Wastewater surveillance (WWS) is a powerful, population-level approach to infectious disease monitoring that complements traditional clinical surveillance, which often under-detects asymptomatic or unreported cases. This study presents validated methods for Salmonella detection in wastewater, including sample concentration using PEG precipitation, nucleic acid extraction with magnetic beads, and quantitative PCR targeting. When WWS for Salmonella was implemented across Oklahoma from 2021 to 2022, results showed seasonal patterns in transmission, provided a 1-week lead time over clinical case r

The rise of multidrug resistance (MDR) poses a significant challenge to global health, reducing the effectiveness of conventional antibiotics. The World Health Organization (WHO) classifies Salmonella as a priority pathogen due to its global burden in foodborne disease and the escalating threat of MDR strains. Bacteriophages (phages) are gaining attention as alternatives to traditional antibiotics. Their strict host specificity has potential applications in both therapeutic and food safety contexts. However, the first step toward practical application is the successful isolation of phages from

The stable expression of fluorescent reporters in cell lines is crucial for the analysis of host-pathogen interactions. Here, an optimized protocol for generating a HeLa cell line that indicates membrane damage, including lentiviral particle production, transduction, functional tests, and fluorescence-activated sorting (FACS) of HeLa cells, is described. In addition, an image-based segmentation protocol is described for quantifying membrane damage to Salmonella-containing vacuoles (SCV) and the presence of cytosolic Salmonella enterica. This enables the analysis of virulence factors that affec

Polymerase chain reaction (PCR) is a molecular-based technology that has revolutionized diagnostics and characterization of pathogens, thus affecting how we understand the disease landscape. This technology has been found amenable to application on various strategies for management and control of infectious diseases. The main advantage with PCR technologies, when applied optimally, is the high sensitivity, specificity, efficiency, and short-turn-around time for results, thus rendering the strategy attractive to researchers in infectious diseases and public health. In this chapter, we describe

Biochemical testing plays a crucial role in the detection and confirmation of Salmonella spp. Since the cultural characteristics of Salmonella often resemble those of other enteric pathogens, differentiation based solely on growth features can be challenging. Biochemical tests allow for accurate identification by exploiting the distinct metabolic traits of Salmonella. In this study, we described three key tests, including methyl red, hydrogen sulfide production, and urease production, which are particularly important for the confirmation of Salmonella.

The conventional method for Salmonella detection, as recommended by ISO 6579-1:2017(E), involves four sequential steps: preenrichment, selective enrichment, plating on selective media, and confirmation. The second step consists of Selective enrichment, which is performed using Muller-Kauffmann tetrathionate-novobiocin broth, Rappaport-Vassiliadis Soya Peptone Broth, or Modified Semisolid Rappaport-Vassiliadis (MSRV) agar. MSRV agar, widely used in food industry testing, exploits Salmonella motility to facilitate selective isolation by forming characteristic opaque halos. Its use reduces proces

Salmonella is an important zoonotic pathogen that is widely distributed in livestock, poultry, and other animals. Salmonella contamination not only affects the healthy development of the livestock industry but also poses a serious threat to public health. Therefore, establishing detection methods capable of simultaneously identifying different Salmonella serotypes is crucial. Among current detection methods for Salmonella, immunological detection methods using MAbs offer high throughput, shorter processing times, and enhanced specificity, making them useful for rapid Salmonella detection. The

Salmonella infection increases the risk of chronic diseases, e.g., inflammatory bowel diseases, colon cancer, and gallbladder cancer. However, the mechanisms leading to the development of these conditions are still not completely understood. Here, we describe the methods to establish in vivo mouse models to study Salmonella-induced chronic infection and colon cancer. These models will allow us to investigate the initiation and progression of diseases.

Whole-genome sequencing (WGS) provides high-resolution evidence for investigating bacterial transmission during outbreaks of invasive non-typhoidal Salmonella (iNTS). Here we describe a reproducible workflow for genomic tracing of transmission dynamics using short-read sequencing data. The protocol includes read quality assessment, genome assembly, reference-based single nucleotide polymorphism (SNP) calling, core-genome alignment, pairwise SNP distance analysis, phylogenetic reconstruction, and characterization of mobile genetic elements (MGEs) and antimicrobial resistance (AMR) determinants.

