Diagnostic accuracy of Mycobacterium tuberculosis antigen-induced interferon-γ-inducible protein 10 for latent tuberculosis infection among high-risk groups: a systematic review and meta-analysis.

BACKGROUND: Latent tuberculosis infection (LTBI) affects nearly a quarter of the world's population, yet current diagnostic tools are suboptimal. Interferon-γ-inducible protein 10 (IP-10) has been proposed as a candidate biomarker, but its diagnostic value for LTBI among high-risk groups is unclear. To address this, we performed a systematic review and meta-analysis to assess the accuracy of Mycobacterium tuberculosis (Mtb) antigen-induced IP-10 for detecting LTBI among high-risk groups. METHODS: PubMed, Web of Science, Embase, Scopus, and the Cochrane Library were searched up to April 2026.

Acquisition of a toxin-antitoxin system within a mobile genetic element likely enhances Mycobacterium abscessus fitness by enforcing cargo retention.

UNLABELLED: Infections caused by the emerging pathogen Mycobacterium abscessus (Mab) require prolonged multidrug treatment regimens that frequently fail because its accessory genome has incorporated a daunting array of poorly defined resistance mechanisms. However, the processes governing acquisition and retention of new genetic material within the Mab accessory genome remain poorly understood. Here, we first identified two Mab clinical strains that harbor the same uncharacterized toxin-antitoxin (TA) system at different genomic positions. Using computational analyses of the surrounding DNA co

Mycobacterium tuberculosis small RNAs MTS0997 and MTS1338 regulate both pathogen virulence and host immune responses.

Previously, others and we demonstrated that small RNAs MTS1338 and MTS0997 of Mycobacterium tuberculosis, the causative agent of tuberculosis (TB), are essential for persistence inside host macrophages. However, their role in mycobacterial transcriptional profile regulation was characterized incompletely, and potential influence on host immune responses and the outcome of infection remained obscure. To obtain new insights in M. tuberculosis biology and TB pathogenesis we established unmarked single-knockout (KO) M. tuberculosis strains with deleted genes for MTS0997, or MTS1338, as well as the

Genomic diversity of human-associated Mycobacterium bovis in Canada.

Human infections caused by Mycobacterium bovis in Canada are not well documented. The following study analyzed M. bovis (excluding BCG) isolates identified in public health laboratories across Canadian provinces between 2000 and 2025. Whole-genome sequencing was used to catalog mutations linked to antimicrobial resistance and evaluate the genomic relatedness of isolates. The majority of samples in this study were non-respiratory (60%). Of 74 study isolates, 69 (93%) were phenotypically sensitive to first-line drugs isoniazid, rifampin, and ethambutol. The most frequently encountered M. bovis c

Nonlinear analysis of factors influencing tuberculosis outpatient visits in China: an explainable machine learning approach.

This study aims to analyze the temporal trends, interprovincial differences, and key predictors of Tuberculosis Outpatient Visits (TOV) in China. It provides empirical evidence for understanding regional differences in tuberculosis-related outpatient service uti- lization. Panel data from 30 Chinese provinces from 2012 to 2024 were used. The study included variables related to economic development, health resources, education resources, environmental governance, pollutant emissions, and technological innovation. Correlation analysis, model comparison, and XGBoost-SHAP analysis were applied. Fr

Diagnosis of tuberculous lymphadenopathy: the role of droplet digital PCR.

BACKGROUND: Tuberculous lymphadenopathy (TB-LAP) is the most common form of extrapulmonary TB and remains diagnostically challenging due to its paucibacillary nature. We evaluated the diagnostic performance of droplet digital PCR (ddPCR) for detecting Mycobacterium tuberculosis (Mtb) DNA in formalin-fixed paraffin-embedded (FFPE) lymph node specimens. METHODS: We retrospectively analysed 102 lymph node FFPE specimens with microbiologically confirmed or pathology-suspected TB-LAP. We performed ddPCR targeting the IS6110 gene and correlated it with pathological findings and mycobacterial cultur

DNA damage repair-related gene signatures delineate immune-distinct subtypes and diagnostic cues in tuberculosis.

BACKGROUND: DNA damage repair-related genes (DDR-RGs) may shape tuberculosis (TB) pathogenesis, but their molecular characteristics, diagnostic utility, and cellular context remain insufficiently defined. METHODS: We integrated multiple GEO transcriptome datasets, identified differentially expressed genes, constructed weighted gene co-expression network analysis modules, and intersected these results with curated DDR-RGs to obtain candidate genes. An ensemble framework of 110 machine-learning model combinations was used to select diagnostic markers, followed by external validation, consensus

Toxin-mediated tRNATrp depletion drives Trp-codon-dependent reprogramming in Mycobacterium tuberculosis while selectively enriching the PDIM biosynthetic machinery.

