Multifunctional hydrogel assembled from phenylboronic acid-chitosan and Polygonati Rhizoma polysaccharide for accelerated radiation-combined wound healing.

Combined radiation-wound injury (CRWI) is difficult to heal because of persistent oxidative stress, inflammation, and impaired angiogenesis. Herein, a multifunctional pH and reactive oxygen species (ROS)-responsive hydrogel was prepared by dynamic crosslinking of Polygonati Rhizoma-derived polysaccharide fraction (PC) with phenylboronic acid-modified chitosan (CS-BA) through reversible boronate ester bonds. The obtained PC/CS-BA hydrogel was injectable and exhibited tissue adhesion, self-healing ability, and pH/ROS-responsive network disassembly. The optimized PC/CS-BA-1 hydrogel showed favora

TLR4 epidermal genetic deletion protects against acute and tumorigenic UV exposure in SKH-1 mouse skin.

Epidermal keratinocytes play a key role in regulating skin immune responses. However, their specific involvement in inflammatory signaling triggered by solar ultraviolet (UV) radiation, which contributes to the development of cutaneous squamous cell carcinoma, is not yet fully understood. It is now known that environmental UV exposure activates Toll-Like Receptor 4 (TLR4), initiating inflammatory signaling pathways. Given the emerging evidence linking TLR4 to UV-induced skin damage, we recently tested the TLR4 antagonist resatorvid (TAK-242) in SKH-1 mouse models. Our findings show that topica

Sustained low disease activity during long-term mepolizumab treatment in FIP1L1::PDGFRA-negative hypereosinophilic syndrome: a multicenter real-world study.

BACKGROUND: Mepolizumab reduces relapses and glucocorticoid exposure in hypereosinophilic syndrome (HES), but evidence on long-term effectiveness remains limited. Growing consensus emphasizes sustained low disease activity over single-timepoint response in chronic inflammatory diseases. OBJECTIVE: To evaluate the long-term safety and effectiveness of mepolizumab in HES using a pragmatic definition of Eosinophilic Low Disease Activity State (EoLDAS). METHODS: We conducted a retrospective multicenter study of 67 patients with glucocorticoid-responsive FIP1L1::PDGFRA-negative HES treated with m

Pseudoaneurysm Following Stereotactic Breast Biopsy.

Stereotactic breast biopsy is a widely utilized and generally low-risk method for evaluating suspicious breast findings, though rare vascular complications, such as pseudoaneurysm, can occur. This case describes a patient who developed a delayed pseudoaneurysm approximately two weeks after an otherwise routine biopsy with benign pathology. The patient initially developed a post-procedural hematoma and subsequently presented with worsening pain, prompting further evaluation. Additional imaging revealed a pseudoaneurysm. Although first-line treatment with ultrasound-guided thrombin injection was

Rezpegaldesleukin and Regulatory T Cell Restoration in Immune-Mediated Skin Diseases.

Inflammatory skin diseases such as atopic dermatitis (AD), psoriasis, and alopecia areata (AA) are characterized by immune dysregulation involving impaired regulatory T cell (Treg) function and persistent activation of effector inflammatory pathways. Current targeted therapies largely suppress downstream inflammatory signaling rather than correcting upstream regulatory abnormalities. Rezpegaldesleukin (REZPEG; NKTR-358) is a first-in-class, Treg-selective interleukin-2 (IL-2) receptor agonist engineered to preferentially expand cluster of differentiation 4 positive (CD4+) FOXP3+ Tregs while mi

Irritant contact dermatitis: beyond a diagnosis of exclusion.

Irritant contact dermatitis (ICD) is the most common form of contact dermatitis and a leading cause of occupational skin disease, but remains underrecognized relative to allergic contact dermatitis (ACD). Direct chemical or physical injury to the epidermal barrier causes ICD, which can develop in anyone when exposure intensity or duration exceeds the skin's repair capacity. ICD is increasingly understood to involve more than barrier disruption: it is an innate immune-mediated inflammatory disorder involving keratinocyte-derived cytokines, neutrophil recruitment, C-C motif chemokine ligand 2-C-

Case Report: Triple-negative metaplastic breast carcinoma with primary resistance to neoadjuvant chemoimmunotherapy, refractory tumor necrosis-related fever, and rapid postoperative disease progression.

BACKGROUND: Metaplastic breast carcinoma (MpBC) is a rare and highly heterogeneous breast malignancy. Although most cases exhibit a triple-negative phenotype, MpBC differs from conventional triple-negative breast cancer in its biological behavior, treatment response, and metastatic pattern. Neoadjuvant chemoimmunotherapy has become an important strategy for high-risk early-stage triple-negative breast cancer; however, the evidence supporting its use in MpBC remains limited. CASE PRESENTATION: A 54-year-old woman was initially diagnosed with locally advanced triple-negative breast cancer of th

A novel stop-loss variant in ISG15 causes C-terminal dysfunction and impairs USP18-mediated negative feedback.

INTRODUCTION: ISG15 is a type I interferon (IFN-I)-inducible ubiquitin-like protein that restrains IFN-I signaling. Classical ISG15 deficiency is thought to cause disease mainly by disrupting USP18 stabilization, which leads to sustained IFN-I signaling and interferon-driven autoinflammation. Whether patient-derived C-terminal ISG15 variants can impair IFN-I negative feedback through altered C-terminal functional presentation has remained unclear. METHODS: We studied a Chinese boy with early-onset recurrent fever, severe ulcerative skin lesions, and elevated liver enzymes. Whole-exome sequenc

Cancer Survival at a Comprehensive Cancer Center Compared With Surveillance, Epidemiology, and End Results (SEER) Estimates.

