ARHGAP12 suppresses F-actin assembly to control epithelial tight junction mechanics and paracellular leak pathway permeability.

Tight junctions (TJs) control the paracellular transport of ions, solutes, and macromolecules across epithelial barriers. There is evidence that claudin-based ion transport (the pore pathway) and the paracellular transport of macromolecules (the leak pathway) are controlled independently. However, how leak pathway flux is regulated is unclear. Here, we have identified the Cdc42/Rac GTPase-activating protein ARHGAP12 as a specific activator of the leak pathway. ARHGAP12 is recruited to TJs via an interaction between its Src homology (SH3) domain and the TJ protein ZO-2 to suppress N-WASP-mediat

How I treat Wiskott-Aldrich syndrome.

Wiskott-Aldrich syndrome (WAS) is a rare X-linked disorder, characterized by thrombocytopenia, eczema, recurrent infections, autoimmunity, and malignancy. Here, we discuss current conservative and definitive approaches to treating WAS, based on recently published evidence. Disease severity in WAS is highly variable. Recent studies confirm that the probability of disease progression depends on the type of genetic variant, supporting early diagnosis and tailored treatment strategies. Milder cases, historically termed X-linked thrombocytopenia (XLT), received supportive care, whereas severe cases

Human Papillomavirus Type 16 Stimulates WAVE1- and WAVE2-Dependent Actin Protrusions for Endocytic Entry.

Human papillomavirus type 16 (HPV16) is an etiological agent of human cancers that requires endocytosis to initiate infection. HPV16 entry into epithelial cells occurs through a non-canonical endocytic pathway that is actin-driven, but it is not well understood how HPV16-cell surface interactions trigger actin reorganization in a way that facilitates entry. This study provides evidence that Wiskott-Aldrich syndrome protein family verprolin-homologous proteins 1 and 2 (WAVE1 and WAVE2) are molecular mediators of actin protrusions that occur at the cellular surface upon HPV addition to cells, an

Competitive binding of actin and SH3 domains at proline-rich regions of Las17/WASP regulates actin polymerisation.

Eukaryotic actin filaments bind factors that regulate their assembly and disassembly creating a self-organising system, the actin cytoskeleton. Despite extensive knowledge of signals that modulate actin organisation, significant gaps remain in our understanding of spatiotemporal regulation of de novo filament initiation. Yeast Las17/WASP is essential for actin polymerisation initiation supporting membrane invagination in Saccharomyces cerevisiae endocytosis and therefore its tight regulation is critical. The adaptor protein Sla1 inhibits Las17 but mechanisms underpinning Las17 activation remai

Functional divergence of plant SCAR/WAVE proteins is determined by intrinsically disordered regions.

Dynamic actin cytoskeleton reorganization enables plant developmental processes requiring polarized transport such as root hair and leaf trichome formation. The SCAR/WAVE complex plays a crucial role in regulating these dynamics through ARP2/3-mediated actin branching. SCAR/WAVE genes occur as small families across a wide range of plant species, but whether and how they fulfill different functions remains unclear. We use a systematic chimera approach to define the differential functionality of two closely related Medicago truncatula SCAR proteins in plant development. We show that SCAR/WAVE co

Efficacy and safety of romiplostim and eltrombopag in management of thrombocytopenia in Wiskott-Aldrich syndrome patients.

Wiskott-Aldrich syndrome (WAS) is a life-threatening inborn error of immunity associated with bleeding risk due to profound thrombocytopenia. We previously demonstrated that romiplostim is effective for thrombocytopenia treatment in 60% of WAS patients. In this prospective trial, 28 WAS patients (aged 0-17 years) were treated with thrombopoietin receptor agonists (TPO-RAs) to evaluate the comparative efficacy and safety of romiplostim versus eltrombopag and to assess the benefit of switching these molecules in individual subjects. We demonstrate that both drugs are effective, with the probabil

Bioequivalent Letermovir Usage for Prophylaxis in Haploidentical Stem Cell Transplantation at High Risk of CMV Reactivation: A Report of Two Cases From India.

Cytomegalovirus (CMV) reactivation is a major complication in allogeneic hematopoietic stem cell transplantation (HSCT), increasing nonrelapse mortality (NRM) and transplant-related complications. Letermovir, a CMV deoxyribonucleic acid (DNA) terminase inhibitor, has demonstrated efficacy in reducing CMV reactivation without the hematologic toxicity associated with traditional antivirals. We report two HSCT cases, one with a T-cell receptor (TCR) alpha/beta-depleted haploidentical transplant and another haploidentical (6/10 HLA-matched) stem cell transplant, both at significant risk for CMV re

Disruption of Retriever Function Impacts Retrograde Trafficking From Endosomes.

Within endosomes cargo proteins are sorted and packaged into endosomal-transport carriers (ETCs) enabling their delivery to other intracellular compartments. Retromer, a conserved multimeric protein complex, has defined functions in sorting cargo mediating formation of ETCs for both retrograde trafficking back to the trans-Golgi network (TGN) and recycling of cargo to the cell surface. Recent studies have identified the retriever complex, which is structurally like retromer, that also can function in the recycling of cargo from endosomes. However, retriever's function in retrograde trafficking

Unveiling Oncogenic Power of WAVE1 in Bladder Cancer Progression.

