Novel α-amylolyzates derived from enzymatically synthesized α-glucans using diverse glycogen branching enzymes decelerate glucose release by modulation of intestinal α-glucosidases.

In this study, the internal branched structure of highly branched α-glucans (HBαGs) was regulated by employing glycogen branching enzymes (GBEs) from different microbial sources and amylosucrase, with the purpose of synthesizing structurally diverse α-amylolyzates. Variations in GBE origin resulted in HBαGs with distinct fine structural features, including differences in α-1,6 branching degree and molecular weight. Following hydrolysis with Aspergillus oryzae α-amylase, the resulting α-amylolyzates exhibited a high degree of branching (from 26.6 to 30.9%) and large molecular weight (1.07 × 106

Temporal and spatial atlas of eosinophil specialization across tissues.

Eosinophils are multifunctional granulocytes involved in immune regulation, metabolism and tissue repair; however, the extent of their heterogeneity across tissues and the principles governing their specialization are not well defined. Here, we present a single-cell transcriptomic and proteomic atlas of mouse eosinophils spanning immune, barrier and metabolic sites. By integrating transcriptional profiling, high-dimensional surface proteomics and in vivo fate mapping, we show that eosinophil identity is shaped by both tissue-derived cues and the duration of local residency. Long-lived eosinoph

Transcriptional stratification of necrotizing enterocolitis identifies distinct disease subtypes.

Necrotizing enterocolitis (NEC) is a severe inflammatory disorder of the intestine that affects neonatal babies, with a mortality rate of 30-50%. Despite the severe nature of the disease, its rare incidence and early onset prevent further understanding of its etiology and the development of treatment strategies. To further understand the pathogenic mechanism of this disease, we subjected intestinal tissues from ten NEC patients and three control individuals to RNA-sequencing. Clustering of the affected tissues revealed three clusters, each with a distinct transcriptional profile characterized

Genetic dissection of serum pro-neurotensin suggests potential causal impact on brain structure.

BACKGROUND: Pro-neurotensin (pro-NT) is the stable circulating precursor of neurotensin (NT), a neuropeptide expressed mainly in the central nervous system and small intestine that regulates key physiological processes like fatty acid absorption in the gut and suppresses appetite via central mechanisms. Studies in NT-deficient mice and humans implicate NT in obesity and insulin resistance, highlighting its role in metabolic regulation. METHODS: To explore the genetic determinants of circulating pro-NT and its causal relationships with obesity and brain phenotypes, we conducted a genome-wide m

Brain irradiation drives remote liver changes via senescence-independent mechanisms.

BACKGROUND AND PURPOSE: Although radiotherapy is essential for treating brain tumors, it often damages surrounding healthy brain tissue, causing long-term neurocognitive dysfunction. Cellular senescence, a stable state of cell cycle arrest triggered by various stressors including radiation-induced DNA damage, has been implicated in aging- and neurodegeneration-associated cognitive decline via its pro-inflammatory secretome and has been reported to exert paracrine effects on distant organs. However, whether brain irradiation, particularly through the induction of cellular senescence, can trigge

A NEW SPECIES OF TRICHOLEIPERIA (NEMATA: TRICHOSTRONGYLOIDEA: MOLINEIDAE) FROM LAMPRONYCTERIS BRACHYOTIS (CHIROPTERA: PHYLLOSTOMIDAE) IN COLOMBIA.

Tricholeiperia albae n. sp. is described from the small intestine of the yellow-throated big-eared bat, Lampronycteris brachyotis (Chiroptera: Phyllostomidae). Morphologic comparisons and a review of nematode parasites of neotropical bats indicate that this represents an undescribed species. Herein, we describe this species as new and show that this is a novel bat-parasite association. More than 50 species of bats have been reported from the Apure-Villavicencio dry forests ecoregion, but little is known of the parasite fauna. Continued exploration of new localities in Colombia underscores the

Petersen's Space Hernia: An Uncommon Diagnosis With Potentially Fatal Consequences.

Petersen's space hernias are a rare but potentially life-threatening complication following Roux-en-Y gastric bypass, seen in both bariatric and oncologic contexts. Although evidence is mixed, some studies suggest a higher risk of internal herniation after laparoscopic procedures. A 52-year-old male patient with a prior history of a minimally invasive bariatric Roux-en-Y gastric bypass presented with acute left upper quadrant pain and nausea. Imaging revealed small bowel obstruction with mesenteric twisting, consistent with an internal hernia. Exploratory laparotomy confirmed a Petersen's spac

Early-in-life inhalation of ferrocene-derived diesel exhaust-induced metabolic and small intestinal toxicities: Roles of peroxisome proliferator-activated receptor gamma and ferroptosis.

Ferrocene-derived diesel exhaust (FDE) had been demonstrated to exhibit enhanced cardiopulmonary toxicities. This study evaluated the potential metabolic and small intestinal toxicities following early-in-life inhalation exposure to FDE using an in ovo air cell inhalation model in chicken embryos. Ferrocene was added to standard diesel fuel at 205, 410 or 820 mg/L (corresponding to 75, 150 or 300 ppm iron) and combusted in a diesel generator. The resulting FDE was exposed to embryonic day 18-19 chicken embryos via air cell inhalation. Serum, small intestine, white adipose tissue, and pancreas

Small Intestine Length Measurement Using 3D CT Volumetry and in Vivo Laparoscopic Measurement Using Pre-marked Graspers: A Comparative Study.

