiple virulence genes and belong to seven sequence types (STs), including three novel STs. Two isolates obtained from different bird species differed by only a single core-genome SNP despite being collecte

Antimicrobial susceptibility of E. coli isolated from dairy cow milk in Egypt and antibacterial activity of selected nanomaterials against MDR strains.

Escherichia coli (E. coli) is a significant livestock and foodborne pathogen. This study was designed to isolate E. coli from raw cow milk, evaluate its antimicrobial susceptibility, and investigate the antimicrobial activity of silver nanoparticles (nAg), zinc oxide nanoparticles (nZnO), and Moringa nano-emulsion (MNE) against multidrug resistant (MDR) strains. A total of 146 individual milk samples were obtained from dairy cows in Egypt. E. coli were isolated by standard culture methods and pathotyped using molecular techniques. The antibiotic resistance of the isolates was determined, and t

Genotypic and phylogenetic characterization of Salmonella Minnesota ST548 strains with potential risk to public health.

Salmonella Minnesota ST548 has emerged as an important serovar in the poultry industry and has been detected in food products. Five nonclonal S. Minnesota isolates from poultry in southeastern Brazil were characterized using whole-genome sequencing (WGS), and 263 database sequences were analyzed for virulence genes, antimicrobial resistance genes, and phylogenetic relationships. Among the analyzed sequences, 191 multidrug-resistant (MDR) strains carried antimicrobial resistance genes (ARGs) conferring resistance to aminoglycosides (aac(6')-Iaa, aph(3')-Ia and aadA1), quinolones (qnrB19), tetra

Impact of Charge Distribution on the Membrane Binding and Antimicrobial Activity of Cationic Heptapeptides.

Antimicrobial peptides (AMPs) hold immense potential in combating community-acquired and MDR infections. The therapeutic efficiency of AMPs depends on their various physicochemical properties. We investigated how the hydrophobic-hydrophilic landscape of tryptophan-containing cationic heptapeptides (CAMPs) influenced their ability to bind to microbial membranes and their antimicrobial effectiveness against several ESKAPE pathogens. We found that charge delocalization over the peptide backbone enhanced antimicrobial activity, while charge localization at the C- and N-termini reduced efficacy (1.

Gastric microbiota‑derived postbiotics as antimicrobial and anti‑biofilm agents.

BACKGROUND: The human gastric microbiota is recognized as a diverse and metabolically active ecosystem with significant roles in health and disease. Beyond probiotics, postbiotics, non-viable microbial products with antimicrobial and anti-biofilm properties, offer stable and safer therapeutic alternatives. Rising antimicrobial resistance and biofilm associated infections caused by pathogens such as Staphylococcus aureus and Candida albicans highlight the urgent need for new strategies. Based on this evidence, the present study aimed to isolate bacterial strains from gastric biopsies of patient

In Vitro Longitudinal Confocal Fluorescence Microscopy and Imaging Cytometry to Quantify Neutrophil Swarming, Opsonophagocytosis, and Vital NETosis During Implant-Associated Infection.

The "Race for the Surface" hypothesis remains the dogma for explaining the competition between host proteins, cells, and bacteria for implant colonization immediately post-op. Recently, we used intravital microscopy to provide direct evidence supporting this hypothesis by demonstrating that the race for the surface in a murine implant-associated infection (IAI) model is complete within 3 h. However, this intravital imaging is impractical for early-stage drug development research, thus warranting a cost-effective screening platform to mechanistically assess neutrophil-S. aureus interaction dyna

Deep Spinal Infection Caused by Corynebacterium simulans: A Case Report.

Corynebacterium simulans is a commensal coryneform bacterium that commonly colonizes human skin and mucosal surfaces. Owing to limitations in conventional microbiological identification, it is often regarded as a contaminant, and clinically significant deep-seated infections caused by this organism are rarely reported. Here, we describe a case of lumbar infectious spondylitis caused by C. simulans in a 62-year-old male farmer who presented with a 2-month history of persistent low back pain accompanied by pain in both lower limbs. The patient had several potential predisposing factors including

Image-Guided Magnetic Microrobots for Microplastic Capture and Biofilm Removal in the Female Reproductive System.

The health of the female reproductive system is increasingly threatened by emerging environmental contaminants that cause potential bacterial infections, cancer, and fertility problems. Yet effective strategies for the localized removal of these contaminants remain largely unexplored. Among these contaminants, microplastics have emerged as a major global health concern, with growing evidence of their accumulation in human tissues and associated with infectious diseases, carcinogenesis, cellular aging, and infertility due to direct or indirect contact with synthetic fabrics, plastic kitchenware

Remote Coacervate Robots with Visualized Bacteria Sensing and Removal in Indwelling Catheters for Infection Prevention.

Catheter-associated urinary tract infections (CAUTIs) have been a common but challenging disease due to difficulties in the accurate prescribing of antibiotics, while existing clinical diagnostic methods are deficient in their quickness, simplicity, and convenience. Here we report a point-of-care sensing technique using untethered coacervate robots, which demonstrate high controllability and adaptation in confined catheters filled with viscous urine. Under different motion modes, the controlled coacervate robots can achieve a quick, visualized, and selective fluorescence response to Gram-negat
