Tuberculosis is a major cause of death worldwide. It affects mainly low- and middle-income countries, but also persists in vulnerable populations in high-income countries. How well tuberculosis treatments work depends on strict adherence, which is compromised by the length of the protocols, the adverse effects of the drugs, and unfavourable psychosocial factors. Therapeutic education aims to strengthen the self-management skills, autonomy, and quality of life of patients with tuberculosis. It applies to both latent tuberculosis infection and active tuberculosis and improves understanding of th

INTRODUCTION: Adrenal infections are an underrecognized but clinically significant cause of primary adrenal insufficiency (PAI).
OBJECTIVES: This review proposes a diagnostic algorithm for the evaluation of suspected cases.
RESULTS: The most common causative pathogens are Mycobacterium tuberculosis and Histoplasma capsulatum; however, bacterial infections and viral pathogens such as cytomegalovirus (CMV) and SARS-CoV-2 (COVID-19) have also been reported. These infections, particularly in tuberculosis and histoplasmosis, can induce granulomatous inflammation, tissue necrosis, and destruction

Tuberculosis (TB) being caused by M. tuberculosis is a major global health burden especially in low and middle income countries especially with the rise of drug resistant (MDR/RR- TB) strains and poor treatment outcomes. Limitations of traditional anti-TB treatment, such as toxicity, cost and emerging drug resistance mean that the development of alternative therapy approaches is needed. Flavonoids are a diverse group of plant-derived polyphenolic compounds which have received considerable attention because of their remarkable potent anti- mycobacterial and host directed therapeutic properties.

Mycobacterium orygis, a member species of Mycobacterium tuberculosis complex (MTBC), is emerging as a major threat to the control of zoonotic tuberculosis. The lack of low-cost diagnostic assays to differentiate M. orygis from other MTBC members has created a critical data gap in estimating its true burden in human and animal populations, especially in the Asian subcontinent. In this study, a multiplex PCR assay was developed to distinguish M. orygis from other MTBCs, based on two M. orygis-specific non-synonymous point mutations in the mbtG and fadD23 genes. The assay exhibited a limit of det

Tuberculosis (TB), caused by Mycobacterium tuberculosis, reclaimed its status as the deadliest single-pathogen infectious disease globally in 2023, accounting for 10.8 million new cases and 1.25 million deaths in 2023. The escalating crisis of drug-resistant TB, suboptimal BCG vaccine efficacy in adults, and persistent treatment gaps for vulnerable populations collectively drive an urgent shift toward mechanism-driven, precision-oriented therapeutic strategies. This review presents a critically integrated appraisal of anti-TB research advances spanning 2020 to 2025, drawing on structural biolog

INTRODUCTION: Lesotho has among the world's highest TB prevalences and the national program finding only 50% of incident cases. Lesotho underwent its first TB prevalence study in 2019 in reviewing this study, the authors recognized that the standard measures for screening and diagnosis TB often failed to detect the presence of culture positive pulmonary TB. This review seeks to determine if the systematic under detection of culture positive TB by standard methods is true among prevalence surveys in Africa.
METHODS: We conducted a review to identify TB prevalence surveys conducted in Africa fr

UNLABELLED: Airway compression caused by intrathoracic non-tuberculous mycobacteria (NTM) lymphadenitis is particularly uncommon in otherwise healthy children and remains poorly characterized. We aimed to describe the clinical, radiological, microbiological, and immunological features of a French pediatric cohort diagnosed with mediastinal Mycobacterium avium complex (MAC) lymphadenitis. We conducted a retrospective multicenter study that included children diagnosed with bacteriologically confirmed intrathoracic MAC lymphadenitis in France between 2009 and 2024. Patients with HIV-related immun

BACKGROUND: Tuberculosis (TB) remains the leading infectious-disease cause of death globally, with an estimated 10.7 million incident cases and 1.23 million deaths in 2024. Its persistence is driven not by microbial factors alone, but by a complex interplay of biological, psychological, and social determinants influencing exposure, vulnerability, and access to care. Despite growing recognition of these biopsychosocial dimensions, the available evidence remains fragmented across disciplines, and no previous review has comprehensively mapped their scope while simultaneously characterizing the st

Background and objectives Childhood tuberculosis remains underdiagnosed in India. Although gastric aspiration is recommended to improve microbiological confirmation, its use at lower-tier health facilities is limited by training, infrastructural, and logistical constraints. This study evaluated whether a structured, training-focused implementation strategy could integrate gastric aspiration into routine paediatric TB diagnostic workflows at primary and community health settings, and assessed its feasibility, acceptance, uptake, and diagnostic yield. Methods We conducted a mixed-methods control