BACKGROUND: In low-income and middle-income countries, where typhoid fever remains a major public health problem, WHO recommends the use of typhoid conjugate vaccine (TCV). Here, we report vaccine effectiveness up to 8 years following a single dose of TCV in Nepal.
METHODS: TyVOID was a prospective cohort study that extended the follow-up of children enrolled in a phase 3, double-blind, randomised controlled trial in Lalitpur, Nepal (TyVAC; Nov 20, 2017, to Oct 20, 2021). Children aged 9 months to 15 years were randomly assigned (1:1) to receive Vi-tetanus toxoid conjugate vaccine (Vi-TT) or

Salmonella enterica Newport is one of the most common serotypes linked to fresh produce-associated salmonellosis outbreaks in the United States. In this study, we examined the genetic diversity and epidemiological relevance of 20 Newport strains recovered from Salinas Valley watersheds between 2012 and 2016. Multilocus Sequence Typing (MLST) identified two sequence types (STs), with ST118 representing 70% of isolates and ST45 comprising the remainder. Core-genome MLST (cgMLST) resolved 12 cgSTs, with cgST132506 being the most common. Phenotypic analyses revealed lineage-specific traits, includ

Salmonella enterica serovar Infantis (S. Infantis) is an increasingly important food-borne pathogen, and detection in broiler flocks via the standardized EN/ISO 6579-1:2017 is of utmost importance. Phenotypic characteristics, such as the production of hydrogen sulfide (H2S) and swarming behavior, are essential for detection. However, the occurrence of atypical variants lacking such phenotypic characteristics bears a risk to public health. In this study, we characterized an Austrian human-derived S. Infantis strain showing, in addition to the normal colony phenotype (NCP), also an atypical colo

BACKGROUND: Salmonella enterica serotype Napoli (S Napoli), a non-typhoidal Salmonella serotype that has been emerging in Italy, Switzerland, and France since the mid-1990s, has an unusual epidemiological profile that suggests a role for the environment in the transmission of these bacteria. The genomics of S Napoli has been inadequately characterised; we, therefore, aimed to provide a robust genomic framework for S Napoli surveillance.
METHODS: In this genomic epidemiology study, we performed short-read sequencing of 1145 S Napoli isolates from national surveillance in four European countrie

Rapid qualitative assessments and community feedback mechanisms are increasingly recognized as core components of risk communication and community engagement in public health emergencies, yet few documented examples exist of how this evidence has directly influenced outbreak response decision-making. This case study examines the 2023-2024 cholera outbreak in Zambia, analyzing how social and behavioral data generated through multiple rounds of rapid qualitative assessment, alongside national community feedback mechanisms, informed multisectoral response activities. Drawing on document review, c

Consolation behavior is defined as an increase in affiliative contact toward a distressed individual and is associated with the physical and emotional well-being of social species. Allogrooming a distressed individual represents a primary form of consolation behavior and is considered as a complex type of empathy. However, the neural mechanisms underlying allogrooming remain poorly understood. In this study, using adult male and female monogamous mandarin voles (Lasiopodomys mandarinus, approximately three-month-old), we demonstrate that individuals exhibited significantly more allogrooming to

BACKGROUND: Effective cholera outbreak response requires accurate bedside rapid diagnostic tests (RDTs) because of poor laboratory access. Formative studies suggest that cholera diagnostics have multiple vulnerabilities, including target degradation by virulent bacteriophage (phage) specific to Vibrio cholerae and antibiotics. Our study objectives were to evaluate the performance of RDTs as a function of these antimicrobials to improve deployment and guide future diagnostic development.
METHODS: In this prospective longitudinal study, participants were enrolled at seven hospitals across Bangl

BACKGROUND: LepA is one of the most conserved proteins and is found in nearly all bacteria as well as in eukaryotes. Mutation of lepA increases antibiotic tolerance in Escherichia coli, Salmonella, and rifampicin-resistant clinical mycobacterial isolates. However, the physiological function and its role in virulence of LepA remain elusive, particularly considering that LepA is non-essential under laboratory conditions.
METHODS: We investigated the lepA gene of Salmonella Typhimurium for its role in bacterial invasion of host cells. Invasion assays using lepA mutants were performed in macropha

Salmonella Minnesota ST548 has emerged as an important serovar in the poultry industry and has been detected in food products. Five nonclonal S. Minnesota isolates from poultry in southeastern Brazil were characterized using whole-genome sequencing (WGS), and 263 database sequences were analyzed for virulence genes, antimicrobial resistance genes, and phylogenetic relationships. Among the analyzed sequences, 191 multidrug-resistant (MDR) strains carried antimicrobial resistance genes (ARGs) conferring resistance to aminoglycosides (aac(6')-Iaa, aph(3')-Ia and aadA1), quinolones (qnrB19), tetra
