Periodontitis, Psoriasis and Other Skin Disorders: Epidemiological and Mechanistic Evidence.

Periodontitis is a highly prevalent chronic inflammatory disease characterized by a dysbiotic biofilm-induced host immune dysregulation and sustained low-grade systemic inflammation. Psoriasis and other inflammatory skin disorders, including atopic dermatitis and alopecia, are immune-mediated conditions driven by complex interactions between genetic susceptibility, environmental triggers, microbial factors, and cytokine network activation. Mounting epidemiological and experimental evidence suggests a consistent association between periodontitis and psoriasis, with emerging links to other skin

JAK Inhibitor Safety in Atopic Dermatitis and Alopecia Areata: A 4.5-Year Real-World Retrospective Cohort Study.

INTRODUCTION: JAK inhibitors (JAKi) represent a novel therapeutic approach for treating the immune-mediated dermatologic diseases alopecia areata (AA) and atopic dermatitis (AD). JAKi have shown a favorable safety profile in controlled clinical trials; however, real-world safety data in routine dermatologic practice remain limited. The objective of this study is to evaluate the safety of JAKis (abrocitinib, baricitinib, upadacitinib, and ritlecitinib) in patients with AA or AD in a real-world setting. METHODS: We conducted a single-center retrospective observational study based on electronic

Mondor's disease of the breast in a patient living with sickle cell disease: a case report.

BACKGROUND: Mondor's disease of the breast is a rare benign clinical condition characterised by sclerosing thrombophlebitis occurring in subcutaneous veins of the breast. Typically, a tender, palpable cord in the affected quadrant of the breast is found on physical examination. Ultrasonography is the primary imaging modality for confirming the diagnosis following clinical suspicion. CASE PRESENTATION: This paper reviews the case of a 42 year old Ghanaian female living with sickle cell disease (genotype SS) who reported with a week's history of bilateral breast pain and lumps in her left breas

Serum Homocysteine as a Potential Dynamic Biomarker for Staging and Monitoring Progression in Alzheimer's Disease.

BACKGROUND: Diffuse unpatterned alopecia (DUPA) is an uncommon and incompletely characterized variant of androgenetic alopecia (AGA) marked by diffuse follicular miniaturization across the entire scalp, including the occipital region, which distinguishes it from classic patterned AGA and often precludes hair transplantation. SUMMARY: In this review of the available literature on DUPA, we discuss its clinical presentation, diagnostic features, competing pathophysiologic hypotheses, and management considerations. Current evidence suggests DUPA typically presents with diffuse thinning across all

Prevotella copri Exacerbates Psoriasis Through Keratinocyte-Neutrophil Crosstalk via NF-κB Activation.

Psoriasis is a chronic inflammatory skin disorder driven by systemic immune dysregulation and recent studies have implicated the gut-skin axis in its pathogenesis. However, the specific role of individual gut microbes remains poorly understood. In this study, the impact of Prevotella copri (P. copri) on psoriasis development was examined and the underlying mechanisms were investigated. In an imiquimod (IMQ)-induced mouse model, oral administration of P. copri exacerbated skin inflammation, increased epidermal thickness, promoted neutrophil infiltration and aggravated disease severity. In vitro

Upadacitinib for Severe Alopecia Areata in Adults and Adolescents: Two Phase 3 UP-AA Randomized Clinical Trials.

IMPORTANCE: Alopecia areata (AA) is a chronic, systemic immune-mediated disease characterized by nonscarring hair loss. Many patients do not experience improvements with available treatment options and only ritlecitinib is approved for adolescent patients; therefore, there remains a clear unmet need for additional and improved systemic therapies. OBJECTIVE: To evaluate the efficacy and safety of upadacitinib-an oral selective Janus kinase inhibitor-in adult and adolescent patients with severe AA. DESIGN, SETTING, AND PARTICIPANTS: This global clinical program included 2 parallel phase 3 repl

A rapidly involuting congenital haemangioma of the lower limb.

