TCRαβ+CD4-CD8- double negative T (TCRβ+DNT) cells are highly accumulated in human and murine lupus, although the origin and function of TCRβ+DNT cells remains largely elusive and controversy. Remarkably, conditional deletion of Egr2 in lymphocytes significantly reduced TCRβ+NK1.1- DNT cells, the dominant DNT type in B6/lpr mice. TCRβ+DNT cells of B6/lpr mice surprisingly had a lower expression of IFNγ, IL-17, and IL-10 compared to normal B6 mice, which was likely due to high CD138 expression in the TCRβ+DNT cells of B6/lpr mice. Conditionally deleting Egr2 in B6/lpr mice increased IFNγ and IL-

The absence of maternal immune rejection of a haploidentical fetus remains unexplained. We hypothesize that the presence of different HLA antigens on trophoblasts and maternal cells via killer cell immunoglobulin-like receptors (KIRs) provides two-way tuning (education, licensing) of decidual natural killer cells (dNK) with the appearance of alloreactive (HvG) and autoreactive (HvH) dNK. Disturbances in dNKHvG and dNKHvH representation may lead to abnormalities in placental development and pregnancy pathology. Our data show that recurrent pregnancy loss is not associated with specific HLA and

INTRODUCTION: Multiple myeloma (MM) is a heterogeneous plasma cell (PC) malignancy in both clinical presentation and genetic profile. Cytogenetic characterization, particularly by interphase fluorescence in situ hybridization (FISH), is critical for risk stratification and treatment decisions. FISH results depend on the PC infiltration rate in bone marrow (BM); therefore, CD138-positive immunoselection is required to overcome reduced FISH sensitivity due to low infiltration. We evaluated the performance of automated PC immunoselection compared to the manual method.
METHODS: We retrospectively

Cyclic neutropenia (CyN) is a rare haematological disorder most often caused by mutations in the neutrophil elastase gene (ELANE). We describe three novel ELANE mutations in CyN patients and link them to impaired neutrophil extracellular trap (NET) formation. ELANE sequencing was performed using the Sanger method, and AlphaFold2 was used to visualize the impact of mutations on the structure of neutrophil elastase (NE). NET formation was assessed by colocalization of MPO, NE, and DNA using fluorescence microscopy, and free DNA was quantified fluorometrically. We identified two missense substitu

Autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy (APECED) is a rare monogenic autoimmune disease caused by loss-of-function mutations in the Autoimmune regulator gene and the subsequent impairment of negative selection in the thymus. Previous studies have identified many T cell abnormalities in APECED patients, with increased signs of activation and failure of regulation. These findings, however, have been demonstrated using traditional flow cytometry studies, prone to human misinterpretation of multidimensional data. Here we present an unsupervised flow cytometry analysis of CD4

INTRODUCTION: Combined immunodeficiencies (CIDs) are a severe class of inborn errors of immunity characterized by defective T cell development and function, often accompanied by impaired humoral and natural killer (NK) cell responses. Despite their shared clinical classification, the immunological heterogeneity within CIDs remains incompletely understood. This study aims to characterize the immune cell landscape in CID patients caused by disease-causing mutations in different genes to identify immunophenotypic patterns.
METHODS: We analyzed peripheral blood immune cells from four CID patients

BACKGROUND: Chronic lymphocytic leukemia (CLL) is a hematological malignancy with the accumulation of mature, monoclonal B lymphocytes. CLL cells can escape the host's immune response by inducing T-cell exhaustion. This occurs by upregulation of inhibitory checkpoint receptors, such as T-cell immunoglobulin and mucin domain-containing molecule-3 (TIM-3). Galectin-9 (Gal-9) is a soluble ligand for TIM-3. Its interaction with TIM-3 can induce T-cell exhaustion in various cancers. Therefore, targeting these inhibitory pathways may be a potential therapy for CLL.