OBJECTIVE: We compared outcomes across common cancer types at an NCI-designated Comprehensive Cancer Center (NCI-CCC) (City of Hope) vs the U.S. Surveillance, Epidemiology, and End Results (SEER) national database. Cancer Centers offer a comprehensive approach to prevention, diagnosis, and treatment through tertiary services and by providing coordinated, personalized care. METHODS: Propensity score matching (PSM) methods were employed in a 1:5 ratio for patients with incident cancers from 2004 to 2020 treated at COH vs reported in SEER. RESULTS: The COH cohort had significantly superior over

Effects of Supplementing the Diets of Mice With Different Concentrations of Conjugated Linoleic Acid on the Anti-Inflammatory Function of the Mammary Gland.

In this study, we evaluated the impact of the dietary addition of different concentrations of conjugated linoleic acid (CLA) on the antioxidant capacity and anti-inflammatory effects of serum and breast tissue in mice. Twenty-four ICR mice (BW = 37.79 ± 2.17 g) at 7 days of pregnancy were divided into four groups (n = 6 per group): a control (CN) group, which was fed a normal diet, and three groups fed a normal diet supplemented with 0.5% CLA, 1% CLA, or 2% CLA. Each mouse had free access to food and water. After 49 days of feeding, mice were injected with lipopolysaccharide (LPS) to induce in

Zinc ion-releasing immunomodulatory hydrogels for enhanced wound healing.

Effective wound healing requires a pro-regenerative immune microenvironment that supports tissue regeneration. Therefore, immunomodulatory biomaterials have gained increasing attention for wound healing. In this context, hydrogels have been widely investigated owing to their biocompatibility and structural similarity to the natural extracellular matrix. However, traditional hydrogels mainly provide physical protection and moisture retention while offering limited capacity for active immune modulation. To overcome this limitation, we fabricated zinc ion (Zn2+)-releasing immunomodulatory hydroge

Collagen-mediated biofilm formation and fibrillar organization in Staphylococcus epidermidis.

The symbiotic skin bacterium Staphylococcus epidermidis moisturizes human skin, contributes to protection against external pathogens, and supports innate immune stimulation. However, the interaction between collagen, a major skin protein, and S. epidermidis remains insufficiently explored. In this study, biofilm development was investigated using scanning electron microscopy with ionic liquid pretreatment, confocal laser scanning microscopy, biofilm quantification assays, and growth curve analysis. In the presence of collagen, cells adhered to the substrate, secreted microvesicles, formed web-

PAK4 exacerbates melanoma immunosuppression by promoting exosome release.

Melanoma remains a lethal malignancy with frequent resistance to immune checkpoint blockade therapy. Clinical evidence associates elevated PAK4 expression with poor response to anti-PD-1 treatment in melanoma patients. While tumor-derived exosomes are recognized as key immunosuppressive mediators, the role of PAK4 in regulating exosome secretion to induce immunosuppression remains incompletely understood. Here, we demonstrate that PAK4-mediated phosphorylation of Annexin-A2 (ANXA2) at Ser26 promotes its nuclear-to-cytoplasmic translocation, facilitating the formation of S100A10-ANXA2 heterotet

Migratory dermal γδ T cells determine the balance between lung immunity and tissue damage during helminth infection.

Many helminth parasites migrate through multiple host organs during infection, but how immune responses are relayed across these tissues is still poorly understood. To investigate the immune aspects of intertissue communication during infection, we established a percutaneous infection model with the tissue-migrating nematode Nippostrongylus brasiliensis. Profiling of the initial cutaneous immune response revealed that dermal γδ T cells become activated via an interleukin 1 receptor (IL-1R)-dependent mechanism, engage cell motility-associated transcriptional pathways, and leave the skin after p

Depletion of regulatory T cells breaks self-tolerance and induces epidermolysis bullosa acquisita in resistant C57BL/6J mice.

Pemphigoid diseases (PDs) are antibody-mediated autoimmune blistering diseases caused by loss of tolerance to basement membrane proteins such as type VII collagen (COL7), triggering pathogenic T and B cell responses. The contribution of regulatory T cells (Tregs) to disease resistance remains unclear. Using a COL7 immunization-induced PD mouse model, we show that transient Treg depletion breaks tolerance and induces skin blistering in otherwise PD-resistant H2b mice. Disease occurred predominantly in females and was associated with impaired Treg recovery, early skin inflammation, altered B-cel

Strategies for mitigating severe COVID-19 in patients with haematological malignancy during the omicron era.

Despite a decrease in disease severity since the emergence of the severe acute respiratory syndrome coronavirus 2 Omicron variant, coronavirus disease-2019 (COVID-19) continues to pose a significant threat to patients with haematological malignancies (HM). Although repeated booster vaccinations enhance protection against severe illnesses in immunocompromised individuals, they remain at heightened risk of adverse outcomes. This underscores the crucial need for effective pharmacologic strategies to prevent and treat infection. This review examines current strategies for preventing severe COVID-1

Gut microbiota and systemic sclerosis: Mechanistic insights and microbial therapies.

Systemic sclerosis (SSc) is an autoimmune disease characterized by progressive fibrosis in the skin and multiple organs. The cause and mechanism of SSc remain largely elusive, and there are limited therapeutic options available that effectively alter the progression of fibrosis associated with the disease. Emerging evidence indicates that the gut microbiota and its metabolites play a crucial regulatory role in maintaining immune homeostasis and modulating fibrotic responses in SSc. In this review, we provide a comprehensive summary of clinical evidence regarding gut microbiota dysbiosis in SSc