We report the case of a full-term male neonate born with a 5 cm vascular mass on the inner surface of the right thigh, diagnosed as a congenital haemangioma. Blood tests, including platelet count and coagulation parameters, were normal. Doppler ultrasound supported the diagnosis. Follow-up examinations at 1 week, 1 month and 2 months revealed progressive involution of the lesion, characterised by a growing peripheral pale halo, without ulceration or complications. Conservative management was adopted, and no interventions were required. This case highlights the natural regression of rapidly inv

Self-Healing Hydrogel-Enabled Modular Assembly of Bilayered Skin Construct for Hair Follicle Regeneration.

Alopecia remains a pervasive clinical challenge, largely owing to the limited ability of existing therapies to regenerate functional hair follicles. Hair follicle morphogenesis relies on precisely coordinated epithelial-mesenchymal interactions (EMIs) within a spatially defined epidermal-dermal niche, which is difficult to reconstruct using conventional tissue engineering approaches. We present a modular tissue engineering strategy using a self-healing N-carboxyethyl chitosan-oxidized dextran/collagen-genipin hydrogel. The hydrogel exhibits tunable mechanical properties, achieving a mechanical

Cutaneous Manifestations of Fertility Treatments: A Scoping Review.

Women are increasingly utilizing assisted reproductive technologies when pursuing pregnancy, particularly in vitro fertilization (IVF), which requires supraphysiologic hormonal exposures and intensive pharmacotherapy. As IVF use continues to rise, recognition of associated cutaneous adverse events has become increasingly important, with dermatologic manifestations reported in up to one-quarter of patients. These manifestations encompass a broad clinical spectrum, including progestogen hypersensitivity, alopecia, acneiform eruptions, injection-related cutaneous complications, urticaria and othe

A dual role of PIEZO2 in neuropathic pain: driving peripheral sensitization and enabling mechanical loading-induced analgesia.

BACKGROUND: Mechanotherapy, a form of mechanical loading (ML), effectively relieves tactile allodynia-associated neuropathic pain by targeting sensitization points stimulated by PIEZO2, a mechanosensitive ion channel. However, the molecular mechanisms underlying peripheral sensitization and ML-induced analgesia remain unclear. The study aimed to investigate the role of PIEZO2 in neuroinflammatory processes in a rat model of chronic compression injury (CCD) of dorsal root ganglia (DRG). METHODS: Mechanical allodynia was assessed via paw withdrawal threshold in male Sprague-Dawley rats. ML was

Clove oil in combination with flurbiprofen attenuated imiquimod-induced psoriasis-like skin inflammation in a mouse model: A mechanistic study.

Psoriasis is an immune-mediated chronic inflammatory skin disease characterized by scaly plaques, erythema, and thick skin. Psoriasis management is complicated and multifaceted as this disease is linked to serious systemic consequences like infections, psoriatic arthritis, cardiovascular diseases, and psychological complications. In the current study, a mice model of psoriasis was established by topical application of 5% imiquimod (IMQ) cream on the shaved dorsal skin and left ear pinna of mice for 7 days. The animals in treatment groups were treated topically with different concentrations of

From the Perspective of the Peripheral Sensory Nervous System: The Role of Vitamin D in Neurogenic Inflammatory Skin Disorders.

Vitamin D (VD) is a secosteroid hormone with critical physiologic roles in calcium homeostasis and bone mineralization. Recently, studies have uncovered VD's broader effects on immune system and nervous system function. VD receptors (VDRs) and enzymes responsible for the metabolism of VD have been discovered in dorsal root ganglion (DRG) neurons and nociceptive sensory neurons, implicating VD as a potential regulator of neurogenic inflammation. Neurogenic inflammation is a phenomenon in which sensory nerve fibers in the peripheral nervous system (PNS) are activated and release neuropeptides su

Macrophage-Driven Bidirectional Exacerbation in Psoriasis-Atherosclerosis Comorbidity: Insights From a Novel Mouse Model.

