Icaritin Attenuates HSC Activation by Down-regulating the HIF-1α and TGF-β/Smad Signaling Pathways to Ameliorate Liver Fibrosis.

INTRODUCTION: Icaritin is a bioactive flavonol isolated from the Chinese medicinal herb Epimedium. The comprehensive understanding of antifibrotic effects and associated molecular mechanisms of icaritin remains incomplete. This study aims to explore the protective effects of icaritin against liver fibrosis and to further elucidate the mechanisms involved. METHODS: Human hepatic stellate LX-2 cells stimulated with TGF-β1 and a carbon tetrachloride (CCl4)-induced liver fibrosis mouse model were employed. In vitro assays were carried out to evaluate collagen type I (COL I) and α-smooth muscle ac

Low dose IL-2 administration prevents but fails to treat experimental autoimmune anti-myeloperoxidase glomerulonephritis.

BACKGROUND AND HYPOTHESIS: Anti-neutrophil cytoplasmic antibody (ANCA) associated vasculitis (AAV) is a major cause of glomerulonephritis (GN) and kidney failure and current therapies are often limited by significant toxicity and limitations. Regulatory T cells (Tregs) play a crucial role in maintaining immune tolerance, and their dysfunction has been implicated in AAV pathogenesis. Low dose interleukin-2 (IL-2) has emerged as a promising strategy to selectively expand Tregs and restore immune balance in several autoimmune diseases. However, low dose IL-2's therapeutic potential in AAV remains

Hepatic myelopathy improved by embolization of spontaneous portosystemic shunt and partial splenic artery embolization ‒ Literature review.

Hepatic myelopathy (HM) is a serious and debilitating complication associated with chronic liver disease. Currently, there is no definitive treatment; however, it is widely accepted that early liver transplantation and stent-restricted surgery may alleviate HM to some extent. We report a case of an adult male with a history of hepatitis B cirrhosis who developed a complex spontaneous shunt following portal vein occlusion, which resulted in recurrent hepatic encephalopathy and was later complicated by HM. We successfully relieved HM in this patient through embolization of the inferior mesenteri

Protection Against Type 1 Diabetes Development in Mice With 4E-BP2 Deletion.

UNLABELLED: Type 1 diabetes (T1D) is an autoimmune disease characterized by β-cell destruction promoted by autoreactive T cells. Eukaryotic translation initiation factor 4E (eIF4E)–binding protein 1 (4E-BP1) and 4E-BP2 are translational repressors and downstream targets of mammalian target of rapamycin complex 1 (mTORC1). Activation of the 4E-BP2/eIF4E pathway by 4E-BP2 deletion promotes translation initiation, inducing β-cell expansion and proliferation and regulating adaptive immunity. However, the involvement of 4E-BP2 in T1D remains unexplored. This study aimed to determine the role of 4E-

Evaluation of trained immunity as a potential strategy for a universal Streptococcus suis vaccine using immunogenic proteins in a murine model.

BACKGROUND: Universal vaccine against Streptococcus suis (S. suis) remains a challenge due to the high number of serotypes/strains, the absence of cross-protection among them and the existence of different immune escape strategies. In this study, the immunomodulatory potential of live and inactivated porcine derived Bacillus Calmette-Guérin (dpB) in combination with specific immunogenic proteins S. suis is evaluated in murine model. RESULTS: Results revealed different immune responses depending on dpB formulation. Live-dpB administered intravenously induced consistent results with trained imm

Brucella abortus infection exploits ZDHHC3-mediated STAT3 palmitoylation to regulate host responses and promote persistence.

Brucella abortus, an intracellular bacterium, employs intricate mechanisms to manipulate host signaling for persistence. This study investigates the role of STAT3 palmitoylation in B. abortus-infected macrophages. We demonstrate that B. abortus infection induces STAT3 palmitoylation, which is critical for its membrane recruitment. Among zinc finger DHHC-type palmitoyl acyltransferases (ZDHHC) family members, ZDHHC3 specifically mediates STAT3 palmitoylation in infected macrophages. ZDHHC3-induced STAT3 palmitoylation promotes STAT3 phosphorylation at Y705, independently of IL-6. Functionally,

The Spleen Promotes Liver Sinusoidal Endothelial Cell Angiogenesis by Stimulating the TGFβ/Smad2/3 Pathway Through GDF15.

BACKGROUND AND AIMS: Liver sinusoidal endothelial cells (LSECs) are crucial nonparenchymal cells involved in liver fibrosis progression. Liver fibrosis develops from various chronic liver diseases, with pathological progression associated with the spleen. However, the precise mechanism by which the spleen influences LSECs during fibrosis is not fully understood. This study investigated the spleen's effect on LSEC angiogenesis and its underlying mechanisms using animal and cellular experiments. METHODS: Splenectomy or sham operations were performed on carbon tetrachloride (CCl4)-induced liver

Harnessing the microbiome to regulate myeloid TREM2 expression and innate immune responses.

