INTEGRIN FUNCTION IN LEUKOCYTE-MEDIATED INFLAMMATION-ACTINOPATHIES IN IMMUNE DISEASES.

Integrins play a critical role in leukocyte recruitment and activation within inflamed tissues. These heterodimeric cell-surface receptors recognize ligands on vascular endothelium or extracellular matrix to initiate intracellular signals leading to leukocyte adhesion, migration, and activation. The best-described role for integrins is in the leukocyte adhesion cascade, which is the process by which leukocytes exit the blood vasculature and enter the tissues in response to infection or injury. During the adhesion cascade, integrin signaling is required for changes in leukocyte cytoskeletal str

Agent-based modelling of the early stages of actin polymerisation required to drive endocytosis in Saccharomyces cerevisiae.

Endocytosis is critical. Its complexity means that many aspects remain poorly understood. We have developed an agent-based model covering key components of actin filament generation in endocytosis in Saccharomyces cerevisiae. The model incorporates realistic values for rates, affinities, concentrations, and mobilities, and reproduces essential features of endocytosis, from the arrival of WASp/Las17 and its inhibitor Sla1 at the membrane up to the burst of actin polymerisation. The model yields relative rates and affinities for interactions that cannot be measured experimentally, and places lim

The Role of the Wiskott-Aldrich Syndrome Protein Family in Cancer Development, Invasion, and Metastasis.

Emerging evidence indicates that cancer metastasis is governed by distinct genetic mechanisms that operate independently of those regulating carcinogenesis and cancer proliferation. The Wiskott-Aldrich syndrome protein (WASP) family members significantly contribute to the regulation of actin cytoskeleton (AC) dynamics and cancer cell invasion and metastasis. Recent research has demonstrated that WASP and its homolog, the WASP family verprolin-homologous protein (WAVE), function as a master regulator and a key scaffolding protein, bringing together the various components of metastatic signaling

Development of an All-Hydrocarbon Stapled Peptide Targeting BRK1 in Triple-Negative Breast Cancer.

Wiskott-Aldrich syndrome protein family (WASF) members are key regulators of actin cytoskeleton dynamics at the leading edge of the cell membrane. WASF3 has been demonstrated to directly promote cancer invasion and metastasis in triple-negative breast cancer. WASF3 is incorporated into a heteropentameric protein complex with BRK1, CYFIP1/2, NCKAP1/1L, and ABI1/2/3 termed the WASF Regulatory Complex (WRC) that links upstream signaling pathways to Arp2/3-mediated actin nucleation. Disruption of the complex inhibits actin remodeling and presents a novel approach to targeting cancer invasion and m

CD9 downregulation activates EGFR/ERK/WAVE2 pathway to remodel F-actin and promote proliferation and migration of cholesteatoma epithelial cells.

Although overproliferation and high migration of keratinocytes are the key pathological features of middle ear cholesteatoma (MEC), the underlying mechanism remains unclear. We found low CD9 and high ADAM17 expression in MEC specimens, alone with their colocalization, and elevated EGF levels. To investigate the role of CD9 in MEC, we created stable CD9-knockdown keratinocyte cell lines. Results showed enhanced proliferation and migration in CD9-knockdown cells, which were inhibited upon restoring CD9. Exogenous EGF reversed the inhibition. Increased ADAM17 sheddase activity in CD9-knockdown ce

Comorbidities in Mild WAS/XLT Require Lifelong Follow-Up and Consideration of Definitive Treatment.

Loss of function mutations in the gene encoding WASP (Wiskott-Aldrich syndrome protein) result in Wiskott-Aldrich syndrome (WAS) and X-linked thrombocytopenia-XLT (WAS/XLT). The clinical severity of the disease can be assessed using the WAS clinical severity score. Typically, patients with a score of 3 or less at 2 years of age are considered to have mild WAS/XLT disease. However, patients with a low score in the first 2 years of life can still experience life-threatening complications, and there are no agreed guidelines for the management of these patients with mild WAS/XLT. We analyzed data

Wiskott-Aldrich syndrome complicated with IgG4-related Sclerosing disease: A case report and literature review.

Wiskott-Aldrich Syndrome (WAS) is an X-linked immunodeficiency characterized by eczema, microthrombocytopenia, and recurrent infections. Immunoglobulin G4-related disease (IgG4-RD) is a fibroinflammatory disorder involving various organs. We present a 34-year-old male with WAS who developed cervical lymphadenopathy and parotid gland swelling. Initial biopsies were inconclusive and imaging suggested pleomorphic adenoma. Given the persistent cervical lymphadenopathy and the underlying immunodeficiency, lymphoma was also considered in the differential diagnosis. However, histopathological examina

Is it time for the A/I (allergist/immunologist) to embrace AI (artificial intelligence) in diagnosis and treatment of the inborn errors of immunity?

