The influencing factors, challenges, optimization strategies and food applications of antimicrobial peptides.

The incidence and mortality caused by bacterial infections have increased significantly in the global context of declining efficacy of traditional antimicrobial drugs, placing considerable economic and social burdens on healthcare systems and the food safety sector. However, conventional antibiotics have several limitations, including the promotion of antimicrobial resistance, relatively narrow antimicrobial spectra, and limited efficacy against complex biofilms. Therefore, the development of novel antimicrobial agents has become a major research priority for the global scientific and medical

A Link Between Immunoglobulin M Levels and Overwhelming Post-Splenectomy Infection Risk in a National Asplenia Cohort.

BACKGROUND: Overwhelming post-splenectomy infection (OPSI) is a rare complication of asplenia that is frequently fatal. OPSI risk remains elevated in patients with asplenia who receive the Advisory Committee on Immunization Practices (ACIP)-recommended vaccines, suggesting additional variables influence OPSI risk. OBJECTIVE: To identify clinical and immunologic factors associated with OPSI in individuals with anatomic or functional asplenia. METHODS: A retrospective cohort study was conducted of 2,992 individuals with physician-confirmed anatomic or functional asplenia and identified 47 OPSI

Microbial Etiology of Postoperative Spinal Fusion Infections and Factors Associated with Gram-Negative Infections, United States, 2021-2022.

Accurate knowledge of microbial etiology is essential for optimizing spinal fusion surgical site infection (SSI) management, yet contemporary national data in the United States, particularly prevalence and factors associated with gram-negative infections, remain poorly understood. We conducted a cross-sectional analysis of US data reported to the Centers for Disease Control and Prevention National Healthcare Safety Network (2021-2022) to characterize the microbiology of spinal fusion SSI and identify factors associated with gram-negative infection. Among 7,187 cases, gram-negative organisms ca

Probiotic-loaded biomaterial for wound healing and tissue regeneration: a unique medical tool.

Deeper skin wounds lead to infections prolonging the healing process resulting in further complications. Although the treatment protocol includes mostly the use of antibiotics, the main challenge comes with their resistance and poor pharmacokinetics. Selective strain specific probiotic infused hydrogel system is one such newer approach in wound management to counter these issues. The highly sensitive probiotic strain needs proper conditions for colonization and protection against external environment. In this study, an economically feasible and simple attempt was made to incorporate the probio

Fluorescent tagging for assessment of environmental cleaning in an equine hospital.

BACKGROUND: Healthcare-associated infections increase morbidity, mortality, and treatment costs in both human and veterinary medicine, and environmental contamination can contribute to their occurrence. Monitoring of environmental cleaning is an important component of multimodal infection control strategies. Fluorescent tagging is a low-cost, practical approach that correlates with reduced environmental contamination in human and small animal hospitals. Its applicability in equine hospitals, where surface characteristics and contamination risks differ, has not yet been evaluated. OBJECTIVES:

Intratumoral Fusobacterium nucleatum promotes ferroptosis resistance and a disulfide-stress-associated vulnerability under glucose restriction.

Fusobacterium nucleatum (Fn) is an intracellular oncogenic bacterium implicated in multiple cancers. Although Fn has been linked to resistance to anticancer therapies, its effects on therapies targeting metabolic cell-death programs remain unclear. Here, we show that intratumoral Fn abundance is positively associated with SLC7A11 expression and that Fn infection attenuates Era- and RSL3-induced ferroptosis in tumor cells. Live-versus-killed, donor-based, and pharmacological evidence suggests that H2S generated during Fn infection contributes to SLC7A11 induction and engagement of the downstrea

A Sulfur-Modified Cryptolepine-Based Type I/II AIE Photosensitizer for Efficient Eradication of Planktonic Bacteria and Biofilms.

In recent years, the application of aggregation-induced emission photosensitizers (AIE PSs) in photodynamic therapy has gained significant traction as an effective approach for eradicating bacteria and biofilms. We developed a series of AIE PSs based on structural modifications of the natural product cryptolepine. Among them, TPA-CRYS exhibited exceptional type I/II reactive oxygen species (ROS) generation capability. At a low concentration of 20 nM, TPA-CRYS exerted potent antibacterial activity, achieving over 99.7% eradication of both Staphylococcus aureus and MRSA. Notably, TPA-CRYS effect

Triple Combination Therapy: A Promising Path Forward against β-Lactamase-Mediated Resistance.

The silent pandemic of antimicrobial resistance (AMR) continues to drive global morbidity and mortality. Resistance is often mediated by co-expression of serine β-lactamases (SBLs) and metallo-β-lactamases (MBLs), limiting treatment options. While several SBL inhibitors are clinically available, MBL inhibitors are yet to reach the market, and only a few candidates are in late-stage development. Dual-β-lactamase inhibitors remain an unmet goal. To preserve existing antibiotics, alternative strategies are required. This perspective highlights triple combination therapy comprising a β-lactam anti

Artificial Intelligence-Integrated Proteome-Wide Target Prioritization for Drug Discovery against Multidrug-Resistant Neisseria gonorrhoeae.

Neisseria gonorrhoeae causes ∼82.4 million infections annually and is rapidly approaching an untreatable status. The pathogen has developed resistance to every antibiotic class introduced since the sulfonamide era. Ceftriaxone, the last WHO- and CDC-recommended empirical therapy, is now compromised by high-level resistance in extensively drug-resistant lineages reported across multiple continents. No licensed vaccine exists, and no antibiotic with a novel mechanism has been introduced for this pathogen in over 40 years. Conventional drug discovery is too slow and insufficiently resistance-awar

Sequence type-associated variation in blood brain barrier invasion capacity among predominant Listeria monocytogenes strains in China.

Listeria monocytogenes (LM) can cause severe central nervous system infections; however, current understanding of its neuroinvasion is largely based on studies using a limited number of reference strains, leaving differences among sequence types (STs) in their capacity to invade across the blood-brain barrier (BBB) poorly defined. In this study, 12 LM isolates from China, representing ST5, ST8, ST9, and ST87, were investigated using whole-genome sequencing, an in vivo oral murine infection model, and an hCMEC/D3-based in vitro BBB model to assess virulence gene profiles, organ colonization, an
