A Consistently Elevated Pineal Gland Uptake of 18F-MFBG as a Physiological Normal Variant.

A 3-year-old girl with high-risk neuroblastoma underwent 18F-MFBG PET/CT for therapy response evaluation as part of a clinical trial. The scan revealed focal increased uptake in the pineal gland, which was also observed on a concurrent 68Ga-DOTATATE PET/CT. Brain MRI showed no signs of metastasis, and importantly, follow-up 18F-MFBG PET/CT demonstrated consistently elevated uptake in the same region without interval change. These combined findings, negative MRI, concordant physiological 68Ga-DOTATATE uptake, and stable appearance on serial 18F-MFBG PET/CT, support the interpretation of this pi

The effects of pinealectomy and melatonin on bisphenol a diglycidyl ether (BADGE)-induced hepatotoxicity in rats: involvement of apoptotic, inflammatory, and oxidative mechanisms.

In this study, rats were deprived of melatonin by removing their pineal gland (PinX), and the effects of Bisphenol A Diglycidyl Ether (BADGE) on oxidative stress, inflammation, apoptosis, histopathology, liver cell count, and mean liver volume were investigated. Furthermore, the protective effects of melatonin against the potential toxic effects of BADGE were assessed through exogenous melatonin administration. Fortyeight rats were divided into 6 groups (n = 8): Sham PinX, Sham PinX+BADGE, Sham PinX+BADGE+Mel, PinX, PinX+BADGE, and PinX+BADGE+Mel. The rats' pineal glands were either removed or

Melatonin Regulates Sheep Granulosa Cell Proliferation and Apoptosis Through a Novel Micropeptide, IGF1Raa.

Melatonin is a hormone primarily secreted by the pineal gland, best known for regulating circadian rhythms. It also plays a critical role in ovarian physiology by modulating granulosa cell (GC) function during follicular development, although the underlying molecular mechanisms remain incompletely understood. Recent advances in ribosome sequencing (Ribo-seq) have revealed that many small open reading frames (sORFs), previously overlooked in conventional annotations, actually encode functional micropeptides. However, whether melatonin exerts its ovarian effects through such micropeptides has re

Constant Light Versus Intermittent Constant Light/Recovery Regimen: Differential Effects on Pineal Ultrastructure, Clock Gene Protein Abundance, and Systemic Metabolism in Rats.

Disruption of the natural light regimen is considered a key pathogenetic factor in the development of desynchronosis and associated diseases. The pineal gland, as a neuroendocrine organ, is a central component of the circadian system and the primary target of the pathogenic effects of disrupted lighting. However, it remains unclear whether the mechanisms of pineal damage differ between different types of light stress. In this study, 120 male Wistar rats underwent a comprehensive analysis of the morphofunctional state of the pineal gland under two lighting regimens: constant lighting and interm

Smaller pineal volume in Alzheimer's disease than in dementia with Lewy bodies.

BACKGROUND: Pineal gland volume reduction in Alzheimer's disease (AD) has been reported. Recent findings also indicate an association between pineal volume reduction and dementia with Lewy bodies (DLB). OBJECTIVE: This study aimed to compare pineal volume between two major neurodegenerative disorders: AD and DLB. METHODS: This cross-sectional analysis included patients with AD or DLB who underwent brain magnetic resonance imaging (MRI) and Mini-Mental State Examination (MMSE). AD was diagnosed based on the National Institute on Aging-Alzheimer's Association criteria and confirmed by positive

Papillary Tumor of the Pineal Region Presenting With Obstructive Hydrocephalus: A Diagnostic and Management Challenge.

We report a 45-year-old woman with a papillary tumor of the pineal region (PTPR), a neuroepithelial neoplasm of the pineal gland, who presented with a one-year history of headaches, visual disturbance, and gait impairment and was found to have a pineal mass with obstructive hydrocephalus. She underwent endoscopic third ventriculostomy (ETV) with biopsy; the initial radiologic impression favored pineocytoma. Immunohistochemistry was nondiagnostic on the limited sample, and the diagnosis of PTPR was established only after review at two academic neuropathology centers. DNA methylation profiling w

Pineal melatonin modulates responsiveness to exogenous melatonin via the IFN-γ-CXCL9/10-CXCR3 axis in gastric cancer.