Mycobacterium tuberculosis (Mtb) must adapt to host-imposed stresses that limit its growth and ability to establish an infection. One important immune-regulated host defense mechanism involves competition between host and pathogen for tryptophan (Trp). Although Mtb is a Trp prototroph capable of synthesizing its own Trp, it normally acquires this amino acid from host macrophages because of the high energetic cost of its synthesis. However, interferon-γ-activated macrophages attempt to restrict Mtb growth by inducing indoleamine 2,3-dioxygenase 1 (IDO1) that degrades Trp. In response, Mtb sense

Therapeutic and Pharmacokinetic Challenges With Treatment of Nontuberculous Mycobacteria Infection in Two Lung Transplant Recipients: Case Report.

Lung transplant (LTx) recipients are susceptible to pulmonary nontuberculous mycobacteria (NTM) infections due to pre-transplant colonization and post-transplant immunosuppression. This presents a challenge when managing immunosuppression and infection prophylaxis post-LTx due to drug-drug interactions (DDIs) with antimycobacterial agents. We present two cases of LTx recipients requiring concomitant rifabutin and isavuconazole. A 62-year-old male receiving rifabutin for pre-LTx pulmonary Mycobacterium avium complex (MAC) infection was initiated on isavuconazole for post-LTx fungal prophylaxis.

Association between tuberculosis, HIV infection, and diabetes mellitus: should we integrate screening and care? A systematic review.

Tuberculosis (TB) and HIV/AIDS are among the leading infectious diseases worldwide, while diabetes mellitus (DM) is a major chronic endocrine and metabolic disorder. Together, these conditions contribute substantially to the global burden of disease and mortality and are increasingly recognized as interconnected comorbidities. The combination of these three diseases can worsen patients' health conditions, increase mortality risk, and complicate treatment, which is why this study aims to summarize the relationship between HIV, TB, and DM. To achieve this, a systematic literature search was cond

Tuberculosis disease burden trends, drivers, and projections to 2050 in China and the wider Central Asian region, 1990-2023.

BACKGROUND: Tuberculosis (TB) remains a leading infectious killer, with high burden concentrated in China and wider Central Asia. Integrated long-term trend analysis and forecasting for this region are scarce. We aimed to quantify TB burden trends, drivers, inequalities, and 2050 projections across China and wider Central Asia (1990-2023). METHODS: We used Global Burden of Disease 2023 data to estimate age-standardized incidence, prevalence, mortality, and disability-adjusted life years (DALYs). Joinpoint, age-period-cohort, decomposition, and Bayesian APC models were applied. Socioeconomic i

[The role of therapeutic education in the care of tuberculosis patients].

Tuberculosis is a major cause of death worldwide. It affects mainly low- and middle-income countries, but also persists in vulnerable populations in high-income countries. How well tuberculosis treatments work depends on strict adherence, which is compromised by the length of the protocols, the adverse effects of the drugs, and unfavourable psychosocial factors. Therapeutic education aims to strengthen the self-management skills, autonomy, and quality of life of patients with tuberculosis. It applies to both latent tuberculosis infection and active tuberculosis and improves understanding of th

Flavonoids in treatment of tuberculosis: from molecular mechanisms to structure activity relationships.

Tuberculosis (TB) being caused by M. tuberculosis is a major global health burden especially in low and middle income countries especially with the rise of drug resistant (MDR/RR- TB) strains and poor treatment outcomes. Limitations of traditional anti-TB treatment, such as toxicity, cost and emerging drug resistance mean that the development of alternative therapy approaches is needed. Flavonoids are a diverse group of plant-derived polyphenolic compounds which have received considerable attention because of their remarkable potent anti- mycobacterial and host directed therapeutic properties.

Development of a multiplex PCR for detection of pathogenic Mycobacterium orygis in cattle tissues harboring tuberculous-like lesions: a pilot study.

Mycobacterium orygis, a member species of Mycobacterium tuberculosis complex (MTBC), is emerging as a major threat to the control of zoonotic tuberculosis. The lack of low-cost diagnostic assays to differentiate M. orygis from other MTBC members has created a critical data gap in estimating its true burden in human and animal populations, especially in the Asian subcontinent. In this study, a multiplex PCR assay was developed to distinguish M. orygis from other MTBCs, based on two M. orygis-specific non-synonymous point mutations in the mbtG and fadD23 genes. The assay exhibited a limit of det