Background: In 1970, W.B. Schwartz predicted that computers would revolutionize medicine by enhancing the physician's intellect, a vision that has largely materialized in the past 5 decades. Recent advancements in artificial intelligence (AI), especially in health care, have transformed AI from a conceptual tool into a fundamental part of clinical practice. AI has been successfully applied in diagnostic imaging, health system management, and patient care workflows. Within immunology, AI's potential for diagnosing and managing complex conditions such as inborn errors of immunity (IEI) is increa

Enhancing lentiviral production for WAS gene therapy: a comparative analysis of stable producer cell lines evaluating flatware system and adherent bioreactors in perfusion mode.

Ex-vivo gene therapies require scalable, high-quality lentivirus (LV) with excellent transduction efficiency. Achieving this involves a synergistic approach combining efficient vector design and LV process optimization. In our study, we evaluated transfection reagents for generating stable producer cell lines from two Tet-off regulated adherent stable LV packaging PCLs, GPRG and GPRTG, to produce lentivirus (LV) to treat Wiskott Aldrich Syndrome (WAS). Stable producer cell lines expressing the WAS transgene or GFP transgene were generated from GPRG and GPRTG PCLs. The GPRTG producer cell line

Immunogenetic investigation of WAS patients revealing impaired IL-6/STAT3 signaling in T cells.

Wiskott-Aldrich syndrome (WAS) is an inborn error of immunity caused by loss-of-function mutations in the WAS gene, which encodes WASp, a key regulator of cytoskeletal remodeling. In addition to microthrombocytopenia, affected individuals often present with recurrent infections, eczema, eosinophilia and elevated IgE levels, suggesting a potential pathophysiological overlap with STAT3 hyper-IgE syndrome (HIES). Given these shared features, we investigated the immunogenetic characteristics of three WAS patients and explored the IL-6/STAT3 pathway as a potential underlying mechanism. Flow cytomet

Intrinsically disordered enteropathogenic E. coli EspF exploits motif mimicry in high-affinity binding to neural Wiskott-Aldrich syndrome protein and sorting nexin 9.

EspF is an enteropathogenic Escherichia coli (EPEC) effector protein that interferes with intestinal epithelial cell signaling by binding to the Src homology 3 (SH3) domain of sorting nexin 9 (SNX9) and the GTPase-binding domain (GBD) of neural Wiskott-Aldrich syndrome protein (N-WASP) with its C-terminal proline-rich repeats. To understand the molecular basis of these interactions, we characterized the structure, dynamics, and binding thermodynamics of EspF and its target protein domain complexes. We also elaborated on our previous study on EspFU, a homologous effector in enterohemorrhagic E.

Endophilin-lamellipodin-VASP, key components in fast endophilin-mediated endocytosis, control actin polymerization within liquid-like condensates.

Actin polymerization is essential in several clathrin-independent endocytic pathways including fast endophilin-mediated endocytosis (FEME); however, the actin machinery involved in FEME has been elusive. Here, we show that the actin polymerase VASP colocalizes and interacts directly with the FEME priming complex. We identify endophilin as a VASP binding partner and establish novel noncanonical interactions between the SH3 domain of endophilin and the Ena/VASP homology 1 or 2 (EVH1 and EVH2) domains of VASP. We show that liquid-like condensates formed by endophilin, lamellipodin, and VASP local

Atopic dermatitis in inborn errors of immunity: at the interface of immunodeficiency and immune dysregulation.

Inborn errors of immunity (IEI) encompass a broad spectrum of immunodeficiency disorders characterized by variability in genetic background, individual immunophenotype, and clinical manifestations with organ-specific immunopathology and immune dysregulation in the form of atopy, autoimmunity, polyclonal lymphoproliferation, and malignancy. With the ever-expanding insight in the pathophysiology of IEI, atopy may be perceived as an integral part and even a hallmark of IEI diseases. This review is aimed at gathering, delineating, and summarizing the immunogenetic underpinnings of IEI diseases acc

Advancements in gene therapy for Wiskott-Aldrich syndrome: from early trials to emerging approaches.

Wiskott-Aldrich syndrome (WAS) is a rare X-linked recessive disorder characterized by microthrombocytopenia, eczema, recurrent infections, and immune dysregulation, affecting approximately 1 in 100,000 live births. While the disorder was historically fatal in early childhood, advances in hematopoietic stem cell transplantation (HSCT), gene therapy, and supportive care have improved survival, though morbidity remains high. Mutations in the WAS gene disrupt the WAS protein (WASp), which is essential for actin cytoskeleton dynamics, thereby impairing immune cell function. Over 466 mutations corre