BACKGROUND: Melatonin is a pineal-derived neuroendocrine hormone with pleiotropic actions on tumor biology and immune regulation. Although exogenous melatonin has been reported to exert antitumor and immunomodulatory effects, whether these actions depend on an intact endogenous pineal melatonin background remains unclear. In particular, the role of endogenous melatonin in conditioning chemokine-mediated antitumor effects has not been defined. METHODS: A male mouse gastric cancer model was used to investigate the interplay between endogenous and exogenous melatonin in tumor growth, immune infi

Serotonergic Control of Spine Morphology During Growth in Zebrafish.

Scoliosis is a three-dimensional deformity of the vertebral column that commonly emerges during growth, yet the biological mechanisms that initiate and drive curve progression remain poorly understood. Serotonin (5-hydroxytryptamine [5-HT]) has long been linked to scoliosis through its relationship with pineal melatonin signaling and through genetic associations with human disease, but its role in spinal morphogenesis has remained unclear. Here, using zebrafish, we show that serotonergic signaling is required for normal spine morphology. Transient exposure to 5-HT during development induces la

Beyond Brain Parenchyma: A Pictorial Review of Non-Parenchymal and Extracranial Metastases on Cranial MRI.

Brain MRI is the primary imaging technique used to detect, stage, and monitor intracranial metastatic disease. While parenchymal metastases are easily identifiable, metastatic involvement beyond the brain parenchyma is frequently subtle and underreported, despite representing a significant proportion of metastatic findings identified during cranial examinations. Failure to identify these metastases, or identification at a late stage, can lead to incomplete staging, inappropriate treatment selection, and delays in initiating targeted therapies. This pictorial review presents a systematic, compa

Influence of Intravenous Norepinephrine Administration on Melatonin Plasma Concentration in Awake and Anaesthetised Healthy Adult Volunteers.

Melatonin (MEL) is an endogenous hormone that regulates circadian rhythm, immune function and oxidative stress responses. Norepinephrine (NE), an endogenous catecholamine, increases blood pressure through the sympathetic nervous system and is commonly administered intraoperatively or in the clinically ill patient for hemodynamic support. Although endogenous NE is known to stimulate MEL synthesis, the effects of exogenous intravenous NE on plasma MEL concentration, particularly under general anaesthesia (GA), remain poorly characterised. This study examined the influence of intravenous NE on pl

Variable and fixed rate debt: Gambling on the future state of the economy after Covid-19.

Melatonin is a product of the amino acid tryptophan in the pineal gland. Once synthesized, the specific mechanisms governing the release of melatonin from the pineal gland and its functions are largely unknown. Besides its regulatory role in circadian rhythms in mammals, because of its widespread subcellular distribution, melatonin contributes to the reduction of oxidative damage in both the lipid and the aqueous environments of the cell. This postulate is widely supported by the experimental observations showing that melatonin protects lipids in membranes, proteins in the cytosol, and DNA in

Transcriptomic analysis of pigeon retina and pineal gland illuminated with red light.

1. This study investigated the transcriptomic expression of the pigeon retina and pineal gland when exposed to red light (RL). Light is a crucial environmental factor influencing poultry production, physiology and behaviour. Different wavelengths of light have distinct effects on photoreceptors, including those in the retina and pineal gland, ultimately regulating various production benefits.2. Using transcriptome sequencing, this trial examined the gene expression profiles of the retina and pineal gland of White King pigeons exposed to either red light (RL; 660 nm) or White light (WL; 400-760

Melatonin alleviates gentamicin-induced acute kidney injury through the Keap1/Nrf2/HO-1 signaling pathway.

Drug-related factors represent a primary cause of acute kidney injury. Gentamicin (GM), while being one of the most effective and commonly used clinical agents against Gram-negative bacteria, frequently induces nephrotoxicity and triggers acute kidney injury during treatment. Melatonin, a natural antioxidant produced by the pineal gland, has been shown in recent studies to mitigate drug-induced nephrotoxicity. This study aimed to delineate the dose-dependent effects and underlying mechanisms of GM-induced acute kidney injury, along with the protective role of melatonin. Results demonstrated th

Physiological relevance of autocrine melatonin signaling in pineal and extrapineal sites: a systematic review.

Beyond the pineal gland, melatonin is produced locally in many extrapineal organs, where it mediates local tissue homeostasis. However, little attention is paid to the physiological role of autocrine melatonin signaling across body organs. This study synthesizes original data to address this gap, as gaining insight into this topic could lead to new therapeutic approaches for diseases associated with melatonin. This systematic review used a narrative synthesis, following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) 2020 guideline standards to synthesize origin