INTRODUCTION: The method for assessing the bowel length is debatable but crucial in bariatric surgeries, especially revisional ones. It may be linked to improved weight loss and reducing nutritional deficiencies; however, it can be time-consuming, increasing the likelihood of complications during and after the surgery. 3D CT volumetry could offer a non-invasive, time-saving alternative for accurate Total Bowel Length (TBL) measurement, potentially reducing operative time and associated risks. OBJECTIVE: This study compares small bowel length using 3D CT volumetry versus in vivo laparoscopic m

Delta-9-Tetrahydrocannabinol alleviates doxorubicin-induced weight loss but does not affect dextran sodium sulphate-induced colitis.

BACKGROUND: Gastrointestinal mucositis is a common adverse effect of chemotherapy. Inflammatory bowel diseases (IBD) are a spectrum of chronic inflammatory disorders of the digestive tract. Mucositis and IBD/colitis are characterized by weight loss and intestinal inflammation. Previous animal studies suggest that the cannabinoid, ∆9-tetrahydrocannabinol (THC), attenuates intestinal inflammation. We aimed to investigate the effect of THC oil on doxorubicin-induced gastrointestinal mucositis and dextran sodium sulphate (DSS) induced colitis in mice. METHODS: Wild-type C57BL6 mice were used for t

Non-occlusive mesenteric ischemia induced by methamphetamine: an autopsy case report.

Non-occlusive mesenteric ischemia (NOMI) is an acute form of intestinal ischemia that occurs in the absence of organic stenosis of the mesenteric arteries. Vasoconstrictive agents have been recognized as potential etiological factors. This report describes the first documented Japanese autopsy case of NOMI induced by methamphetamine (MA), a potent sympathomimetic agent. A forensic autopsy was performed on a man in his 60s who died suddenly. Gross and histopathological findings of the small intestine and mesenteric arteries were examined, and post-mortem toxicological analysis was performed to

Lactobacillus-vectored nanobodies improve broiler productivity under sub-clinical necrotic enteritis with associated microbiome and transcriptome changes.

Sub-Clinical Necrotic Enteritis (SCNE), caused by toxin-producing Clostridium perfringens, is a major challenge in poultry production. SCNE has traditionally been managed with in-feed antibiotics; however, increasing concerns about the spread of antimicrobial resistance call for antibiotic-free strategies for its control. We recently described an NE control strategy leveraging Limosilactobacillus reuteri probiotic strains genetically engineered to deliver nanobodies against alpha toxin and NetB from C. perfringens in the poultry gut. Here, in a controlled study under SCNE conditions, we found

Microbiota-induced T cell plasticity enables immune-mediated tumour control.

Therapies that harness the immune system to target and eliminate tumour cells have revolutionized cancer care. Immune checkpoint blockade (ICB), which boosts the anti-tumour immune response by inhibiting negative regulators of T cell activation1-3, is remarkably successful in a subset of cancer patients. Yet a significant proportion do not respond to treatment, emphasizing the need to understand factors influencing the therapeutic efficacy of ICB4-9. The gut microbiota, consisting of trillions of microorganisms residing in the gastrointestinal tract, has emerged as a critical determinant of im

Cell-body curvature modulates stall frequency to enhance Vibrio cholerae swimming and chemotaxis through hydrogels.

The swimming motility of Vibrio cholerae is a virulence factor that aids in breaching the mucus layer of the small intestine. V. cholerae cells have a curved cell shape and previous work demonstrated that loss of curvature decreases infectivity. Here we investigate the mechanism by which V. cholerae's curvature affects single-cell motility within mucus-mimicking environments. Using a multiscale chemotaxis assay, we compared the chemotactic performance of wild-type curved cells (O1 El Tor C6706) and straight mutants under linear chemical gradients in liquid solutions, viscous solutions, and sof

Two'-Fucosyllactose, a Human Milk Oligosaccharide, Promotes Intestinal Remodeling and Enhances Nutrient and Functional Component Absorption.

2'-Fucosyllactose (2'FL) is an abundant human milk oligosaccharide and is recognized for its diverse biological benefits. The aim of this study was to investigate the effects of 2'FL supplementation on small intestinal remodeling and the associated enhancement of nutrient absorption capabilities. Mice were orally supplemented with 2'FL daily for 20 d. This supplementation significantly increased the length of the small intestine and the density of villi, with no adverse effects on intestinal barrier function. Mice treated with 2'FL exhibited an enhanced absorption of triglycerides containing d

Mechanisms and pathophysiology leading to development of small intestinal microbial dysbiosis.

Small intestinal microbial dysbiosis (SIMD) describes clinical syndromes attributed to overabundance of microbes within the small intestine. Usually attributed to bacteria (but also less commonly to archaea and fungi), SIMD arises as a secondary condition, when there is disruption of one or more key physiological mechanisms. We provide a comprehensive review of the current understanding of SIMD pathophysiology. We highlight the key physiological factors including gastric acid, pancreatic enzymes, bile acids, small intestinal dysmotility and obstruction, colonic motility, and intestinal mucosal