Tuberculosis (TB) can progress despite substantial systemic immune activation, indicating that protection and immunopathology are shaped not only by the magnitude of host responses but also by their organization within individual lesions. TB granulomas are heterogeneous tissue ecosystems in which cellular composition, necrosis, vascular access, extracellular-matrix remodeling, metabolic stress, and local drug exposure vary across lesions and over time. Spatial and single-cell technologies now enable these features to be investigated at increasing resolution; however, spatial or molecular assoc

RATIONALE: Near point-of-care nucleic acid amplification tests (NPOC-NAATs) are a new class of diagnostics that can be used for tuberculosis diagnosis. NPOC-NAATs are suitable for use in peripheral levels of healthcare systems as they are battery-operated instrument-based tests without special infrastructure requirements and can be performed by health care workers with basic technical skills. We evaluated the accuracy of NPOC-NAATs in diagnosing pulmonary tuberculosis using respiratory samples.
OBJECTIVES: To assess the diagnostic accuracy of NPOC-NAATs for detecting pulmonary tuberculosis us

BACKGROUND: Active case finding (ACF) for tuberculosis (TB) can reduce transmission, yet efficient targeting requires high-quality surveillance data. We investigated the potential costs and impact of targeted ACF, guided by local estimates of the annual risk of TB infection (ARTI) derived from Mycobacterium tuberculosis (Mtb) immunoreactivity survey data in children aged <5 years.
METHODS AND FINDINGS: Using mathematical models parameterized with local data, we compared three case-finding approaches across 33 urban neighborhoods in Blantyre, Malawi: passive case finding (PCF) only; PCF with u

BACKGROUND: Accurate forecasting of pulmonary tuberculosis (PTB) is frequently impeded by surveillance artifacts, most notably health policy shifts and festival holidays. This study proposes a practical epidemiological forecasting framework to address these real-world disruptions within China's monitoring data.
METHODS: We utilized an 11-year historical cohort (2014-2024) from Jiulongpo District, Chongqing. To ensure longitudinal comparability, we first performed an epidemiological standardization to mitigate the artificial structural break caused by the 2018 diagnostic reclassification. To e

BACKGROUND: Tuberculosis (TB) preventive treatment (TPT) is a core component of TB control. Levofloxacin was recently recommended by the World Health Organization as TPT in children exposed to multidrug-resistant tuberculosis (MDR-TB). However, the affordability and feasibility of TPT provision for governments, health services, individual households and global donors is uncertain.
METHODS: This costing analysis used empirical data collected alongside the TB-CHAMP clinical trial which investigated levofloxacin TPT in children exposed to MDR-TB in South Africa. We modelled the societal (health

AIM: Among Aotearoa New Zealand-born people with tuberculosis (TB), half are Māori. TB is preventable, and Aotearoa New Zealand has signed up to the global strategy to end the TB epidemic. TB among Māori has endured since the late 1800s and occurs in a complex political, social and wellbeing context. We aimed to review current public health practices used for contacting and following up Māori with TB and their contacts.
METHODS: We undertook a retrospective case review of Māori notified with pulmonary TB (2018- 2023) in the Waikato Region. Data included TB notification, treatment start and en

BACKGROUND: Prostatic tuberculosis is a rare manifestation of genitourinary tuberculosis and an uncommon form of extrapulmonary tuberculosis. Because its clinical presentation is often nonspecific and closely resembles benign prostatic hyperplasia (BPH), chronic prostatitis, or prostate carcinoma, the diagnosis is frequently delayed and often established only after histopathological examination of prostatic tissue. Reporting such cases is important to increase clinical awareness and facilitate timely diagnosis, particularly in tuberculosis-endemic regions.

BACKGROUND: Leprosy and other skin neglected tropical diseases, including Buruli ulcer and yaws, remain significant public health challenges in Ghana. Despite control efforts, these conditions continue to cause disability, stigma, and social exclusion, particularly in rural and underserved communities. Misconceptions rooted in cultural and spiritual beliefs further delay care-seeking and reinforce discrimination.
OBJECTIVE: This study assessed community awareness, symptom recognition, and perceived causes of leprosy and other skin NTDs across four regions of Ghana, and examined how these vari