WAVE1 (Wiskott-Aldrich syndrome verprolin-homologous protein family member 1), a gene associated with the Wiskott-Aldrich syndrome, has been identified as an oncogene with high expression in various tumors and has been linked to metastatic conditions. Nevertheless, expression pattern of WAVE1 and its function in bladder cancer (BC) remain unknown. This study aimed to uncover the effect of WAVE1 on migration and metastasis in BC. In this study, four different human urinary BC cell lines (T24, RT4, J82, and BIU87) were examined using immunohistochemical and Western blot assays to assess the patt

The tumor suppressor RASSF8: A WAVE interaction partner controlling migration and cohesion of invasive border cells in Drosophila.

Collective cell migration is a key driver of tissue morphogenesis and cancer invasion. Here, we identified the tumor suppressor Ras association domain-containing protein 8 (RASSF8) as a WAVE interactor required for border cell migration. RASSF8 colocalizes with F-actin and cell adhesion molecules at border cell- border cell contacts. Loss of RASSF8 function results in border cell cohesion defects, a phenotype associated with changes in the localization of the Echinoid (Ed) and Coracle (Cora). Cell-type-specific RNA interference (RNAi) experiments suggest that cohesion defects are caused by cha

Unraveling a novel missense mutation (c.A248C) in Wiskott-Aldrich syndrome gene by whole exome sequencing: Insights from dynamic simulation, molecular docking and in-silico studies.

Wiskott-Aldrich syndrome (WAS) is an uncommon genetic condition that is inherited via the X chromosome and is caused by mutations in the Wiskott-Aldrich syndrome (WAS) gene. This disease is defined by the presence of persistent thrombocytopenia, eczema, and immunological dysfunction. It is also linked to a higher likelihood of developing autoimmunity and cancer. Using whole-exome sequencing (WES) on the genomic DNA of a family that had two affected members, we detected a new missense hemizygous mutation (c.A248C, Y83S) in the Wiskott-Aldrich syndrome gene (WAS, Xp11.23). The tyrosine residue a

Long-term outcome in Wiskott-Aldrich syndrome and X-linked thrombocytopenia patients: an observational -prospective multi-center study of the Italian Primary Immune Deficiency Network (IPINET).

BACKGROUND: Wiskott-Aldrich Syndrome (WAS) is characterized by eczema, infections, and severe bleeding, but may also include autoimmunity and malignancy. Subjects with X-linked thrombocytopenia (XLT) can display a mild phenotype, although severe complications may occur at any age. WAS and XLT are caused by mutations of the WAS gene. However, retrospective studies have shown conflicting results about their genotype-phenotype correlation and their relative risk of complications. METHODS: To evaluate the outcome of patients with WAS or XLT, since January 2004, patients with identified WAS mutati

Ena/VASP-EVH1 inhibition prevents chemotaxis and metastasis by blocking the EVH1-WAVE2 interaction.

Cancer therapy would benefit from suppressing cancer cell motility in the process of metastasis. Such directed cell migration relies on the propulsive force established by the filamentous actin network within lamellipodia. Proteins of the Ena/VASP family and the WAVE regulatory complex orchestrate lamellar protrusions and therefore provide promising targets for pharmacological interventions. Here, we report a cross-talk between Ena/VASP proteins and WAVE2 that is important for cancer cell extravasation. Mutating the EVH1 domain recognition motif in WAVE2 abrogates chemotaxis of triple-negative

Phenotypic screens for SIRPA expression reveal RAB21 as a general regulator of macrophage surface identity.

Factors influencing the macrophage surfaceome define macrophage identity and behavior. Here, we use genome-wide phenotypic screens to identify genes affecting the accessibility and surface expression of macrophage signal regulatory protein alpha (SIRPA). Our data are consistent with previous evidence but also implicate glutaminyl-peptide cyclotransferase-like (QPCTL) in cis CD47-SIRPA interactions. We also identify endolysosomal factors encoded by Ras-associated binding protein 21 (RAB21) and members of the CCC (COMMD/CCDC22/CCDC93) and Wiskott-Aldrich syndrome protein and SCAR homolog (WASH)

Expression and Regulatory Roles of SKAP2 and Cortactin in Mouse Ovarian Tissue and Oocyte Maturation.

Oocyte maturation is a critical phase in mammalian reproduction. Src kinase-associated phosphoprotein 2 (SKAP2) and cortactin, both cytoskeletal regulatory proteins that play pivotal roles in cell motility and signal transduction. However, their specific functions and mechanisms during oocyte maturation are currently unknown. This study revealed the roles of SKAP2 and cortactin in the oocyte maturation process. To investigate the distribution, activity, and functional significance of SKAP2 during murine oocyte asymmetric division, ovarian tissues were infected with lentiviruses expressing SKAP

Allergic manifestations of actinopathies: A review.

Actinopathies are conditions that impact the maturation and formation of actin filaments. When actinopathies affect immune cells, immune dysregulation leads to atopic disease, autoimmunity, autoinflammation, and increased infection. Many actinopathies, including Wiskott-Aldrich syndrome and Dedicator of cytokinesis 8 (DOCK8) deficiency, have prominent atopic manifestations. With emerging research and new clinical descriptions of patients diagnosed with actinopathies, the impact of actinopathies on allergic pathways has expanded. Furthermore, effective treatments related to atopy have been iden

WAVE complex forms linear arrays at negative membrane curvature to instruct lamellipodia formation.

Different actin nucleation-promoting factors (NPFs) orchestrate different patterns of cell protrusions, likely reflecting their distinct patterns of self-organization. Here, we leveraged in vivo biochemical approaches to investigate how the WAVE complex instructs the formation of sheet-like lamellipodia. We show that the WAVE complex is a core constituent of a linear multilayered protein array at the plasma membrane, expected for an NPF that builds sheet-like actin-based protrusions. Negative membrane curvature is both necessary and sufficient for WAVE complex linear membrane association in th