BACKGROUND: Psoriasis (PSO) and atherosclerosis (AS) are chronic inflammatory diseases that frequently coexist, with each condition exacerbating the other. However, the underlying mechanisms remain poorly understood, partly due to the lack of a suitable animal model that recapitulates this bidirectional comorbidity. METHODS: PSO-AS mouse models were constructed by combining high-fat diet (HFD)-induced AS with imiquimod (IMQ)-triggered psoriatic inflammation in ApoE-/- mice. PSO severity was assessed via skin manifestation, hematoxylin and eosin (H&E) staining, PSO area and severity index (PAS

A case of rheumatoid neutrophilic dermatosis complicated by neutrophilic tubulointerstitial nephritis.

As an extra-articular manifestation of rheumatoid arthritis (RA), neutrophilic dermatosis may occur. Here, we report a rare case of RA complicated by both neutrophilic dermatosis and neutrophilic tubulointerstitial nephritis. In addition to polyarthritis, the patient exhibited purpura and erythema with haemorrhagic crusts and ulceration accompanied by marked neutrophilic infiltration, extending from the fingers to the forearms and from both lower legs to the toes, as well as interstitial lung disease, leading to a diagnosis of RA. After the initiation of tocilizumab, the arthritis improved. Ho

Molecular diagnosis of an immature Ascaris lumbricoides infection with negative parasitological examinations.

Ascariasis is one of the most prevalent parasitic infections worldwide, particularly in developing countries, and is caused by Ascaris lumbricoides. Diagnosis can be challenging during the early stages of infection, especially when adult helminths have not yet become established in the intestine and routine parasitological examinations yield negative results. A 40-year-old female patient was referred to a hospital in South Tehran, Iran, with various clinical manifestations, including gastrointestinal symptoms, abdominal cramps, respiratory distress, cough, wheezing, localized skin itching, and

Novel insights into the role of microRNAs in atopic dermatitis and psoriasis: diagnostic and therapeutic aspects.

Atopic dermatitis (AD) and psoriasis are prevalent chronic inflammatory skin diseases with distinct clinical features and significant impacts on patients' quality of life. AD typically presents as recurrent eczema-like lesions accompanied by intense itching, whereas psoriasis is characterized by well-defined erythematous, scaly epidermal plaques. AD is associated with skin barrier dysfunction, allergen sensitization, and a type 2 immune response, while psoriasis involves hyperproliferation, abnormal differentiation of epidermal keratinocytes, and immune cell infiltration. However, the precise

NMJ-associated transcriptomic remodeling correlates with disease severity in juvenile dermatomyositis.

BACKGROUND: Juvenile dermatomyositis (JDM) is a rare systemic autoimmune disease primarily affecting children, with a female predominance. While muscle fiber inflammation has been extensively investigated, the contribution of the neuromuscular junction (NMJ) and its cellular microenvironment to JDM pathogenesis remains poorly understood. This study aimed to characterize NMJ-related gene expression patterns in JDM to identify potential pathogenic mechanisms and candidate biomarkers. METHODS: An integrated transcriptomic analysis was conducted using two publicly available microarray datasets co

Anti-transcription Intermediary Factor-1 Gamma Antibody-positive Dermatomyositis with Interstitial Lung Disease and Multisystem Involvement: A Case Report.

Dermatomyositis (DM) is a type of inflammatory myopathy of unknown origin that is marked by weakness in the proximal muscles, skin manifestations, and involvement of other systems. The existence of anti-transcription intermediary factor-1 gamma (anti-TIF1 γ) antibodies identifies a specific subgroup linked to cancer, interstitial lung disease (ILD), and unusual clinical presentations. We present a rare and complex case of DM positive for anti-TIF1 γ antibodies, which manifested as inflammatory myopathy, ILD, neuropathy, symptoms of sicca syndrome, and gastrointestinal involvement in an elderly