The composition of the gastrointestinal microbiome is correlated with numerous immune-mediated systemic diseases, but underlying mechanisms remain unclear. In murine studies, we recently identified microbiome Bacteroidota-derived bacterial molecules, serine-glycine lipodipeptides (S/G lipids), as mediators of microbiome-systemic innate immune system crosstalk. By altering microbiome production of S/G lipids, we documented that proinflammatory responses of splenic monocytes could be regulated. Transcriptomic analysis revealed that this regulation occurred by modulating the mRNA levels of inhibi

Radiological Approach to Splenomegaly: Etiologies, Pathophysiologies, and Diagnostic Strategies.

The spleen is frequently referred to as the "silent and forgotten" organ of the abdomen by clinicians and radiologists, primarily because of the secondary nature of most splenic pathologies and their relative rarity compared with hepatic diseases. Nevertheless, the term splenomegaly frequently appears in radiological reports. Although it is often reported in the context of a known underlying cause, it is also commonly used as a purely descriptive term without reference to the underlying etiology. Although splenomegaly may occasionally be idiopathic, it commonly represents an underlying patholo

Repeated administration of cannabidiol decreases splenic lymphocyte subset numbers in rats.

OBJECTIVE: Cannabidiol (CBD) administration (5 mg/kg) in healthy rats has been shown to significantly decrease lymphocyte subset numbers in peripheral blood without involvement of natural killer cells. The aim was to evaluate whether lymphocyte numbers also decrease in the spleen. INTRODUCTION: CBD, the major non-psychotropic compound of Cannabis sp., is an effective treatment for inflammatory and autoimmune diseases with various anti-tumor effects, but the mechanisms of its long-term actions in vivo remain unclear. METHODS: To examine the effects of CBD on lymphocyte subsets in the spleen a

Immunomodulatory effect of mesenchymal stromal cell overexpressing HLA-G1 in cell-based therapy for myocardial infarction.

BACKGROUND: Our previous study revealed that intravenous administration of mesenchymal stromal cells (MSCs) increases local cell engraftment and improves heart function. This study aims to investigate whether MSCs overexpressing HLA-G1 have further increased local transplanted cells engraftment and improved heart function. METHODS: The mice were intravenously administered saline or human umbilical cord blood-derived MSCs (hUCB-MSCs) 7 days before myocardial infarction (MI) induction. Then, the MI mouse model was established by ligating the left anterior descending coronary artery. The mice we

Intraparenchymal B cell therapy protects neurological function and modulates local neuroinflammation in experimental hemorrhagic stroke.

Intracerebral hemorrhage (ICH) is a devastating subtype of stroke, with a 5-year mortality of up to 70 %. Disease-modifying treatments to improve neurological outcomes in ICH survivors represent a critical unmet need. Neuroinflammation contributes significantly to ongoing secondary brain injury and long-term morbidity in ICH. We and others have shown that B cells are potent modulators of the inflammatory response, capable of acquiring a regulatory phenotype within injured microenvironments. Here, we investigated the effects of a single intraparenchymal application of mature naïve splenic B lym

Infarct-to-spleen volume ratio as a novel volumetric predictor of splenectomy in splenic infarction.

BACKGROUND: Splenic infarction is a rare and often underrecognized condition with diverse etiologies and variable clinical presentations. While most cases are managed conservatively, identifying patients who may require surgical intervention remains a clinical challenge. This study aimed to evaluate the infarct-to-spleen volume ratio (ISR) as a radiologic predictor for splenectomy in patients with splenic infarction and to propose a risk stratification model incorporating ISR and fever. METHODS: In this retrospective, two-center cross-sectional study, 236 patients diagnosed with splenic infarc

Prostate cancer with incidental thoracic splenosis: a diagnostic challenge unveiled by 18 F-PSMA PET/CT imaging.

BACKGROUND: Thoracic splenosis is a rare condition caused by autotransplantation of splenic tissue into the thoracic cavity following thoracoabdominal trauma or surgery. Due to its radiological presentation, this condition often mimics malignancies, leading to potential misdiagnosis and unnecessary invasive procedures. A thorough clinical history, including prior splenic trauma or splenectomy, is therefore critical to raise suspicion. When imaging is inconclusive, non-invasive nuclear medicine techniques such as Tc-99 m heat-damaged erythrocyte scintigraphy represent the diagnostic gold standa

Imperforate tricuspid valve with severe hypoplasia associated with an atypical vascular ring in the set of left isomerism. First case report.

Bodily isomerism, often referred to as "Heterotaxy," is the clinical entity in which the thoracic organs are themselves mirror-imaged in the same individual. The thoracic isomerism can be right or left. The left thoracic isomerism always has both atria with their atrial appendages of left morphology and frequently a mixed atrioventricular connection. Usually, there are two well-developed ventricles; however, in some cases, there may be univentricular physiology. In the set of left thoracic isomerism, no case of imperforate tricuspid valve with severe hypoplasia associated with a vascular ring

[Diagnostics and stage-conform treatment of splenic injuries].

Splenic injuries often represent severe internal injuries in cases of abdominal trauma. Contrast-enhanced computed tomography (CT) and laboratory diagnostics play a particularly important role in the diagnostics. Traumatic splenic injuries are classified according to the American Association for the Surgery of Trauma (AAST) and World Society of Emergency Surgery (WSES) criteria. The degree of injury and the hemodynamic and clinical situation of the patient are decisive factors in the choice of treatment. Nonoperative management (NOM) is used in cases of hemodynamic stability and minor splenic