This study investigated the effects of Dalbergiae Odoriferae Lignum flavonoids(DOLF) on microcirculatory disorders through the regulation of neutrophil extracellular traps(NETs). Sixty SPF-grade male Sprague-Dawley(SD) rats were randomly divided into a sham operation group, a model group, a nicorandil group(1 mg·kg~(-1)), and low-, medium-, and high-dose DOLF groups(1, 2, and 4 g·kg~(-1), respectively), and were administered the corresponding treatments by gavage for 7 consecutive days. A model of microcirculatory disorders was established by injecting polyethylene microspheres into the left v

BACKGROUND: Acute lymphoblastic leukaemia (ALL) in children is often associated with immune dysfunction, including alterations in regulatory T cells (Treg), impaired T-cell activation, and disrupted natural killer (NK) cell function. ALL comprises immunophenotypically heterogeneous entities, primarily B-cell ALL (B-ALL) and T-cell ALL (T-ALL). However, the dynamics of Treg subsets and their relationship with broader immune alterations across treatment stages remain insufficiently characterised.
METHODS: In this prospective case-control study, peripheral blood samples were analysed from 31 chi

BACKGROUND: Heart failure with preserved ejection fraction (HFpEF) arises from chronic cardiometabolic and vascular stress and is increasingly recognized as an inflammatory syndrome with immune dysregulation. Regulatory T cells (Tregs) are critical modulators of cardiovascular inflammation, yet the mechanisms driving Treg dysfunction in HFpEF remain poorly defined. stromal interaction molecule 1 (STIM1)-dependent calcium signaling is a key stress-responsive pathway in immune cells; however, its role in Treg maladaptation during HFpEF remains unknown.
METHODS: Circulating Tregs from patients w

Our group discovered a mutation in a French-Canadian family causing a severe and early-onset form of emphysema. The heterozygote mutation located in the PTPN6 gene causes an alanine to threonine switch near the catalytic site of SHP-1, leading to a reduction of its enzymatic activity. In this study, we aimed to characterize the respiratory and immune abnormalities caused by this mutation in human carriers (PTPN6Ala455Thr) and mice carrying the equivalent mutation (Ptpn6Ala457Thr/+). Ptpn6Ala457Thr/+ mice spontaneously developed pulmonary tertiary lymphoid tissues (TLTs). They also exhibited a

Metabolic syndrome (MeS) is a major risk factor for cardiovascular disease and is characterized by chronic low-grade inflammation, immune dysregulation, and metabolic abnormalities. However, the molecular mechanisms linking MeS to diabetic coronary artery disease (DMCAD) remain incompletely understood. Publicly available peripheral blood mononuclear cell (PBMC) transcriptomic datasets of MeS and DMCAD were analyzed using an integrative bioinformatics approach. Differentially expressed genes (DEGs) were identified using the limma package, followed by functional enrichment, protein-protein inter

Postzygotic somatic variants affecting the Ras/MAPK signaling pathway can cause mosaic RASopathies, characterized by craniofacial abnormalities, cardiac defects, growth impairment, and various cutaneous manifestations in localized areas of the body. Herein, we present a rare case of mosaic RASopathy caused by a somatic HRAS variant. A 52-year-old Japanese patient visited our department because of skin metastasis of malignant melanoma. Besides, he had a sebaceous nevus on the forehead, linear epidermal nevus on the trunk along with the Blaschko line, and focal and linear non-epidermolytic palmo

Alpha-mannosidosis is a rare lysosomal storage disease caused by a deficiency of the enzyme alpha-mannosidase. It manifests as a continuous spectrum of signs and symptoms characterized by dysmorphic features, skeletal abnormalities, delayed psychomotor and speech development, impaired hearing, and psychiatric involvement. When suspected, alpha-mannosidosis must be confirmed by biochemical and molecular testing, namely, assessment of blood levels of alpha-mannosidase in leukocytes or fibroblasts and Sanger or next-generation sequencing of the MAN2B1 gene. The disease must be diagnosed and treat
